A cell-cell communication signal from Enterobacter cloacae interfering with the signaling systems and virulence in Shigella sonnei.

UNLABELLED: Quorum sensing (QS) is widely utilized by both bacteria and fungi to mediate cell-cell communication. Previous studies have demonstrated that the indole derivative indole-3-ethanol (also known as tryptophol) controls morphogenesis as a QS molecule in fungi. However, whether this QS signal is involved in the modulation of biological functions in bacteria remains unknown. Here, we report that indole-3-ethanol controls the biological functions and pathogenicity of Enterobacter cloacae subsp. cloacae ATCC 13047. The biosynthesis of indole-3-ethanol is performed by YjgB (ECL_RS22935), a

FlyPhoneDB2: A computational framework for analyzing cell-cell communication in Drosophila scRNA-seq data integrating AlphaFold-multimer predictions.

Cell-cell communication (CCC) plays a critical role in the physiological regulation of organisms and has been implicated in numerous diseases. Previously, we introduced FlyPhoneDB, a tool designed to explore CCC in Drosophila single-cell RNA-sequencing datasets. The core algorithm of FlyPhoneDB infers tissue-specific signaling events between cell types by calculating cell-cell interaction scores based on curated ligand-receptor (L-R) expression across major signaling pathways. However, the utility of FlyPhoneDB was limited by the relatively small number of available L-R pairs. Here, we present

Extracellular vesicles contribute to early cyst development in autosomal dominant polycystic kidney disease by cell-to-cell communication.

Autosomal dominant polycystic kidney disease (ADPKD) is characterized by the formation of fluid-filled cysts within the kidney due to mutations in PKD1 or PKD2. Although the disease remains incompletely understood, one of the factors associated with ADPKD progression is the release of nucleotides (including ATP), which can initiate autocrine or paracrine purinergic signaling by binding to their receptors. Recently, we and others have shown that increased extracellular vesicle (EVs) release from PKD1 knockout cells can stimulate cyst growth through effects on recipient cells. Given that EVs are

The Regulation of Flavivirus Infection by Hijacking Exosome-Mediated Cell-Cell Communication: New Insights on Virus-Host Interactions.

The arthropod-borne flaviviruses are important human pathogens, and a deeper understanding of the virus-host cell interaction is required to identify cellular targets that can be used as therapeutic candidates. It is well reported that the flaviviruses hijack several cellular functions, such as exosome-mediated cell communication during infection, which is modulated by the delivery of the exosomal cargo of pro- or antiviral molecules to the receiving host cells. Therefore, to study the role of exosomes during flavivirus infections is essential, not only to understand its relevance in virus-hos

Imatinib sensitizes T-cell lymphocytes from chronic myeloid leukemia patients to FasL-induced cell death: a brief communication.

There is now substantial evidence that imatinib may affect immune responses, especially those mediated by T lymphocytes. Fas (CD95/Apo-1), a cell death receptor, is a key regulator of the immune system. We have explored the consequences of treatment on the Fas system in chronic myeloid leukemia patients treated with imatinib. In comparison with healthy controls, we found not only a mild blood lymphopenia but also impairment of phytohemagglutinin activation in CD4Fas and CD8Fas lymphocytes of imatinib-treated patients. Moreover, these lymphocyte populations were more sensitive to FasL-induced c

[The Role of Connexins and Pannexins in the Cell Communications of Bone Cells].

Connexins and Pannexins play important roles in osteocytes and osteoblasts differentiation, intracellular signal transduction, maintenance of bone balance, and bone regeneration. This article reviews the progress and limitations of Connexins-mediated gap junctions and Pannexins mediated hemichannel in bone. Current research has shown that these molecules, in the form of gap junctions or separate hemichannels, deliver external stimuli to the skeletal system. However, little is known about the role of other cell types in bone development and homeostasis, such as pre-osteoblasts and bone marrow m

Single-cell transcriptomics reveals cellular heterogeneity and cell-cell communication networks in free gossypol-induced enteritis in juvenile largemouth bass (Micropterus salmoides).

This study utilized single-cell transcriptomics to reveal the cellular heterogeneity and cell-cell communication networks in free gossypol-induced enteritis in juvenile largemouth bass (Micropterus salmoides). Four isonitrogenous and isolipidic diets were formulated: a fishmeal-based control (FM) and three diets supplemented with 150, 300, or 600 mg/kg gossypol acetate (FG150, FG300, or FG600). Healthy fish (14.80 ± 0.10 g) were randomly assigned to four groups (four replicates of 25 fish each) and fed for eight weeks. Results showed that increasing dietary FG progressively impaired growth per

Caregiver communication and psychosocial outcomes among adolescents and young adults with sickle cell disease.

OBJECTIVE: The transition from pediatric to adult medical care is associated with increased symptoms of anxiety, depression, and experiences with disease stigma for adolescents and young adults (AYA) with sickle cell disease (SCD). Community-informed conceptual frameworks suggest that caregiver communication could play an important role in enhancing AYA well-being during this critical period; yet this association has yet to be empirically investigated in this population. This study examined the influence of caregiver-AYA communication on psychosocial outcomes among AYA with SCD. METHODS: AYA

Luteolin Disrupts Keratinocyte-Dendritic Cell Communication in Psoriasis by Targeting Rh Family C Glycoprotein.

Psoriasis, a chronic inflammatory skin disease, arises from a dysregulated interaction between keratinocytes (KCs) and dendritic cells (DCs). We previously identified Rh family C glycoprotein (RHCG) as a key mediator of KC inflammation and DC activation. Here, we demonstrate that luteolin, a bioactive compound derived from the traditional Chinese formula cooling blood and detoxicating formula (CBDF), directly binds to RHCG, as confirmed by multiple computational methods and in vitro experiments. In vitro, luteolin suppressed RHCG expression in KCs, reducing CXCL14 secretion and subsequent DC a

Monocyte-plasma cell communication characterizes treatment-associated peripheral immune remodeling in Guillain-Barré syndrome.

BACKGROUND: Guillain -Barré syndrome (GBS) is an acute immune-mediated polyradiculoneuropathy involving multifaceted pathogenic mechanisms. Although humoral immunity is central to GBS pathogenesis, longitudinal changes in peripheral immune circuits across symptomatic disease and clinical recovery remain incompletely understood at single-cell resolution. METHODS: We performed longitudinal single-cell RNA sequencing of peripheral blood mononuclear cells collected during the symptomatic GBS and clinical recovery phases from a patient with GBS. Changes in immune cell composition, transcriptional

Antioxidant sericin averts the disruption of oocyte-follicular cell communication triggered by oxidative stress.

Antioxidants are free radical scavengers that increase oocyte quality and improve female fertility by suppressing oxidative stress. However, the related mechanisms remain unclear. The present study was designed to examine whether a reduction of oxidative stress from using the antioxidant sericin led to expanded cumulus cell (CC)-oocyte communication and oocyte developmental acquisition in a bovine model. We found that cumulus-oocyte complexes (COCs) matured in the presence of sericin showed a significantly increased oocyte meiotic maturation rate (P < 0.01) and accelerated subsequent blastocys

Lidocaine Ameliorates Psoriasis by Obstructing Pathogenic CGRP Signaling‒Mediated Sensory Neuron‒Dendritic Cell Communication.

Psoriasis is a chronic immune-mediated skin disorder with the nervous system contributing to its pathology. The neurogenic mediators of psoriasis are elusive, and whether the intervention of the cutaneous nervous system can treat psoriasis remains to be determined. In this study, we conducted a pilot study using an epidural injection of lidocaine to treat patients with psoriasis. Lidocaine treatment markedly reduced patients' clinical scores and improved an imiquimod-induced rat model of psoriasis as competent as systemic delivery of a TNF-α antibody. Imiquimod application elicited aberrant cu

Association between hospital admissions and healthcare provider communication for individuals with sickle cell disease.

Objective: To test the hypothesis that caregivers' or adult participants' low ratings of provider communication are associated with more hospital admissions among adults and children with sickle cell disease (SCD), respectively. Secondarily, we determined whether there was an association between the caregivers' or participants' health literacy and rating of providers' communication. Methods: Primary data were collected from participants through surveys between 2014 and 2016, across six sickle cell centers throughout the U.S. In this cross-sectional cohort study, 211 adults with SCD and 331 car

Tackling sickle cell crisis in Nigeria: the need for newer therapeutic solutions in sickle cell crisis management - short communication.

Sickle cell disease (SCD) is a group of inherited haematological disorders, which affects the shape of the oxygen-carrying haemoglobin component of erythrocytes, giving it an abnormal sickle-shaped appearance. This disease is one of the most common haematological disorders in Nigeria and is generally characterized by anaemia, painful crises, and multi-organ dysfunction. Recurrent episodes of painful crises account for most of the morbidities and mortalities observed in SCD, particularly sickle cell anaemia. This has been a critical concern in the field of haematology and molecular genetics as

Anesthetics and Cell-Cell Communication: Potential Ca2+-Calmodulin Role in Gap Junction Channel Gating by Heptanol, Halothane and Isoflurane.

Cell-cell communication via gap junction channels is known to be inhibited by the anesthetics heptanol, halothane and isoflurane; however, despite numerous studies, the mechanism of gap junction channel gating by anesthetics is still poorly understood. In the early nineties, we reported that gating by anesthetics is strongly potentiated by caffeine and theophylline and inhibited by 4-Aminopyridine. Neither Ca2+ channel blockers nor 3-isobutyl-1-methylxanthine (IBMX), forskolin, CPT-cAMP, 8Br-cGMP, adenosine, phorbol ester or H7 had significant effects on gating by anesthetics. In our publicati
