Opportunity and means: horizontal gene transfer from the human host to a bacterial pathogen.

The acquisition and incorporation of genetic material between nonmating species, or horizontal gene transfer (HGT), has been frequently described for phylogenetically related organisms, but far less evidence exists for HGT between highly divergent organisms. Here we report the identification and characterization of a horizontally transferred fragment of the human long interspersed nuclear element L1 to the genome of the strictly human pathogen Neisseria gonorrhoeae. A 685-bp sequence exhibiting 98 to 100% identity to copies of the human L1 element was identified adjacent to the irg4 gene in so

Use of a bacterial antimicrobial resistance gene microarray for the identification of resistant Staphylococcus aureus.

As diagnostic and surveillance activities are vital to determine measures needed to control antimicrobial resistance (AMR), new and rapid laboratory methods are necessary to facilitate this important effort. DNA microarray technology allows the detection of a large number of genes in a single reaction. This technology is simple, specific and high-throughput. We have developed a bacterial antimicrobial resistance gene DNA microarray that will allow rapid antimicrobial resistance gene screening for all Gram-positive and Gram-negative bacteria. A prototype microarray was designed using a 70-mer b

Association of chronic non-bacterial osteomyelitis with Crohn's disease but not with CARD15 gene variants.

Chronic non-bacterial osteomyelitis (CNO) is an inflammatory, non-infectious disorder of the skeletal system with unknown etiology. Besides bone-inflammation, patients may present with inflammatory involvement of other tissues. Chronic recurrent multifocal osteomyelitis (CRMO) is the most severe form of CNO. We describe the occurrence of Crohn's disease (CD) in four patients, previously diagnosed with CRMO. Mutations in CARD15, encoding the NOD2 protein, have recently been found in patients with CD. Based on the occurence of CNO and CD in these four and several reported patients, we hypothesiz

Infective Arthritis: Bacterial 23S rRNA Gene Sequencing as a Supplementary Diagnostic Method.

Consecutively collected synovial fluids were examined for presence of bacterial DNA (a 700-bp fragment of the bacterial 23S rRNA gene) followed by DNA sequencing of amplicons, and by conventional bacteriological methods. One or more microorganisms were identified in 22 of the 227 synovial fluids (9,7%) originating from 17 patients. Sixteen of the patients had clinical signs of arthritis. For 11 patients molecular and conventional bacterial examinations were in agreement. Staphylococcus aureus, Streptococcus dysgalactiae subspecies equisimilis and Streptococcus pneumoniae, were detected in syno

Gene exchange of thyA for interleukin-10 secures live GMO bacterial therapeutics.

Extract: The exponential outburst of knowledge in molecular immunology has provided us with an in depth insight into the biological activity of cytokines. These are small, freely diffusible proteins that, together with numerous growth factors and chemokines, act as messengers by which cells of the immune system communicate with each other and with most other tissues in the body. As such, these molecules are able to regulate many aspects of the immune response in which numerous cells and tissues may be involved at any one time. Most often, cytokines are active in extremely low concentrations. I

Characterisation of a gene cluster involved in bacterial degradation of the cyanobacterial toxin microcystin LR.

A novel pathway for degradation of the cyanobacterial heptapeptide hepatotoxin microcystin LR was identified in a newly isolated Sphingomonas sp. (Bourne et al. 1996 Appl. Environ. Microbiol. 62: 4086-4094). We now report the cloning and molecular characterisation of four genes from this Sphingomonas sp. that exist on a 5.8-kb genomic fragment and encode the three hydrolytic enzymes involved in this pathway together with a putative oligopeptide transporter. The heterologously expressed degradation pathway proteins are enzymatically active. Microcystinase (MlrA), the first enzyme in the degrada

Identification and characterization of a Salmonella typhimurium oxygen-regulated gene required for bacterial internalization.

Growth of Salmonella typhimurium in a low-oxygen environment induces the ability of these bacteria to enter mammalian cells. We have carried out a search for invasion genes that are expressed under low-oxygen conditions by using Tn5lacZY transcriptional fusions. Several noninvasive oxygen-regulated lacZY insertion strains have been identified. The invasion defect in one of these noninvasive S. typhimurium strains, BJ66, has been complemented by introduction of a cosmid (pBDJ125) from an S. typhimurium SL1344 gene bank. A 1.9-kb EcoRV DNA fragment subcloned from this cosmid, containing a single

Metagenomic insights into the ecological risks of multiple heavy metals on soil bacterial communities and resistance-related genes.

The simultaneous contamination of soils by various heavy metals (HMs) significantly disrupts microbial ecosystems, leading to notable shifts in bacterial community structures and the prevalence of resistance-related genes, including metal resistance genes (MRGs) and antibiotic resistance genes (ARGs). Conventional indices, such as the Nemerow Integrated Pollution Index (NIPI), primarily assess contamination from a chemical standpoint but often overlook the complex ecological responses elicited by multiple HMs. In this study, we used metagenomic analyses, network assessments, and neutral commun

Environmental antibiotic stress and high-risk resistance genes in bacterial communities of the Gomti and Ganga Rivers, India.

BACKGROUND: The river ecosystems provide habitats and source of water for a number of species including humans. The uncontrolled accumulation of pollutants in the aquatic environment enhances the development of antibiotic-resistant bacteria and genes. METHODS: Water samples were collected seasonally from different sites of Gomti and Ganga River. Bacteria were isolated by plating on nutrient agar supplemented with individual antibiotics (100 µg/ml) to select the resistant strains. These isolates were subsequently tested for cross-resistance to other antibiotics using the disc diffusion method.

Metagenomics-based antibiotic resistance genes diversity and prevalence risk revealed by pathogenic bacterial host in Taihu Lake, China.

In recent decades, antibiotics and antibiotic resistance genes (ARGs) have been regarded as emerging pollutants. In Taihu Lake, as a typical representative of freshwater system in China, the ARGs occurrence and abundance was of great importance for ecological risk control and public health protection. In this research, high-throughput sequencing and metagenomics technique were used to investigate the seasonal ARGs profile in overlying water and sediment at typical area of Taihu Lake. Besides, taxonomy pattern of ARGs host bacteria and potential pathogens were identified. The results showed tha

Safety of composts consisting of hydrothermally treated penicillin fermentation residue: Degradation products, antibiotic resistance genes and bacterial diversity.

Combining hydrothermal treatment and composting is an effective method to dispose of penicillin fermentation residue (PFR), but the safety and related mechanism are still unclear. In this study, penicillin solution was hydrothermally treated to decipher its degradation mechanism, and then hydrothermally treated PFR (HT-PFR) was mixed with bulking agents at ratios of 2:0 (CK), 2:1.5 (T1), and 2:5 (T2) to determine the absolute abundance of antibiotic resistance genes (ARGs) and the succession of bacterial community. Results showed that penicillin was degraded to several new compounds without th

A new targeted capture method using bacterial artificial chromosome (BAC) libraries as baits for sequencing relatively large genes.

To analyze a specific genome region using next-generation sequencing technologies, the enrichment of DNA libraries with targeted capture methods has been standardized. For enrichment of mitochondrial genome, a previous study developed an original targeted capture method that use baits constructed from long-range polymerase chain reaction (PCR) amplicons, common laboratory reagents, and equipment. In this study, a new targeted capture method is presented, that of bacterial artificial chromosome (BAC) double capture (BDC), modifying the previous method, but using BAC libraries as baits for seque

Two HMGB1 genes from grass carp Ctenopharyngodon idella mediate immune responses to viral/bacterial PAMPs and GCRV challenge.

High mobility group box 1 (HMGB1) is a nuclear weapon in the immune arsenal and a master regulator of innate immunity, at the crossroads between innate and adaptive immunity. To clarify the immune characterizations of HMGB1 in fishes, two co-orthologs of HMGB1 (CiHMGB1a and CiHMGB1b) were identified in grass carp Ctenopharyngodon idella by local EST database searching and RACE techniques. mRNA expressions of the two HMGB1 genes are widespread in fifteen tissues investigated. The transcripts of CiHMGB1a and CiHMGB1b were significantly up-regulated and reached peak at 24h post GCRV challenge in

Molecular characterization of peach PR genes and their induction kinetics in response to bacterial infection and signaling molecules.

'Venture' and 'BabyGold 5' are two peach cultivars with a demonstrated resistance and susceptibility, respectively, to bacterial spot disease caused by Xanthomonas campestris pv. pruni (Xcp). To explore the differences between these cultivars at the molecular level, two PR1 (Pp-PR1a, Pp-PR1b) and three PR5 (Pp-TLP1, Pp-TLP2 and Pp-TLP3) genes were isolated from peach (Prunus persica L.) and investigated by in silico and in situ approaches. The analysis of gene expression by qRT-PCR indicated that all PR genes, except Pp-PR1a, were induced to a significantly higher degree in the resistant culti

Reorganisation of the caecal extracellular matrix upon Salmonella infection--relation between bacterial invasiveness and expression of virulence genes.

Interactions of Salmonella (S.) outer membrane structures with extracellular matrix (ECM) of host tissues seem to be crucial for bacterial adhesion and invasion. To evaluate the relationship between the ECM and bacterial invasiveness, the reorganisation of fibronectin, tenascin-C and laminin after Salmonella exposure in vivo, the Salmonella adhesiveness to ECM proteins in vitro and the virulence gene expression upon co-cultivation of salmonellae and ECM proteins were elucidated for two Salmonella strains with different capabilities to enter the intestinal mucosa. Comparison of bacterial diversity along the human intestinal tract by direct cloning and sequencing of 16S rRNA genes.

Bacterial diversity of the mucosal biopsies from human jejunum, distal ileum, ascending colon and rectum were compared by analysis of PCR-amplified 16S rDNA clone libraries. A total of 347 clones from the mucosal biopsies were partially sequenced and assigned to six phylogenetic phyla of the domain Bacteria: Firmicutes, Bacteroidetes, Proteobacteria, Fusobacteria, Verrucomicrobia, and Actinobacteria. The jejunum sample had least microbial diversity compared to the other samples and a trend towards highest diversity in ascending colon was observed. The clone libraries of distal ileum, ascending

Differential expression of genes encoding calmodulin-binding proteins in response to bacterial pathogens and inducers of defense responses.

Calmodulin (CaM) plays an important role in sensing and transducing changes in cellular Ca2+ concentration in response to several biotic and abiotic stresses. Although CaM is implicated in plant-pathogen interactions, its molecular targets and their role in defense signaling pathway(s) are poorly understood. To elucidate the signaling pathways that link CaM to defense responses, we screened a cDNA library constructed from bean leaves undergoing a hypersensitive response (HR) with radiolabeled CaM isoforms. A total of 26 putative CBPs were identified. Sequencing of the cDNAs revealed that they

Single gene target bacterial identification. groEL gene sequencing for discriminating clinical isolates of Burkholderia pseudomallei and Burkholderia thailandensis.

Proper identification of Burkholderia pseudomallei and Burkholderia thailandensis is crucial in guiding clinical management of patients with suspected melioidosis, as more than 99% of cases of melioidosis are caused by B. pseudomallei, whereas B. thailandensis is only responsible for causing less than 1% of the cases. However, the difference between the 16S ribosomal RNA gene sequences of B. pseudomallei and that of B. thailandensis is only 1%, and is therefore not discriminative enough for distinguishing the 2 species confidently. In this study, we amplified and sequenced the groEL genes of 7

Global Analysis of the Co-Occurrence of Antimicrobial and Metal Resistance Genes in Bacterial Genomes From Dairy-Isolated Pathogens.

Metal contamination in dairy production can drive antimicrobial resistance co-selection independently of antibiotic use, yet the genomic architecture linking metal resistance and antimicrobial resistance across dairy-isolated pathogens remains largely uncharacterized globally. We analysed 3303 genomes from dairy environments across 52 countries, creating the first genome-resolved map of co-occurring resistance determinants in this critical agricultural interface. Metal/stress resistance genes were present at levels comparable to AMR genes (28.85% of isolates showed metal dominance), with arsen
