Reverse genetic systems are mainly used to rescue recombinant viral strains in cell culture. These tools have also been used to generate, by inoculating infectious clones, viral strains directly in living animals. We previously developed the "Infectious Subgenomic Amplicons" (ISA) method, which enables the rescue of single-stranded positive sense RNA viruses in vitro by transfecting overlapping subgenomic DNA fragments. Here, we provide proof-of-concept for direct in vivo generation of infectious particles following the inoculation of subgenomic amplicons. First, we rescued a strain of tick-bo

The paper systematizes the data of the literature on ultrastructural changes in the central nervous system (CNS) of experimental animals infected with arboviruses of the Togaviridae family. The main site of virus reproduction in the CNS was found to be nerve cells in which the ultrastructural lesions typical for this group of viruses developed. The main features of these lesions consist in degeneration of rough membranes, ribosomes, and polysomes of the cell, hyperplasia and hypertrophy of smooth membranes, formation of various vacuoles and vesicles in the cell hyaloplasm. Mature virions accum

The first reported outbreak of St. Louis encephalitis in Canada occurred in the summer of 1975 in southern Ontario -- in the Windsor-Sarnia-Chatham area, the Niagara region and the city of Toronto. Hemmagglutination inhibition and complement fixation testing of serum samples collected during the outbreak confirmed that St. Louis encephalitis virus was the etiologic agent. Furthermore, this virus was isolated from brain tissue of a patient who died. This outbreak was probably an extension of the outbreak that occurred in the United States that summer. It was the first outbreak of arbovirus ence

In South America, the arboviruses that are most noted are those that lead to dengue, Zika, and chikungunya. Nevertheless, there are additional arboviruses that trigger outbreaks, yet do not receive adequate attention, including Oropouche virus (OROV), Mayaro virus (MAYV), Saint Louis Encephalitis virus, Madariaga virus (MADV), and Rocío virus (ROCV). A rapid review was conducted using the databases PubMed, LILACS, Scopus, and Google Scholar. This analysis focused on studies that outline the clinical signs and diagnostic methods of neglected arboviruses impacting human populations in South Amer

Multiplex immunoassays can facilitate the parallel measurement of antibody responses against multiple antigenically related pathogens, generating a wealth of high-dimensional data that depict complex antibody-antigen relationships. In this study, we developed a generalizable analytical framework to maximize inferences from multipathogen serological studies. We fit the model to measurements of immunoglobulin antibody binding against 10 arboviral pathogens from a cross-sectional study in northwest Bangladesh with 1453 participants. We used our framework to jointly infer the prevalence of each pa

Arthropod-borne viruses, referred to collectively as arboviruses, infect millions of people worldwide each year and have the potential to cause severe disease. They are predominately transmitted to humans through blood-feeding behavior of three main groups of biting arthropods: ticks, mosquitoes, and sandflies. The pathogens harbored by these blood-feeding arthropods (BFA) are transferred to animal hosts through deposition of virus-rich saliva into the skin. Sometimes these infections become systemic and can lead to neuro-invasion and life-threatening viral encephalitis. Factors intrinsic to t

The Greater Everglades Region of South Florida is one of the largest natural wetlands and the only subtropical ecosystem found in the continental United States. Mosquitoes are seasonally abundant in the Everglades where several potentially pathogenic mosquito-borne arboviruses are maintained in natural transmission cycles involving vector-competent mosquitoes and reservoir-competent vertebrate hosts. The fragile nature of this ecosystem is vulnerable to many sources of environmental change, including a wetlands restoration project, climate change, invasive species and residential development.

Coues rice rat (Oryzomys couesi), a species abundant throughout Central America, was evaluated experimentally for the ability to serve as an amplifying host for three arboviruses: Patois (Bunyaviridae, Orthobunyavirus), Nepuyo (Orthobunyavirus), and Venezuelan equine encephalitis virus subtype ID (Togaviridae, Alphavirus). These three viruses have similar ecologies and are known to co-circulate in nature. Animals from all three cohorts survived infection and developed viremia with no apparent signs of illness and long-lasting antibodies. Thus, O. couesi may play a role in the general maintenan

Mosquitoes were collected for virus isolation tests from 36 sites in Bradford, Lake, Leon, Manatee, Orange, Osceola, Pasco, Putnam, and Sarasota counties, FL, from April 6 through October 11, 1993. A total of 158,129 adult specimens were collected in 726 trap nights using CDC light traps, usually baited with dry ice. At least 35 species were represented, although 60% of the collections was made up of 3 species (Aedes infirmatus, 6.5%; Anopheles crucians, 14.4%; and Culex nigripalpus, 39.4%). Four of the 36 collecting sites were located at waste-tire sites, where 254 trap nights yielded 27,455

Sixty-two sera horse collected from two stables at Lagos, Nigeria, were tested for complement fixing antibody to 8 arbovirus antigens; Chikungunya, Igbo-Ora, Yellow fever, Wesselsbron, West Nile, Potiskum, Uganda S and Rift Valley fever. Ten per cent of the horse sera examined contained CF antibody to one or more of the test antigens and indicated considerable arbovirus activity in the two stables. Reactions with flavivirus antigens were most common and the highest antibody titres were obtained with Wesselsbron and Yellow fever viruses. Eleven per cent of the sera tested reacted with alphaviru

BACKGROUND: Brazil has witnessed the co-circulation of dengue virus (DENV), Zika virus (ZIKV), and chikungunya virus (CHIKV), with outbreaks exacerbated by environmental factors, social determinants, and poor sanitation. The recent re-emergence of Oropouche virus (OROV) has added complexity to vector control strategies, emphasizing the need for integrated approaches to curb arboviruses spread. We aimed to analyze temporal trends and spatial distributions with national scope of these emerging arboviruses.
METHODS: An ecological study using data from the Brazilian Notifiable Diseases Informatio

Arbovirus surveillance in marmosets (Callithrix spp.) that live close to humans helps identify viral circulation in the environment and contributes to public health. We investigated the exposure to arboviral infections in 47 captive and free-living Callithrix from urban and peri-urban areas in the semiarid region of northeastern Brazil (SNB) in 2018. The samples were tested for antibodies against 23 arbovirus antigens and the presence of Orthoflavivirus zikaense (ZIKV), Orthoflavivirus denguei, Alphavirus chikungunya, and Orthoflavivirus flavi using hemagglutination inhibition (HI) and RT-qPCR

Mosquito-borne diseases (MBDs) are increasingly prevalent due to the resultant impact of global change with significant health and economic impacts worldwide. Dengue virus (DENV), chikungunya virus (CHIKV), Zika virus (ZIKV), yellow fever virus (YFV), Japanese encephalitis (JEV), and West Nile virus (WNV) transmitted by Aedes and Culex species have been identified as arboviruses of public health interest. The vertical transmission (VT) refers to the process where infected mosquitoes transmit viruses to their offspring; this has been often overlooked in MBD epidemiology. We conducted a systemat

Japanese encephalitis virus (JEV) and dengue virus (DENV) are two major arboviruses known as significant public health issues worldwide. Arboviruses are a heterogeneous group of vector-borne viruses that are clinically associated with various consequences ranging from asymptomatic infections to serious forms of haemorrhagic fever marked by bleeding complications. Despite advancements in understanding the pathogenesis of arboviruses the molecular mechanisms underlying clinical outcomes remain incompletely understood. Autophagy, a cellular process crucial for maintaining homeostasis through the
