Polyethylene microplastics disrupt focal adhesion kinase (FAK) signaling and sertoli cell metabolism, compromising blood-testis barrier function and spermatogenesis.

Polyethylene microplastics (PE-MPs), used extensively in personal care applications, are non-biodegradable pollutants with demonstrated bioaccumulation potential and toxicological relevance. Recent studies have detected microplastics in human semen and testicular tissues, raising concerns about their effect on male reproductive health. This study investigates the toxicological effects of orally administered polyethylene microplastics (PE-MPs) at 20 µg/mL, 200 µg/mL, and 2000 µg/mL doses in rats for 56 days, with a focus on disruption of the blood-testis barrier (BTB) and destabilization of ect

Oral administration of antimicrobial peptide MPX can effectively regulate LPS-mediated testicular oxidative stress and blood‒testis barrier damage.

Oxidative stress and disruption of blood‒testis barrier permeability are considered key factors in the pathogenesis of testicular inflammation, degeneration, and functional impairment, which play crucial roles in male infertility. Antimicrobial peptides (AMPs) are internationally recognized as some of the most promising alternatives to antibiotics. However, the molecular mechanisms by which AMPs regulate oxidative stress and the blood‒testis barrier in the testis are still poorly understood. In this study, we orally administered 0.5 mg/kg antimicrobial peptide MPX (MPX) to mice for 20 and 40 d

A High-Fat Diet Damages the Blood-Testis Barrier Through the PCSK9-LOX1 Positive Feedback Loop in Sertoli Cells.

We primarily investigated the correlation between long-term high-fat diet and male infertility, aiming to reveal the potential mechanisms underlying PCSK9-induced spermatogenic dysfunction in this context. Clinical data revealed that infertile male patients typically exhibited hyperlipidemia and significantly elevated serum PCSK9 concentrations. To further explore this, we utilized wild-type C57BL/6 male mice and their homologous PCSK9-deficient mice, adhering to experimental control principles. After administering a high-fat diet to the experimental groups, we observed local testicular change

Study on the mechanisms of defective spermatogenesis induced by TiO2 NPs based on 3D blood-testis barrier microfluidic chip.

Titanium dioxide nanoparticles (TiO2 NPs) can reduce sperm number, but the mechanisms of defective spermatogenesis induced by TiO2 NPs have not been studied through cell-cell interactions at present. A kind of biomimetic three-dimensional blood-testis barrier microfluidic chip capable of intercellular communication was constructed with soft lithography techniques, including Sertoli cell (TM4), spermatogonia (GC-1) and vascular endothelial cell units, to study the mechanisms of TiO2 NPs-induced defective spermatogenesis. TM4 and GC-1 cells cultured in TiO2 NPs exposure and control chips were co

BDE-209 disrupted the blood-testis barrier integrity by inhibiting estrogen receptor α signaling pathway in Sprague-Dawley rats.

Deca brominated diphenyl ether (BDE-209) is a widely used flame retardant with endocrine-disrupting activity which reportedly caused sperm quality decline and damaged blood-testis barrier (BTB). However, whether BDE-209 exposure led to BTB integrity dysfunction through affecting microtubule cytoskeletal organization and junctions was not well-elucidated. This study aimed to investigate the role of estrogen receptor α (ERα) in BDE-209-mediated perturbation of BTB integrity. Male rats and primary culture Sertoli cells were co-treated with BDE-209 and propylpyrazoletriol (PPT). The data demonstra

Atorvastatin improves spermatogenesis in murine and in vitro human chronic orchitis models through restoring blood-testis barriers.

Chronic orchitis, which stems from autoimmune responses and infections, significantly impairs the testicular niche and affects male fertility. However, there is a lack of effective therapeutic drugs. In this study, we aimed to explore whether atorvastatin can be used for the treatment of chronic orchitis and the underlying mechanism based on our previous findings and detection using the database. We utilized mouse models of chronic orchitis induced by both autoimmune and infectious causes to demonstrate that atorvastatin can markedly improve spermatogenesis and fertility. Mechanism studies thr

Bisphenols as promoters of the dysregulation of cellular junction proteins of the blood-testis barrier in experimental animals: A systematic review of the literature.

Daily, people are exposed to chemicals and environmental compounds such as bisphenols (BPs). These substances are present in more than 80% of human fluids. Human exposure to BPs is associated with male reproductive health disorders. Some of the main targets of BPs are intercellular junction proteins of the blood-testis barrier (BTB) in Sertoli cells because BPs alter the expression or induce aberrant localization of these proteins. In this systematic review, we explore the effects of BP exposure on the expression of BTB junction proteins and the characteristics of in vivo studies to identify p

Spp1 Contributes to Nano-Antimony Trioxide-Induced Male Reproductive Toxicity Associated with Inflammatory Response and Blood-Testis Barrier-Related Alterations.

Nano-antimony trioxide (Nano-Sb2O3) is extensively utilized in industrial production and consumer products, leading to widespread environmental contamination and human exposure. Accumulating evidence has demonstrated that Nano-Sb2O3 induces male reproductive toxicity, yet the underlying molecular mechanisms remain poorly understood. In this study, male C57BL/6 mice were exposed to Nano-Sb2O3 (2.5, 5.0, and 7.5 mg/kg/day). Exposure to Nano-Sb2O3 induced dose-dependent reproductive toxicity, evidenced by dose-dependent reductions in sperm motility (56.70% to 36.10%) and sperm density (15.76 × 10

Deoxynivalenol Induces Blood-Testis Barrier Dysfunction through Disrupting p38 Signaling Pathway-Mediated Tight Junction Protein Expression and Distribution in Mice.

Deoxynivalenol (DON) is widely present in cereals and processed grains. It can disrupt the blood-testicular barrier (BTB), leading to sterility in males; however, the mechanism is unknown. In this study, 30 Kunming mice and TM4 cells were exposed to 0 or 4.8 mg/kg (28 d) and 0-2.4 μM (24 h) of DON, respectively. Histopathological findings showed that DON increased BTB permeability in mice, leading to tight junction (TJ) structural damage. Immunofluorescence results indicated that DON disrupted the localization of zonula occludens (ZO)-1. The results of protein and mRNA expression showed that t

rpS6 regulates blood-testis barrier dynamics through Arp3-mediated actin microfilament organization in rat sertoli cells. An in vitro study.

In the seminiferous epithelium of rat testes, preleptotene spermatocytes residing in the basal compartment are transported across the blood-testis barrier (BTB) to enter the adluminal compartment at stage VIII of the epithelial cycle. This process involves redistribution of tight junction (TJ) proteins via reorganization of actin cytoskeleton in Sertoli cells that serves as attachment site for adhesion protein complexes. Ribosomal protein S6 (rpS6), a downstream molecule of mTORC1 (mammalian target of rapamycin complex 1), participates in this process via a yet-to-be defined mechanism. Here, w

Perfluorooctanoic acid disrupts the blood-testis barrier and activates the TNFα/p38 MAPK signaling pathway in vivo and in vitro.

Perfluorooctanoic acid (PFOA) is correlated with male reproductive dysfunction in animals and humans, but the underlying mechanisms for this remain unknown. To explore the potential reproductive toxicity of PFOA, we studied blood-testis barrier (BTB) damage using in vivo and in vitro models. Male mice were gavage-administered PFOA (0-20 mg/kg/d) for 28 consecutive days, and breeding capacity and permeability of the Sertoli cell-based BTB were estimated. Primary Sertoli cells (SCs) were exposed to PFOA (0-500 μM) for 48 h, and transepithelial electrical resistance (TER) was assessed. Furthermor

Mitochondrial ROS-associated integrated stress response is involved in arsenic-induced blood-testis barrier disruption and protective effect of melatonin.

Arsenic (As) is an environmental metalloid. Previous studies have demonstrated that As exposure resulted in decline of sperm quality. This study aimed to investigate the impact of exposure to As on blood-testis barrier (BTB) in a mouse model. Four-week-old male mice were exposed to NaAsO2 (1 or 15 mg/L) for 6 weeks. Our results found that NaAsO2 exposure disrupted the BTB and reduced sperm counts in adult mice. NaAsO2 activated the integrated stress response (ISR) and downregulated barrier junction protein in mouse testes and Sertoli cells. Ribosome profiling sequencing (Ribo-seq) and Ribosome

FTO mediates bisphenol F-induced blood-testis barrier impairment through regulating ferroptosis via YTHDF1/TfRc and YTHDF2/SLC7A11 signal axis.

Bisphenol F (BPF) has been extensively utilized in daily life, which brings new hazards to male reproductive health. However, the specific functional mechanism is still unclear. Both cell and animal models were utilized for exploring the role of RNA methylation and ferroptosis and its underlying mechanisms in male reproductive injury induced by BPF. In animal model, BPF severely destroyed the integrity of the blood-testis barrier (BTB) and induced ferroptosis. Furthermore, BPF significantly affected the barrier function of TM4 cells and promoted ferroptosis. Importantly, ChIP assays revealed t

Embryonic 6:2 FTOH exposure causes reproductive toxicity by disrupting the formation of the blood-testis barrier in offspring mice.

Previous studies have revealed nephrotoxicity, hepatotoxicity, subchronic developmental and reproductive toxicity in rats exposed to fluorotelomer alcohol (FTOH). However, the effects of embryonic 6:2 FTOH exposure on the reproductive system of offspring mice remain unclear. The purpose of this study is to explore the reproductive toxic effects of embryonic 6:2 FTOH exposure on offspring male mice and the related molecular mechanisms. Therefore, the pregnant mice were given corn oil or 6:2 FTOH by gavage from gestational days 12.5-21.5. The results demonstrated that embryonic 6:2 FTOH exposure

Interrupting the blood-testis barrier with a flutamide-loaded nanostructured lipid carrier: A novel nonsurgical contraceptive approach for male animals.

Flutamide is an antagonist of testosterone, an essential hormone in male reproduction. However, the use of flutamide as a contraceptive agent for nonsurgical castration in veterinary practice remains challenging due to its poor bioavailability. Here, the flutamide-loaded nanostructure lipid carrier (FLT-NLC) was synthesized, and its biological effects were demonstrated by an in vitro blood-testis barrier model. The flutamide was incorporated into the nanostructure lipid carrier by a homogenization method resulting in a high encapsulation efficiency (99.7 ± 0.04%). The FLT-NLC was negatively ch
