Dual action of cholecystokinin-octapeptide on the guinea pig antrum.

We examined the mechanism of action of cholecystokinin-octapeptide (CCK-OP) on longitudinal strips of the lesser and greater curvatures of the guinea pig antrum. In the strips of the lesser curvature, CCK-OP produced a concentration-dependent tonic contraction, which was significantly reduced by atropine, but not by tetrodotoxin. In contrast, CCK-OP caused a relaxation of the preparation of the greater curvature in a concentration-dependent manner. The relaxation induced by the peptide was enhanced by atropine, whereas it was blocked by tetrodotoxin. Propranolol, phentolamine, desensitization

Effects of cholecystokinin-octapeptide on gastric motility of anesthetized dogs.

The present experiments examined the local effects of cholecystokinin-octapeptide (CCK-8) and related peptides on gastric motility of anesthetized dogs. Peptides were injected through the gastroepiploic artery at doses of 1.0, 2.5, 5.0, 10.0, and 20.0 ng/ml. CCK-8 and its analogues (Glt-CCK-8, pGlu-CCK-8, and Suc1-MePhe8-CCK-7) increased gastric smooth muscle contraction in a dose-dependent manner. ED50 of CCK-8 was 2.97 +/- 0.63 ng/ml. Administration of atropine (100-200 micrograms/kg) inhibited the effects of both CCK-8 and pentagastrin; however, hexamethonium (5 mg/kg) failed to block the c

The satiety effect of cholecystokinin. Recent progress and current problems.

Since the discovery of the satiety effect of CCK in 1973, progress has been made and problems have been encountered. The progress has included the accumulation of strong, indirect evidence that exogenous CCK acts in the abdomen to activate vagal afferent fibers and that exogenous CCK may be useful in the treatment of bulimia and obesity in humans. The most pressing problem is the current lack of evidence for the hypothesis that the satiety effect of exogenous CCK reveals a physiological function of endogenous CCK released by food entering the small intestine during a meal. Since clarification

Effect of alcohol on the release of cholecystokinin and pancreatic enzyme secretion.

Ethanol is often implicated in the pathogenesis of acute pancreatitis, but the pathophysiologic processes of alcohol-induced acute pancreatitis remains poorly understood. We found that ingestion of alcohol by healthy volunteers did not stimulate release of cholecystokinin, which is the chief hormonal stimulant of pancreatic enzyme secretion, nor did it significantly alter fasting levels of pancreatic polypeptide, a hormonal inhibitor of pancreatic enzyme secretion. In conscious dogs prepared with chronic pancreatic fistulas, direct intraduodenal instillation of ethanol significantly reduced pa

Cholecystokinin metabolism in normal man and patients with duodenal ulcer.

Using two cholecystokinin (CCK) radioimmunoassays CCK has been shown to be heterogeneous (CCK-33 and CCK-8) after both exogenous infusion and endogenous release. CCK-39 (CCK variant) was not separated after endogenous CCK stimulation but was identified after the exogenous infusion of 16% pure CCK. The disappearance half-time of 16% pure CCK has been shown to be 2.4 min and one major organ of catabolism identified--the kidney. Degradation of CCK by whole blood occurred after standing for 2 h, but if separation was carried out immediately serum could be left for 6 h before any further degradatio

Eating, drinking and temperature responses to intracerebroventricular cholecystokinin in the chick.

The central effect of cholecystokinin-octapeptide (CCK), SQ 19,844 or sincalide, on the intake of food and water and on colonic temperature (Tc) was investigated using the broiler cockerel. Four-week old chicks were maintained in a thermoneutral environment of 23-24 degrees C. After food was removed for a 24 hr interval, CCK was infused in a volume of 10.0 microliters into the lateral cerebral ventricle (ICV) in doses ranging from 10-150 ng. Although lower doses of CCK had no effect on food intake, 100 or 150 ng of CCK significantly reduced consumption of food in a dose-dependent manner; water

Influence of repeated administration of cholecystokinin and secretin on the pancreas of the rat.

Repeated injections of cholecystokinin (CCK) during a period of up to 3 weeks significantly increased the weight of the pancreas in rats. This was associated with an increase in the amount of protein per unit weight of DNA, suggesting hypertrophy of the acinar cells, and with increase in the total amount of pancreatic DNA, indicating additional hyperplasia of the gland. CCK administration also increased the pancreatic content of amylase and trypsin, but the content of lipase remained unchanged. The rate of secretion of the two enzymes increased in the CCK-treated rats, although it appeared tha

Beta cell-derived cholecystokinin drives obesity-associated pancreatic adenocarcinoma development.

Pancreatic endocrine-exocrine crosstalk plays a key role in normal physiology and disease and can be altered by host metabolic states, such as obesity. Classically, endocrine islet beta (β) cell secretion of insulin is thought to promote the development of obesity-associated pancreatic adenocarcinoma (PDAC), an exocrine cell-derived tumor. Here, we show that β cell expression of the peptide hormone cholecystokinin (CCK) is necessary and sufficient for obesity-associated PDAC progression in mice and that CCK expression - rather than insulin - correlates strongly with enhanced tumorigenesis. Sin

Cholecystokinin responsiveness varies across the population dependent on metabolic phenotype.

Background: Cholecystokinin (CCK) is an important satiety factor, acting at type 1 receptors (CCK1Rs) on vagal afferent neurons; however, CCK agonists have failed clinical trials for obesity. We postulated that CCK1R function might be defective in such patients due to abnormal membrane composition, such as that observed in cholesterol gallstone disease. Objective: Due to the challenges in directly studying CCK1Rs relevant to appetite control, our goal was to develop and apply a method to determine the impact of a patient's own cellular environment on CCK stimulus-activity coupling and to determ

Feeding-suppressive mechanism of sulfated cholecystokinin (26-33) in chicks.

The anorexigenic effect of cholecystokinin (CCK) is well documented in mammals, but documentation in neonatal chicks is limited. Thus, the present study investigated the mechanism underlying the anorexigenic effect of CCK in neonatal chicks. Intraperitoneal (IP) injection of sulfated CCK(26-33) (CCK8S) significantly decreased food intake in chicks at 60 and 300 nmol/kg. Non-sulfated CCK(26-33) (CCK8) also significantly decreased food intake, but its anorexigenic effect was observed only at the highest dose (300 nmol/kg) and short-lived. However, CCK(30-33) (CCK4) had no effect on food intake.

Hindbrain leptin receptor stimulation enhances the anorexic response to cholecystokinin.

Leptin is thought to reduce food intake, in part, by increasing sensitivity to satiation signals, including CCK. Leptin action in both forebrain and hindbrain reduces food intake, and forebrain leptin action augments both the anorexic and neuronal activation responses to CCK. Here, we asked whether leptin signaling in hindbrain also enhances these responses to CCK. We found that food intake was strongly inhibited at 30 min after a combination of 4th-intracerebroventricular (4th-icv) leptin injection and intraperitoneal CCK administration, whereas neither hormone affected intake during this per

Cholecystokinin-2 receptor mediated gene expression in neuronal PC12 cells.

Cholecystokinin (CCK) is abundantly expressed in the CNS, in which it regulates feeding behavior and long-term memory. Moreover, CCK has been implicated in mental disorders, such as anxiety and schizophrenia. Despite its manifest physiological and pathophysiological role, the molecular targets of neuronal CCK are incompletely understood. To identify genes regulated by neuronal CCK, we generated neuronal PC12 cells stably expressing the CCK-2 receptor (CCK-2R) and treated the cells with sulphated CCK-8 for 2-16 h, before the global expression profile was examined. The changes in gene expression

Cholecystokinin and panic disorder: past and future clinical research strategies.

The involvement of cholecystokinin (CCK) in human anxiety is well documented. Exogenous administration of CCK-2 receptor agonists, such as cholecystokinin-tetrapeptide and pentagastrin, provoke panic attacks in man. Patients with panic disorder (PD) are hypersensitive to CCK-2 receptor stimulation compared to healthy volunteers and patients with other anxiety disorders, and they differ from healthy subjects in CCK metabolism and genetic characteristics of the CCK-2 receptor system. This article reviews the corpus of work supporting the role of CCK in anxiety and suggests three research approac

Morphine augmentation increases gallbladder visualization in patients pretreated with cholecystokinin.

UNLABELLED: The purpose of this study was to determine if a combination of cholecystokinin (CCK) pretreatment followed by morphine augmentation improved the detection of cystic duct patency compared with CCK pretreatment only. METHODS: One hundred fifty-five patients with suspected acute cholecystitis had scintigraphy performed with 185-481 MBq (5-13 mCi) 99mTc-mebrofenin adjusted to the patients' total bilirubin levels. All patients were pretreated with 0.02 microgram/kg sincalide injected intravenously over 3-5 min. Sequential imaging was performed until gallbladder activity was identified

Changes in cholecystokinin mRNA expression in methamphetamine-induced behavioral sensitization.

Neuropeptide cholecystokinin (CCK) mRNA was measured in the rat brain subchronically treated with methamphetamine (MAP). Male Wistar rats were injected daily with MAP (3 or 6 mg/kg, i.p., once a day) or saline for 14 days. Progressive reinforcement was observed in MAP-induced stereotyped behaviors. After 7 or 14 days of discontinuation, the rats were decapitated and the brains were prepared for Northern blot analysis using 32P-labeled cDNA probes. Northern blot analysis revealed that the levels of CCK mRNA in the frontal cortex and the hippocampus of 3 or 6 mg/kg MAP-treated rats were signific

Intracerebroventricular administration of cholecystokinin reduces stereotypy in dopamine-supersensitive rats.

Cholecystokinin (CCK-8) coexists with dopamine in some neurons and modulates dopaminergic neurotransmission. In the present study we determined the effect of CCK-8 on stereotyped behavior in supersensitive dopaminergic system. Adult male Wistar rats, weighing 200-250 g, were used. Dopaminergic supersensitivity was induced by long-term haloperidol (HAL) treatment (30 days: 1.0 mg/kg twice a day). Seventy-two hours after HAL withdrawal animals received CCK-8 (14.5 nmol/5 microliters) or saline intracerebroventricularly (icv) before being tested for apomorphine (APO, 0.6 mg/kg, sc)-induced stereo

[Cholecystokinin, neurotensin and corticotropin-releasing factor, three important anorexic peptides].

This paper updates the informations on the three most important anorexigenic peptides: cholecystokinin, neurotensin and corticotropin-releasing factor. Their peripheral and/or central effects on food and water intakes as well as on dietary preferences are detailed. Their mechanisms of action and regulation are examined. This includes the interactions with classical neurotransmitters (norepinephrine, dopamine, etc...) as well as the description of the brain nuclei and neuronal networks involved. Finally, their variations in disturbed feeding behavior (hyperphagia, anorexia) in man or in animal

Delayed biliary-to-bowel transit in cholescintigraphy after cholecystokinin treatment.

The authors assessed the influence of cholecystokinin (CCK), administered before cholescintigraphy, on the biliary-to-bowel transit time (BBTT) of technetium-99m disofenin. Fourteen healthy volunteers underwent two separate cholescintigraphic studies with and without CCK treatment. BBTT was less than 1 hour in all 14 studies of subjects not treated with CCK. In 14 subjects treated with CCK, there was no tracer activity in the bowel up to 2 hours in seven (50%) (P = .006). Eighty-three cholescintigrams obtained in patients with suspected acute cholecystitis were also retrospectively analyzed. I
