Acute Kidney Injury Induced by Bothrops Venom: Insights into the Pathogenic Mechanisms.

Acute kidney injury (AKI) following snakebite is common in developing countries and Bothrops genus is the main group of snakes in Latin America. To evaluate the pathogenic mechanisms associated with Bothrops venom nephrotoxicity, we assessed urinary and blood samples of patients after hospital admission resulting from Bothrops snakebite in a prospective cohort study in Northeast Brazil. Urinary and blood samples were evaluated during hospital stay in 63 consenting patients, divided into AKI and No-AKI groups according to the KDIGO criteria. The AKI group showed higher levels of urinary MCP-1 (

Influence of phospholipasic inhibition on neuromuscular activity of Bothrops fonsecai snake venom.

Bothrops fonsecai (B. fonsecai), a pitviper endemic to southeastern Brazil, has a venom mainly composed by snake venom phospholipases (PLA2) and metalloproteases, compounds that could interfere with neuromuscular junction in vitro. In this work, we investigated the role of PLA2 in the myotoxicity and neuromuscular blockade caused by B. fonsecai venom using different procedures frequently associated with PLA2 activity inhibition: 24 °C bath temperature, Ca2+ - Sr2+ replacement and chemical modification with p-bromophenacyl bromide (p-BPB). Mice extensor digitorum longus preparations (EDL) were

Systemic effects induced by the venom of the snake Bothrops caribbaeus in a murine model.

Snakebite envenoming by Bothrops caribbaeus, an endemic viperid from the Lesser Antillean island of Saint Lucia, is clinically characterized by local tissue damage and systemic thrombosis that can lead to cerebral, myocardial or pulmonary infarctions and venous thromboses. Systemic effects (lethality, pulmonary hemorrhage, thrombocytopenia and coagulopathy) induced by intravenous (i.v.) administration of B. caribbaeus venom were studied in mice. The role of snake venom metalloproteinases (SVMPs) in these systemic alterations was assessed by inhibition with the chelating agent calcium disodium

Humoral immune responses to venom and antivenom of patients bitten by Bothrops snakes.

Snake envenomation and its treatment cause the entry of two kind of foreign antigens into the human body: snake toxins and antivenom from animal origin. Samples of patients bitten by snakes in Uruguay were assayed to determine levels of human antibodies against venom and antivenom. The ELISA results showed that most of the patients presented an important increase of IgG and IgM antibodies against antivenom at day 15 post accident. Antibodies were reactive against both equine immunoglobulin chains by western blot assay. In the case of the response against the venom, increase in titre at day 15

Purification of a lectin with antibacterial activity from Bothrops leucurus snake venom.

A novel lectin was isolated from Bothrops leucurus snake venom using a combination of affinity and gel filtration chromatographies. The lectin (BlL) agglutinated glutaraldehyde-treated rabbit and human erythrocytes with preference for rabbit erythrocytes. Galactose, raffinose, lactose, fetal bovine serum and casein inhibited lectin-induced rabbit erythrocyte agglutination. BlL, with a molecular mass of 30 kDa and composed of two subunits of 15 kDa, showed dependence on calcium. BlL is an acidic protein with highest activity over the pH range of 4.0-7.0 and stable under heating to 70°C. Fluores

Prognostic factors for amputation in the case of envenoming by snakes of the Bothrops genus (Viperidae).

The prognostic factors for amputation following envenoming by snakes of the Bothrops genus were identified from the medical records of 3139 patients. Each of these patients had been bitten by Bothrops sp. and treated in the Hospital Vital Brazil, in São Paulo, Brazil, between 1981 and 1990. The 21 (0.67%) of the patients who had undergone amputation were compared with the 3118 who had not, with respect to the characteristics of the accident, the snake, the victim, the local and systemic manifestations of the envenoming and the treatment. There was an association between amputation and the mont

A C-type lectin from Bothrops jararacussu venom reprograms endothelial cell biology.

Snake venoms are intricate mixtures of enzymes and bioactive factors that induce a range of detrimental effects in afflicted hosts. Certain Viperids, including Bothrops jararacussu, harbor C-type lectins (CTLs) known for their modulation of a variety of host cellular responses. In this study, we isolated and purified BjcuL, a CTL from B. jararacussu venom and investigated its impact on endothelial cell behavior, contrasting it with human galectin-1 (Gal-1), a prototype member of the galectin family with shared β-galactoside-binding activity. We found that BjcuL binds to human dermal microvascu

Antibiofilm Activity of Acidic Phospholipase Isoform Isolated from Bothrops erythromelas Snake Venom.

INTRODUCTION: Bacterial resistance is a worldwide public health problem, requiring new therapeutic options. An alternative approach to this problem is the use of animal toxins isolated from snake venom, such as phospholipases A2 (PLA2), which have important antimicrobial activities. Bothropserythromelas is one of the snake species in the northeast of Brazil that attracts great medical-scientific interest. Here, we aimed to purify and characterize a PLA2 from B. erythromelas, searching for heterologous activities against bacterial biofilms. METHODS: Venom extraction and quantification were fol

Muscular regeneration after myonecrosis induced by Bothrops jararacussu snake venom from Argentina.

Venom from Bothrops snake produces severe local symptoms on the envenomed victim, such as hemorrhage, edema and myonecrosis. The latter is perhaps the most important of all, since antivenom therapy is not effective for it, even when antivenom is injected only a few minutes after the accident. In this work, mice weighing 18-20 g (n = 5) were inoculated with 70 micrograms Bothrops jararacussu venom in 0.1 ml PBS in the gastrocnemius muscle. Mice were sacrificed using ether after 1, 12 hours, 3, 5, 7 days and 2, 3, 5, 6 weeks after the injection of the venom to obtain gastrocnemius muscles. They

Muscular lesions induced by a hemorrhagic factor from Bothrops neuwiedi snake venom.

The local tissue effects of crude Bothrops neuwiedi snake venom and of its hemorrhagic factor (NHF) were studied on mouse tibialis anterior muscle in vivo. After 6 h, 8 days and 6 weeks the muscles were examined in paraffin sections stained with hematoxylin and eosin. Both NHF and crude venom produced hemorrhage and myonecrosis, later followed by muscle fiber regeneration. Intramuscular arteries also suffered necrosis. The minimal dose of NHF necessary to produce detectable hemorrhage and myonecrosis was 50 ng, while the minimal venom dose needed to produce the same effect was 20 times higher.

Combining Thiophene-Triazole Hybrids with Bothropic Antivenom to Enhance Its Inhibitory Effect Against the Coagulant Activity of Bothrops Jararaca, B. Neuwiedi, and B. Jararacussu Snake Venoms.

INTRODUCTION: Snakebite envenomation causes approximately 5 million incidents, 130,000 deaths, and 400,000 amputations annually worldwide. Thus, the objective of this work was to assess the ability of 16 thiophene-triazole hybrid compounds (6a-6h and 7a-7h) to inhibit the coagulant activity of Bothrops jararaca, B. neuwiedi, and B. jararacussu venoms in combination with commercial antibothropic antivenom. METHOD: In the experimental prevention protocol, human plasma or commercial fibrinogen was incubated for 60 seconds at 37°C with the study compounds, with or without antivenom, followed by t

Therapeutic potential of Bothrops leucurus phospholipase A2 in preventing dengue virus infection.

BlD-PLA2, a phospholipase A₂ isolated from Bothrops leucurus venom, exhibited notable antiviral activity against dengue virus (DENV) in vitro. Treatment significantly reduced viral RNA levels, particularly when administered during the infection period, suggesting interference with early stages of viral entry. Immunofluorescence and ELISA assays confirmed its interaction with host cell components. Importantly, reseeding assays demonstrated that residual viral RNA detected after treatment was not associated with infectious particles. These findings indicate that BlD-PLA2 effectively disrupts DEN

Bioactive peptides (cryptides) obtained by Bothrops jararaca serine peptidases action on myoglobin.

Serine peptidases and metallopeptidases are the primary toxins found in Bothrops snakes venoms, which act on proteins in the tissues of victims or prey, and release of peptides formed through proteolytic activity. Various studies have indicated that these peptides, released by the proteolytic activity of heterologous enzymes, generate molecules with unidentified functions, referred to as cryptids. To address this, we purified serine peptidases from Bothrops jararaca venom using molecular exclusion chromatography and then incubated them with the endogenous substrate myoglobin. As a control, we

Bothrops venom-induced hemostasis disorders in the rat: Between Scylla and Charybdis.

Hemostasis impairment represents the most threatening consequence of Viperidae envenoming, notably with Bothrops genus. In the French departments of America, B. atrox envenomation in French Guiana may lead to bleeding while B. lanceolatus envenomation in Martinique to thrombosis. Bleeding related to B. atrox envenomation is attributed to vascular damage mediated by venom metalloproteinases and blood uncoagulable state resulting from thrombocytopenia and consumptive coagulopathy. Thrombosis related to B. lanceolatus envenomation are poorly understood. We aimed to compare the effects of B. atrox

Modulation of Adhesion Molecules Expression by Different Metalloproteases Isolated from Bothrops Snakes.

Snake venom metalloproteinases (SVMP) are involved in local inflammatory reactions observed after snakebites. Based on domain composition, they are classified as PI (pro-domain + proteolytic domain), PII (PI + disintegrin-like domains), or PIII (PII + cysteine-rich domains). Here, we studied the role of different SVMPs domains in inducing the expression of adhesion molecules at the microcirculation of the cremaster muscle of mice. We used Jararhagin (Jar)-a PIII SVMP with intense hemorrhagic activity, and Jar-C-a Jar devoid of the catalytic domain, with no hemorrhagic activity, both isolated f
