Kyasanur Forest Disease: Advances in Experimental Animal Models, Immune Assays, and Challenges in Vaccine Development.

Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to forested regions of South India, with case fatality rates ranging from 3% to 10% and seasonal outbreaks causing recurrent illness among at-risk populations such as forest workers and villagers. The historically used formalin-inactivated KFD vaccine showed limited effectiveness in field studies, providing only about 62% protection. This moderate level of protection highlights the need for improved vaccines that offer higher efficacy and longer-lasting immunity for populations living in endemic areas. A major chall

Olfaction in the context of obesity and diabetes: Insights from animal models to humans.

The olfactory system is at the crossroad between sensory processing and metabolic sensing. In addition to being the center of detection and identification of food odors, it is a sensor for most of the hormones and nutrients responsible for feeding behavior regulation. The consequences of modifications in body homeostasis, nutrient overload and alteration of this brain network in the pathological condition of food-induced obesity and type 2 diabetes are still not elucidated. The aim of this review was first to use both humans and animal studies to report on the current knowledge of the conseque

Audiogenic epileptic DBA/2 mice strain as a model of genetic reflex seizures and SUDEP.

Epilepsy is a chronic neurological disease characterized by abnormal brain activity, which results in repeated spontaneous seizures. Sudden unexpected death in epilepsy (SUDEP) is the leading cause of seizure-related premature death, particularly in drug-resistant epilepsy patients. The etiology of SUDEP is a structural injury to the brain that is not fully understood, but it is frequently associated with poorly controlled and repeated generalized tonic-clonic seizures (GTCSs) that cause cardiorespiratory and autonomic dysfunctions, indicating the involvement of the brainstem. Both respiratory

Using One Health assessments to leverage endemic disease frameworks for emerging zoonotic disease threats in Libya.

Continued emergence, re-emergence and spread of zoonotic diseases demonstrates the imperative need for multisectoral communication and joint coordination of disease detection and response. While there are existing international frameworks underpinning One Health capacity building for pandemic prevention and response, often guidance does not account for challenges faced by countries undergoing long-term conflict and sociopolitical instability. The purpose of this research was to identify Libya's laboratory and surveillance networks and routes of inter- and multisectoral communication and coordi

A multiplex oligonucleotide ligation-PCR method for the genoserotyping of common Salmonella using a liquid bead suspension assay.

Salmonella is a major pathogen having a public health and economic impact in both humans and animals. Six serotypes of the Salmonella genus are mentioned in the Belgian and European regulation as to be rapidly excluded from the food chain (EU regulation N°2160/2003, Belgian royal decree 27/04/2017). The reference method for Salmonella serotyping, including slide-agglutination and biochemical tests, is time-consuming, expensive, not always objective, and therefore does not match the fast identification criteria required by the legislation. In this study, a molecular method, using genetic marker

Microglia-associated progression of multiple sclerosis: target identification and therapeutic engagement in human in vitro models.

Chronic progression of multiple sclerosis (MS) is likely to develop on the basis of a highly complex interaction of different mechanisms, which are probably already present at disease onset. While animal models have been instrumental in developing therapies for relapsing forms of MS, they have provided limited insight into the processes driving disease progression. To overcome these limitations, human in vitro models have emerged as powerful tools to dissect cellular mechanisms and identify novel therapeutic targets. Here, we highlight advances in modeling MS progression, using human induced p

Rationally designed chemoenzymatic synthesis of heparan sulfate oligosaccharides with neutralizable anticoagulant activity and low severe complications.

The identification of enzymes involved in biosynthesis of heparan sulfate (HS) and heparin and their successful application in chemoenzymatic synthesis have provided great impetus to rationally design well-defined oligosaccharides as ideal alternatives to animal-derived heterogeneous heparin anticoagulants clinically to treat clotting disorders. Herein, we revisited the substrate specificity of recombinant 2-O-sulfotransferases produced in different expression systems for the highly efficient chemoenzymatic synthesis of HS oligosaccharides containing the rare 2-O-sulfated GlcA (GlcA2S) residue

Revisiting the embryogenesis of lip and palate development.

Clefts of the lip and palate (CLP), the major causes of congenital facial malformation globally, result from failure of fusion of the facial processes during embryogenesis. With a prevalence of 1 in 500-2500 live births, CLP causes major morbidity throughout life as a result of problems with facial appearance, feeding, speaking, obstructive apnoea, hearing and social adjustment and requires complex, multi-disciplinary care at considerable cost to healthcare systems worldwide. Long-term outcomes for affected individuals include increased mortality compared with their unaffected siblings. The fr

Gene-environment interactions: aligning birth defects research with complex etiology.

Developmental biologists rely on genetics-based approaches to understand the origins of congenital abnormalities. Recent advancements in genomics have made it easier than ever to investigate the relationship between genes and disease. However, nonsyndromic birth defects often exhibit non-Mendelian inheritance, incomplete penetrance or variable expressivity. The discordance between genotype and phenotype indicates that extrinsic factors frequently impact the severity of genetic disorders and vice versa. Overlooking gene-environment interactions in birth defect etiology limits our ability to ide

Comprehensive mapping of neurotransmitter networks by MALDI-MS imaging.

We present a mass spectrometry imaging (MSI) approach for the comprehensive mapping of neurotransmitter networks in specific brain regions. Our fluoromethylpyridinium-based reactive matrices facilitate the covalent charge-tagging of molecules containing phenolic hydroxyl and/or primary or secondary amine groups, including dopaminergic and serotonergic neurotransmitters and their associated metabolites. These matrices improved the matrix-assisted laser desorption/ionization (MALDI)-MSI detection limit toward low-abundance neurotransmitters and facilitated the simultaneous imaging of neurotransm

Aspergillus vaccines: Hardly worth studying or worthy of hard study?

Vaccines rank among the greatest advances in the history of public health. Yet, despite the need, there are no licensed vaccines to protect humans against fungal diseases, including aspergillosis. In this focused review, some of the major scientific and logistical challenges to developing vaccines to protect at-risk individuals against aspergillosis are discussed. Approaches that have shown promise in animal models include vaccines that protect against multiple fungal genera and those that are specifically directed to Aspergillus Advances in proteomics and glycomics have facilitated identifica

Prospects for a novel vaccine against tuberculosis.

The development of a new and improved vaccine against tuberculosis has in the last 10 years been accelerated tremendously from the completed Mycobacterium tuberculosis genome and the progress in molecular biology. This has resulted in the identification of a large number of antigens with potential in tuberculosis vaccines. The next phase of this work has now started--putting the most relevant molecules back together as fusion molecules and cocktails. This requires carefully monitoring of aspects as immunodominance, recognition in different populations as well as the influence of different adju

[Genetic factors in typhoid fever].

The study of genetic factors associated to infectious diseases is a promising research field for optimizing their management and control in the future. For typhoid fever, one of our major health problems, this approach has been sporadic an fruitless for having been addressed to disease's overall susceptibility. Moreover, difficulties on recognition of clinical sub-sets have prevented definition of unambiguous clinical phenotypes appropriate for this objective. With the aim of finding the basis for phenotypes identification, or definite disease expressions, a search including foreign and nation

TAAR1 Potential for Treatment of Stress Disorders.

Substantial evidence indicates that stress serves as a major risk factor for various psychiatric disorders, including major depressive disorder (MDD), anxiety disorders, post-traumatic stress disorder (PTSD), and bipolar disorder. Although multiple therapeutic options exist for these conditions, current treatments often fall short of meeting clinical needs. Since the identification of trace amine-associated receptor 1 (TAAR1), studies across diverse animal models have revealed its involvement in modulating stress responses and its potential role in stress-related psychopathologies. CHK1 inhibition rescues abnormal glycogen buildup in a Caenorhabditis elegans model for glycogen storage disease III.

Glycogen Storage Disease Type III (GSDIII) is a rare genetic disorder caused by mutations in the AGL gene, leading to a deficiency in the glycogen debranching enzyme. This results in the accumulation of abnormal glycogen in various tissues, causing a range of symptoms, including liver enlargement and hypoglycemia. Current animal models do not fully recapitulate the severe phenotypes observed in patients, highlighting the need for improved model systems. To the best of our knowledge, this study presents the first Caenorhabditis elegans model of GSDIII, which successfully exhibits disease-releva
