Tick-borne diseases constitute the predominant vector-borne health threat in North America. Recent observations have noted a significant expansion in the range of the black-legged tick (Ixodes scapularis Say, Acari: Ixodidae), alongside a rise in the incidence of diseases caused by its transmitted pathogens: Borrelia burgdorferi Johnson (Spirochaetales: Spirochaetaceae), Babesia microti Starcovici (Piroplasmida: Babesiidae), and Anaplasma phagocytophilium Zhu (Rickettsiales: Anaplasmataceae), the causative agents of Lyme disease, babesiosis, and anaplasmosis, respectively. Rodents are known to be reservoir hosts for at least 60 zoonotic diseases and are known to play an important role in their transmission and spread in different ways. We sampled different rodent communities within and around human settlements in Northern Senegal, an area subjected to major environmental transformations associated with global changes. Herein, we conducted an epidemiological study on their bacterial communities. One hundred and seventy-one (171) invasive and native rodents were captured, 50 from outdoor trapping sites and 121 rodents from indoor habitats, consisting of five speci

Intracellular microcolonies of bacteria (IMC), in some cases developing large extracellular cysts (bacterial aggregates), infecting primarily gill and digestive gland, have been historically reported in a wide diversity of economically important mollusk species worldwide, sometimes associated with severe lesions and mass mortality events. As an effort to characterize those organisms, traditionally named as Rickettsia or Chlamydia-like organisms, 1950 specimens comprising 22 mollusk species were collected over 10 countries and after histology examination, a selection of 99 samples involving 20

Efforts to monitor the expanding geographic distribution of pathogens associated with Ixodes scapularis Say (Acari: Ixodidae), and to evaluate the outcomes of environmental control interventions aiming to reduce the intensity of enzootic transmission of these pathogens, are dependent on sensitive and specific molecular assays for detection of pathogens in ticks and their rodent hosts. In this study, we modified a highly sensitive and specific multiplex PCR amplicon sequencing assay developed to identify pathogens in Ixodes ticks for use in screening blood or ear tissue from rodents (Peromyscus

Wolbachia pipientis is an obligate intracellular bacterium within the family Anaplasmataceae that infects many terrestrial arthropods and arthropod-transmitted nematodes (filariae). Several filarial species are major human pathogens, and antibiotics with activity against Wolbachia offer a promising new therapeutic approach, since the adult worms are relatively refractory to conventional anthelmintics but depend on Wolbachia for reproduction and viability. In a natural filarial parasite of cattle, Onchocerca ochengi, intermittent chemotherapy is adulticidal whereas the equivalent dose administe

The Fea's muntjac (Muntiacus feae) and the northern red muntjac (Muntiacus vaginalis) are Asian cervids maintained in captive conservation programmes in Thailand, yet information on blood-borne pathogens in these species remains limited. This study aimed to investigate Theileria spp. infection in 34 captive muntjacs from four facilities across Thailand using PCR, Sanger sequencing, phylogenetic analysis, and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based serum proteomics. All animals appeared clinically healthy, and no hemoparasites were observed on blood smears. Nevertheless,

Ticks and tick-borne pathogens (TBPs) have a significant economic impact on livestock. In Nepal, only limited information is available for the molecular identification of ticks and TBPs. This study aims to address this major challenge by providing data on the diversity of TBPs in Nepalese cattle, along with molecular sequences to facilitate accurate identification. A total of 145 cattle blood samples were collected from Pokhara (n = 70) and Madi (n = 75). The cattle were also examined for tick infestations, and the detected ticks were subjected to morphological identification, followed by mole

Tick-borne zoonotic pathogens, including Rickettsia, Anaplasma, Ehrlichia, and Babesia spp., pose increasing risks to human and animal health. This study investigated tick species composition and associated zoonotic pathogens across high-(1500-3000 m) and very high-altitude (>3000-5500 m) zones of the Qinghai-Tibetan Plateau area (QTPA). A total of 1025 questing ticks were collected from 11 counties and identified as Haemaphysalis qinghaiensis (n = 316), Dermacentor nuttalli (n = 685), D. silvarum (n = 20), and D. niveus (n = 4). Tick composition showed clear zonal variation. In the high-altit

Wild animals have been recognized as potential reservoirs of vector-borne pathogens. Proximity between these animals and urban areas increases the need to know which pathogens these are and whether they can infect domestic animals and humans. In Mangabeiras Municipal Park in Belo Horizonte, Brazil, coatis live near the urban area, which is mainly occupied by human residents and their domestic animals. Therefore, the objective of this study was to detect, through molecular and direct methods, the presence of ectoparasites and hemoparasites in coatis. A total of 216 samples were collected, of wh

Neorickettsia risticii (N. risticii) is an obligatory intracellular bacterium that causes Potomac horse fever (PHF), a disease clinically characterized by diarrhea, pyrexia, and laminitis in horses. Although sporadic reports of N. risticii infection have been linked to abortion in mares, a detailed retrospective study, including genomic analysis of the pathogen from an aborted fetus, has not been published. This study examined 546 fecal samples from clinically ill horses (January 1, 2017-December 31, 2024) and 833 colon samples from aborted equine fetuses (September 20, 2018-December 31, 2024)

BACKGROUND: This study aimed at investigating the diversity of pathogens in human-biting ixodid ticks, in relation to their seasonality and associated clinical symptoms.
METHODS: Hard ticks, collected from humans in the course of 23 years, were identified to the species level on a morphological basis. This was followed by DNA extraction and molecular analyses. The latter served to confirm tick species, and to detect important tick-borne pathogens, in particular rickettsiae, Anaplasmataceae, borreliae, and piroplasms.
RESULTS: Among 502 ticks, six species were identified, with the predominanc

BACKGROUND: Potomac horse fever (PHF) is a potentially fatal enterocolitis of horses caused by Neorickettsia risticii. The disease was originally recognised almost 40 years ago in the state of Maryland in the US. It is now known to occur in many areas of North America, as well as having been described in South America and Europe. Monocomponent PHF vaccines are available, but clinical protection with vaccination has been reported to be inconsistent.
OBJECTIVES: This study was designed to assess the immunogenicity of a commercially available Potomac Horse Fever (PHF) vaccine when administered a

Obligate intracellular bacteria have remained those for which effective vaccines are unavailable, mostly because protection does not solely rely on an antibody response. Effective antibody-based vaccines, however, have been developed against extracellular bacteria pathogens or toxins. Additionally, obligate intracellular bacteria have evolved many mechanisms to subvert the immune response, making vaccine development complex. Much of what we know about protective immunity for these pathogens has been determined using infection-resolved cases and animal models that mimic disease. These studies h

BACKGROUND: Among the Reduviidae family, triatomines are giant blood-sucking bugs. They are well known in Central and South America where they transmit Trypanosoma cruzi to mammals, including humans, through their feces. This parasitic protozoan is the causative agent of Chagas disease, a major public health issue in endemic areas. Because of the medical and economic impact of Chagas disease, the presence of other arthropod-borne pathogens in triatomines was rarely investigated.
METHODOLOGY/PRINCIPAL FINDINGS: In this study, seven triatomines species involved in the transmission of T. cruzi w

Anaplasma phagocytophilum is the etiologic agent of human granulocytic anaplasmosis (HGA), one of the major tick-borne zoonoses in the United States. The surface of A. phagocytophilum plays a crucial role in subverting the hostile host cell environment. However, except for the P44/Msp2 outer membrane protein family, the surface components of A. phagocytophilum are largely unknown. To identify the major surface proteins of A. phagocytophilum, a membrane-impermeable, cleavable biotin reagent, sulfosuccinimidyl-2-[biotinamido]ethyl-1,3-dithiopropionate (Sulfo-NHS-SS-Biotin), was used to label int
