OBJECTIVE: Three-dimensional (3D) culture systems offer a more physiologically relevant environment than conventional two-dimensional (2D) cultures, particularly for studying tumor biology. Here, we systematically compared two widely used 3D platforms-poly(2-hydroxyethyl methacrylate) (Poly-HEMA, PH)-coating and ultra-low attachment (ULA) plates-to evaluate their impact on pancreatic cancer (PCa) cell behavior. We assessed spheroid morphology, chemotherapeutic response, invasion potential, and adhesion molecule expression in two PCa cell lines (PANC-1 and SU.86.86).
RESULTS: Spheroid morpholo

Biological research groups may face a high barrier to entry when constructing custom 3D cell culture devices to investigate multi-tissue interactions in vitro. Standard fabrication methods such as lithography, etching, or molding are expensive and require specialized equipment and expertise. To address this, we developed an accessible approach for producing polyethylene glycol (PEG)-based cell culture devices using stereolithography 3D printing with a polydimethylsiloxane intermediate mold. Both the intermediate molding steps and the sterilized final device show low cytotoxicity, and the final

Angiogenesis is a multistep biological process involving endothelial cell activation, migration, elongation, lumen formation, branching, and anastomosis. Conventional two-dimensional culture systems are useful for the preliminary evaluation of drugs, genes, or culture conditions, but they do not adequately recapitulate the spatial organization of endothelial morphogenesis within a three-dimensional extracellular matrix. The present protocol describes a microcarrier bead-based three-dimensional angiogenesis assay using human umbilical vein endothelial cells (HUVECs). The HUVECs are first seeded

Although radiotherapy (RT) is one of the main approaches used in cancer treatment, differences in clinical response among patients have not been eliminated, underscoring the need for improved methods to analyze radiobiological responses. Elucidation of variable responses requires a reliable preclinical model, among which three-dimensional (3D) cell aggregates have emerged as a new gold standard due to their physiological relevance and close resemblance to tumor microenvironments. Non-destructive assessment of the effect of RT or other treatments typically involves brightfield imaging to quanti

The epidermis, the outermost layer of the skin, retains moisture and functions as a physical barrier against the external environment. Epidermal cells are continuously replaced by turnover, and thus to understand in detail the dynamic cellular events in the epidermis, techniques to observe live tissues in 3D are required. Here, we established a live 3D imaging technique for epidermis models. We first obtained immortalized human epidermal cell lines which have a normal differentiation capacity and fluorescence-labelled cytoplasm or nuclei. The reconstituted 3D epidermis was prepared with these

Tissue-on-chip systems represent promising platforms for monitoring and controlling tissue functions in vitro for various purposes in biomedical research. The two-dimensional (2D) layouts of these constructs constrain the types of interactions that can be studied and limit their relevance to three-dimensional (3D) tissues. The development of 3D electronic scaffolds and microphysiological devices with geometries and functions tailored to realistic 3D tissues has the potential to create important possibilities in advanced sensing and control. This study presents classes of compliant 3D framework

This study analyses the performance of polylactic acid (PLA)-collagen microcarriers engineered under laboratory-scale conditions with scalable potential for three-dimensional (3D) cell culture applications. The microcarriers were evaluated for their ability to support cell adhesion, viability, proliferation and conglomerate formation using Caco-2 cells. Results show that PLA-collagen microcarriers significantly improve cell density, with notable proliferation by day 7. Scanning Electron Microscopy (SEM) revealed a rough, porous surface topology that fosters cellular aggregation and conglomerat

Adrenocortical carcinoma (ACC) is a rare malignancy of the adrenal cortex that is associated with a poor prognosis. Developing effective treatment options for ACC is challenging owing to the current lack of representative preclinical models. This study addressed this limitation by developing and characterizing 3-dimensional (3D) cell cultures incorporating the ACC cell lines, MUC-1, HAC15, and H295R in a type I collagen matrix. ACC tissue samples were analyzed by immunohistochemistry to determine the presence of type I collagen in the tumor microenvironment. Cell viability and proliferation we

In vitro tumor models might be useful to study tumor growth, invasion and therapy resistance, but also extracellular matrix (ECM) remodeling. This protocol will provide information for the generation of a 3D tumor cell cultures for high content analysis and describes a novel method to monitor the ECM morphology and remodeling as well as therapy responses using live 3D tumor cell spheroids. Using 3D spheroids of thyroid carcinoma (TC) cells as a model, our method involves the treatment of TC cells with inhibitory agents followed by subsequent analysis of ECM components to assess the influence o

Three-dimensional (3D) models to study human disease mechanisms have demonstrated that the third dimension is an essential component for neuronal maturation and function. However, 3D neuronal cell culture is challenging due to their ultra-soft nature and specific extracellular matrix (ECM) organization. This study presents a microfiber reinforcement approach, combining primary mouse spinal cord neurons (SCNs) in a purpose-adaptable hyaluronic-acid-based matrix with melt electrowritten (MEW) frames to study disease mechanisms. The importance of laminins (LNs) is evaluated, which are vital for n

BACKGROUND INFORMATION: Extracellular matrix (ECM)-derived hydrogels are frequently used in three-dimensional (3D) cell culture and organoid formation in several tissues. However, in the 3D cultivation of testicular cells, the hyaluronic acid (HA) hydrogel has not received as much attention. This study examined the effects of three distinct composites, including HA-alginate (HA-Alg), HA-alginate-collagen (HA-Alg-Col), and HA-alginate-decellularized ECM (HA-Alg-dECM), on mouse testicular cell culture and in vitro spermatogenesis.
METHODS: For the creation of composites, the concentration of bi

Neuroendocrine tumors (NETs) of the pancreas are rare neoplasms that present complex challenges to diagnosis and treatment due to their indolent course. The incidence of pancreatic neuroendocrine tumors has increased significantly over the past two decades. A limited number of pancreatic neuroendocrine cell lines are currently available for the research. Here, we present 3D-iNET ORION, a novel 3-dimensional (spheroid) cell line, isolated from human pancreatic neuroendocrine tumor liver metastasis. Three-dimensionally grown (3D) cancer cell lines have gained interest over the past years as 3D c

Barrier organ models need a scaffold structure to create a two compartment culture. Technical filter membranes used most often as scaffolds may impact cell behaviour and present a barrier themselves, ultimately limiting transferability of test results. In this work we present an alternative for technical filter membrane systems: a 3D bioprinted biological membrane in 24 well format. The biological membrane, based on extracellular matrix (ECM), is highly permeable and presents a natural 3D environment for cell culture. Inspired by the human placenta we established a coculture of a trophoblast-d

Intestinal organoids (IOs) are versatile, physiologically relevant models of the human gut. In cystic fibrosis research, IOs help evaluate CFTR correctors, potentiators, and translational readthrough compounds. Here, we present a protocol for generating induced pluripotent stem cell (iPSC)-derived IOs with high phenotypic similarity to primary material. We describe steps for establishing iPSC culture, generating intestinal progenitors via directed differentiation, and maturing these intestinal progenitors using three-dimensional culture. We then detail procedures for establishing intestinal or

This study focuses on advancing the understanding of breast cancer through 3D in vitro models, which provide biomimetic environments superior to many 2D cultures and animal models. Ex vivo analyses show that malignant breast tissues exhibit increased stiffness with higher tumour grade. Tumour stiffening is associated with altered cell phenotype, promoting progression, invasion, and metastasis. This research aims to design 3D models that mimic the evolving tumour microenvironment to study how matrix stiffness affects breast cancer cell behaviour. Using gelatin-methacryloyl (GelMA) hydrogels, we

Three-dimensional (3D) cell cultures offer superior potential in replicating native tissue microenvironments by better supporting cell-cell and cell-extracellular matrix (ECM) interactions that are critical for guiding cellular behavior and functionality in engineered tissues. Among 3D approaches, scaffold-free techniques have gained attention for their ability to produce high-cellular density, and well-organized tissue-like constructs. In particular, cell sheets are uniquely suited for regenerative applications due to their contiguous architecture, large-area coverage, and integration potenti

Head and neck cancer (HNC) is the sixth most prevalent malignancy worldwide and includes a variety of upper gastrointestinal abnormalities. HNC includes oral, throat, voice box, nasal cavity, paranasal sinuses, and salivary gland cancers. Squamous cells in the mouth, nose, and throat cause HNC. Drugs, alcohol, poor diets, smoking, and genetics all contribute to this condition. Cancer research has focused on three-dimensional (3D) models in HNC biology in recent decades. An adequate microenvironmental system and cancer cell culture are the initial steps to understanding cancer cells' complicate

Cancer cell line spheroids provide a three-dimensional (3D) culture platform that enables the evaluation of cancer cell aggregation, tumor initiation, cell proliferation, and overall tumor growth. Compared with conventional two-dimensional monolayer cultures, spheroid models more faithfully recapitulate the architectural, physiological, and microenvironmental features of solid tumors, making them particularly well suited for studying tumor biology and microbe-host interactions. Previous work has leveraged this 3D system to investigate the role of the oral oncobacterium Fusobacterium nucleatum

The extensive reliance on animal models in biomedical research motivates the development of advanced in vitro systems that recapitulate physiological complexity while minimizing animal use. Large cellular spheroids can mimic native tissue architecture; however, scalable fabrication of spheroids exceeding millimeter dimensions remains challenging. Here, we introduce a rigid porous capsule (RPC) in millimeter scale, a mechanically robust yet highly permeable platform that imposes external geometric confinement to enable cell-cell aggregation and three-dimensional proliferation. The RPC is fabric
