Histone deacetylase: a potential therapeutic target for fibrotic disorders.

Histone deacetylases (HDACs) are enzymes that balance the acetylation activities of histone acetyltransferases on chromatin remodeling and play essential roles in regulating gene transcription. In the past several years, the role of HDACs in cancer initiation and progression, as well as the therapeutic effects of HDAC inhibitors in various types of cancer, has been well studied. Recent studies indicated that HDAC activity is also associated with the development and progression of some chronic diseases characterized by fibrosis, including chronic kidney disease, cardiac hypertrophy, and idiopat

Local acetyl-CoA metabolism and acetate recycling as epigenetic regulators of chromatin dynamics.

Histone acetylation depends on acetyl-CoA, a central metabolic intermediate linking nutrient availability to chromatin regulation. Beyond bulk acetyl-CoA abundance, accumulating evidence indicates that chromatin-proximal acetyl-CoA production and acetate recycling provide spatial control over histone acetylation. In budding yeast, acetyl-CoA synthetase Acs2 supports nucleocytosolic acetyl-CoA production and associates with chromatin during transcriptional transitions, enabling local acetyl-CoA generation for histone acetyltransferases. Its mammalian ortholog, ACSS2, can accumulate in the nucle

Epigenomic Evaluation of Cholangiocyte Transforming Growth Factor-β Signaling Identifies a Selective Role for Histone 3 Lysine 9 Acetylation in Biliary Fibrosis.

BACKGROUND & AIMS: Transforming growth factor β (TGFβ) upregulates cholangiocyte-derived signals that activate myofibroblasts and promote fibrosis. Using epigenomic and transcriptomic approaches, we sought to distinguish the epigenetic activation mechanisms downstream of TGFβ that mediate transcription of fibrogenic signals. METHODS: Chromatin immunoprecipitation (ChIP)-seq and RNA-seq were performed to assess histone modifications and transcriptional changes following TGFβ stimulation. Histone modifications and acetyltransferase occupancy were confirmed using ChIP assays. Assay for Transposa

Acetylation of H2AX on lysine 36 plays a key role in the DNA double-strand break repair pathway.

Phosphorylation of H2AX functions to recruit DNA repair complexes to sites of DNA damage. Here, we report that H2AX is constitutively acetylated on lysine 36 (H2AXK36Ac) by the CBP/p300 acetyltransferases. H2AXK36Ac is required for cells to survive exposure to ionizing radiation; however, H2AXK36Ac levels are not increased by DNA damage. Further, acetylation of H2AX did not affect phosphorylation of H2AX or the formation of DNA damage foci. Finally, cells with a double mutation in both the H2AX acetylation and phosphorylation sites were more radiosensitive than cells containing individual muta

Role of Nitric Oxide, Ethylene, Hydrogen Sulfide and Their Interplay With Histone Acetylation in Plants.

Histone acetylation is an essential epigenetic modification that regulates chromatin structure and gene expression by opposing the actions of histone acetyltransferases (HATs) and deacetylases (HDACs). Gas signaling molecules are endogenous gaseous compounds that can freely diffuse across biological membranes. They mediate interplant communications and plant signaling transductions, with representative members including nitric oxide (NO), ethylene (Eth), hydrogen sulfide (H2S), and methane (CH4). Multiple studies have revealed that gas signaling molecules and histone acetylation can interact t

Influence of Histone Acetylation on Chromatin 3D Organization in Drosophila S2 Cells.

Alterations in histone acetylation are frequently detected in various cancers and neurological diseases. Histone acetylation is known to disrupt both intra- and inter-nucleosomal interactions, but its influence on higher-order chromatin organization has been studied to a lesser extent. In this study, we changed global levels of histone acetylation in Drosophila S2 cells by treatment with trichostatin A (TSA; an inhibitor of histone deacetylases) or with curcumin (an inhibitor of histone acetyltransferases and histone deacetylases). We also performed a knockdown of the main histone deacetylase

cAMP responsive element modulator α promotes effector T cells in systemic autoimmune diseases.

T lymphocytes play a crucial role in adaptive immunity. Dysregulation of T cell-derived inflammatory cytokine expression and loss of self-tolerance promote inflammation and tissue damage in several autoimmune/inflammatory diseases, including systemic lupus erythematosus (SLE) and psoriasis. The transcription factor cAMP responsive element modulator α (CREMα) plays a key role in the regulation of T cell homeostasis. Increased expression of CREMα is a hallmark of the T cell-mediated inflammatory diseases SLE and psoriasis. Notably, CREMα regulates the expression of effector molecules through tra

SIRT1 retention in elongating spermatids interferes with histone displacement by counteracting MOF-dependent H4K16 acetylation.

INTRODUCTION: The histone H4 hyperacetylation (i.e., acetylation of H4 at lysines -K5, -K8, -K12, and -K16, here reported as H4tetraAc) occurs in elongating spermatids (eSPTs) during spermiogenesis. Although it is critically involved in histone displacement, the mechanistic involvement of histone -acetyltransferases (HATs) and -deacetylases (HDACs) in the pathway underlying H4 hyperacetylation is poorly defined. Here, we investigate the involvement of SIRT1 deacetylase, and its functional interaction with the histone acetyltransferase MOF, in regulating H4 hyperacetylation underlying histone-t

Switching off the yeast-to-hyphae transition in Yarrowia lipolytica through histone deacetylase inhibitors.

Fungi can develop a variety of morphotypes to survive, colonize, adapt and prevail under different environmental conditions. In general, two morphological shapes encompass the others: yeast (unicellular) and hyphae (multicellular). Under specific conditions, some fungi can adopt these two cellular morphologies, and for this reason, they are called dimorphic. Histone acetylation and deacetylation are well-known important mechanisms of chromatin remodelling that control cell differentiation processes as dimorphism. The reactions involved are catalysed by histone acetyltransferases (HATs) and his

The transcriptional corepressor, PELP1, recruits HDAC2 and masks histones using two separate domains.

PELP1 (proline-, glutamic acid-, and leucine-rich protein 1) has been recognized as a coactivator of estrogen receptor (ER)-recruiting p300/CREB-binding protein histone acetyltransferase to the target chromosome. The present study shows that PELP1 does indeed coactivate ER-mediated transcription but also serves as a corepressor of other nuclear hormone receptors (NR)- and non-NR sequence-specific transcription factors tested, including GR, Nur77, AP1, NF-kappaB, and TCF/SRF. PELP1 expression also retarded the proliferation of mouse fibroblast cell lines in which there was no detectable ER. Thi

The ATM-related cofactor Tra1 is a component of the purified SAGA complex.

The SAGA histone acetyltransferase/transcriptional adaptor complex is composed of multiple transcriptional regulators including Ada, Spt, and TAFII proteins. Here we identify an additional novel subunit of the complex, Tra1, an ATM/PI-3-kinase-related homolog of the human TRRAP cofactor, which is essential for c-Myc and E2F-mediated oncogenic transformation. Mass spectrometry, immunoblotting, and immunoprecipitation experiments confirm the stable association of this protein within SAGA. In addition, the Tra1 protein is a component of at least two other histone acetyltransferase protein complex

Dancing with KAT6A: Current advances and therapeutic potential in oncology of KAT6A inhibitors.

KAT6 A (lysine acetyltransferase 6 A) is a member of the MYST family of histone acetyltransferases (HATs) that plays a pivotal role in regulating gene expression through the acetylation of histones and non-histone proteins. Dysregulation of KAT6A has been implicated in various cancers, making it an attractive target for therapeutic intervention. Over the past, significant advancements have been made in the development of small-molecule KAT6A inhibitors. PF-07248144 has shown antitumor activity in estrogen receptor-positive (ER+) breast cancer models and is currently undergoing clinical evaluat

Structural and functional divergence of cancer hotspot mutations in CREBBP.

CREBBP is among the most frequently mutated genes in human cancer and encodes a lysine acetyltransferase (KAT) protein that acetylates histones and nonhistone proteins to drive transcriptional activation. Mutations of CREBBP most frequently result in missense changes affecting the active site within the KAT domain. However, there has not yet been comprehensive biochemical, structural, and functional studies to determine the impact of recurrent KAT-domain variants. Here we show that the most frequently mutated residue of CREBBP, arginine 1446, alters the stability of acetyl-CoA binding within t

The emerging role of histone acetylation in rheumatic diseases: unraveling mechanisms and therapeutic prospects.

Histone acetylation, a fundamental and reversible epigenetic modification, critically regulates chromatin structure and plays a pivotal role in diverse cellular processes, including gene transcription, cell cycle progression, and DNA damage repair. Rheumatic diseases [such as rheumatoid arthritis (RA), osteoarthritis (OA), and systemic lupus erythematosus (SLE)] represent a group of disorders primarily affecting joints, bones, muscles, and connective tissues, posing a significant global burden. This review systematically elucidated the aberrant regulation of histone acetylation in these rheuma

The role of histone acetylation in mediating the anticancer effects of polyphenols: Insights from genistein, resveratrol, quercetin, curcumin, and epigallocatechin-3-gallate.

Dysregulation of histone acetylation is a central epigenetic driver of oncogenesis. This review details the epigenetic modulating roles of five key dietary polyphenols-genistein (GEN), resveratrol (RES), quercetin (QUE), curcumin (CUR), and epigallocatechin-3-gallate (EGCG)-specifically focusing on their ability to modulate histone acetyltransferases (HATs) and inhibit histone deacetylases (HDACs). It elucidates the molecular mechanisms through which these compounds restore tumor suppressor gene expression, induce cell cycle arrest and apoptosis, and inhibit angiogenesis, metastasis, and drug

Hydroxamic Acid-Based Histone Deacetylase (HDAC) Inhibitors Bearing a Pyrazole Scaffold and a Cinnamoyl Linker.

Genetic abnormalities have been conventionally considered as hallmarks of cancer. However, recent studies have demonstrated that epigenetic mechanisms are also implicated in the insurgence and development of cancer. Patterns of the epigenetic component include DNA methylation and histone modifications. Acetylation of histones is controlled by histone acetyltransferases (HATs) and histone deacetylases (HDACs). Imbalance of these two enzymatic systems is known to be a key factor in tumor progression. Because HDACs have been found to function incorrectly in cancer, various HDAC inhibitors (HDACIs

Divergent SAGA complexes shape the Toxoplasma transcriptome for lytic cycle progression and host interaction.

Histone acetylation governs Toxoplasma gondii gene expression, developmental plasticity and virulence, yet the organization and deployment of its acetyltransferase machinery remain poorly understood. Here, we show that T. gondii rewired this process using a plant-like system built around the acetyltransferase TgGCN5b, which differs from the typical SAGA complex found in other eukaryotes. Interactome and structural analyses reveal a modular assembly that integrates multiple acetyltransferase (GNAT) enzymes and chromatin-reader proteins carrying PHD and PZP domains and Apetala-related transcript

Histone H3 Acetylation Is Involved in Retinoid Acid-Induced Neural Differentiation through Increasing Mitochondrial Function.

Histone acetylation and mitochondrial function contribute importantly to neural differentiation, which is critically associated with neurodevelopmental disorders such as Down Syndrome (DS). However, whether and how histone acetylation regulates mitochondrial function and further affects neural differentiation has not been well described. In this study, when treated with retinoid acid (RA), the human neuroblastoma SH-SY5Y cell line was used as a neural differentiation model. We found that the acetylation of histone H3, especially H3 lysine 14 acetylation (H3K14ac), and mitochondrial function, i
