Modulation of tramadol-induced hyperthermia by risperidone and ambient temperature in rats.

Tramadol (TRA) overdose can induce severe hyperthermia associated with monoaminergic dysregulation, particularly serotonin toxicity, and may lead to fatal outcomes. Although supportive treatments such as hydration and external cooling are commonly used, effective pharmacological interventions are yet to be established. Here, we aimed to evaluate the effects of risperidone (RIS) and ambient temperature on TRA-induced hyperthermia and changes in central monoamines in a rat model. Male Wistar rats received intraperitoneal TRA (100 mg/kg) under ambient temperatures of 18 °C, 23 °C, or 28 °C. RIS (

Performance in the heat-physiological factors of importance for hyperthermia-induced fatigue.

This article presents a historical overview and an up-to-date review of hyperthermia-induced fatigue during exercise in the heat. Exercise in the heat is associated with a thermoregulatory burden which mediates cardiovascular challenges and influence the cerebral function, increase the pulmonary ventilation, and alter muscle metabolism; which all potentially may contribute to fatigue and impair the ability to sustain power output during aerobic exercise. For maximal intensity exercise, the performance impairment is clearly influenced by cardiovascular limitations to simultaneously support ther

Paradoxical [correction of parodoxical] effect of orexin A: hypophagia induced by hyperthermia.

This experiment tested the effect of the sympathetic and thermogenic activation induced by orexin A on eating behavior. The food intake, firing rate (FR) of the sympathetic nerves to interscapular brown adipose tissue (IBAT), IBAT and abdominal temperatures (T(IBAT) and T(ab)), and heart rate (HR) were monitored in 24 h-fasting male Sprague-Dawley rats for 15 h after food presentation. Orexin A (1.5 nmol) was injected into the lateral cerebral ventricle 6 h before food presentation while FR, T(IBAT) and T(ab), and HR were also monitored. The same variables were controlled in rats receiving ore

Evidence for susceptibility to malignant hyperthermia in patients with exercise-induced rhabdomyolysis.

BACKGROUND: Malignant hyperthermia (MH), heat stroke, and exercise-induced rhabdomyolysis (ER) were suspected to be related syndromes. However, it is not known whether individuals with history of ER have an increased incidence of susceptibility to MH. To establish an association between ER and susceptibility to MH, the authors determined the MH status in patients with a history of MH-like episodes induced by physical stress. METHODS: Twelve unrelated patients with ER, 18 patients with anesthesia-induced MH, and 28 controls were investigated with the in vitro contracture test (IVCT) according

Xenon does not induce contracture in human malignant hyperthermia muscle.

Xenon has many characteristics of an ideal anaesthetic agent. It is not known whether xenon is a safe alternative to the potent inhalational anaesthetics in patients susceptible to malignant hyperthermia (MH). We investigated the effect of xenon, halothane and caffeine on muscle specimens of 31 individuals, referred to the MH Unit of the University of Ulm, and performed genetic epidemiology. Thirteen individuals were classified as MH susceptible and 18 as MH negative. Xenon 70% did not cause an increase in baseline tension of any MH-susceptible muscle specimen in contrast to halothane and caff

Hyperthermia induces apoptosis in malignant fibrous histiocytoma cells in vitro.

The effect of mild hyperthermia on a cultured rat malignant fibrous histiocytoma (MFH) cell line, MFH-2NR, was investigated. MFH cells in log-phase (growing phase) were heated at 41 degrees-44 degrees C for 1 hr. Hyperthermic treatment at 41 degrees C did not substantially affect cell proliferation and treatment at 44 degrees C caused necrosis. After hypothermic treatment at 42 degrees or 43 degrees C, proliferation of MFH cells was arrested and morphological changes characteristic of apoptosis, cell shrinkage accompanying apoptotic bodies and chromatin condensation, became apparent. Hyperther

Induced hyperthermia in sedated humans and the concept of critical thermal maximum.

The concept of critical thermal maximum (CTM) has been defined in the literature as the minimal high deep-body temperature that is lethal to an animal. In man the CTM has been estimated at 41.6--42.0 degrees C. Data are presented for sedated, unacclimatized, well-hydrated men heated 1 h at esophageal temperatures of 41.6--42.0 degrees C, without sequelae, except for modest elevation of serum enzymes in two of five patients. These data when combined with other observations in the literature suggest that CTM be redefined as the particular combination of exposure time at elevated body temperature

Hyperthermia induced by amphetamine, p-chloroamphetamine and fenfluramine in the rat-1.

Hyperthemia was elicited in rats by the subcutaneous injection of 4 mg/kg (+)-amphetamine, 4 mg/kg (+)-p-chloroamphetamine, or 8 mg/kg fenfluramine. This hyperthermia could be abolished by oral pretreatment with 300 mg/kg p-chlorophenylalanine. Cyproheptadine, at a dose of 0.05 mg/kg, completely inhibited the response induced by p-chloroamphetamine, but a dose of 0.4 mg/kg was necessary in the case of amphetamine and fenfluramine. Protection against hyperthermia by p-chloroamphetamine was also provided by chloroimipramine. Hypothalamic turnover of 5-HT was enhanced by all three amphetamines. a

Precision hyperthermia-induced HSP20 inhibits gastric cancer cell invasion, migration, and proliferation.

BACKGROUND: Many different types of human malignancies overexpress heat shock proteins (HSPs), which function as oncogenic regulators and control tumorigenesis. However, their contribution to gastric cancer (GC) remains unclear. OBJECTIVES: This study aimed to investigate the roles of HSP20 during high-temperature intraperitoneal chemotherapy for GC. METHOD: Immunohistochemistry and western blotting analysis were used to assess the levels of HSP20. Wild-type human HSP20 was sustainably overexpressed in AGS and HGC-27 cells (HSP20-overexpressing cells). The role of HSP20 in GC cells was furth

Diagnostic Challenges in Malignant Hyperthermia and Anesthesia-Induced Rhabdomyolysis: A Case Study.

BACKGROUND Malignant hyperthermia (MH) and anesthesia-induced rhabdomyolysis (AIR) are rare, yet life-threatening complications that need prompt therapeutic actions and logistic preparedness for treatment success. Both conditions are triggered by general anesthetics, particularly volatiles and depolarizing muscle relaxants. In comparison with MH, which is an inherited pharmacogenomic disease of calcium channel receptor subpopulation and arises only after trigger exposure, AIR has been described mostly in patients with muscular dystrophies. In perioperative settings, rhabdomyolysis is also obse

Inhibition of Heat Shock-Induced H3K9ac Reduction Sensitizes Cancer Cells to Hyperthermia.

Heat stress, clinically known as hyperthermia, is a promising adjunctive modality in cancer treatment. However, the efficacy of hyperthermia as a monotherapy is limited and the underlying mechanism remains poorly understood. Targeting histone modifications is an emerging strategy for cancer therapy, but little is known regarding the role of heat stress in altering these modifications. Here, we report that heat shock inhibits H3K9 acetylation (H3K9ac) via histone deacetylase 6 (HDAC6) regulation. Heat shock inhibits the interaction between HDAC6 and heat shock protein 90 (HSP90), enhances nucle

Therapeutic effects of methimazole on 3,4-methylenedioxymethamphetamine-induced hyperthermia and serotonergic neurotoxicity.

3,4-methylenedioxymethamphetamine (MDMA) is a popular recreational drug, however over 200 studies demonstrate that acute (e.g. hyperthermia, rhabdomyolysis) and chronic (e.g. neurotoxicity) toxicity effects of MDMA were observed in different animals. Methimazole (MMI), an inhibitor of thyroid hormone synthesis, was found to significantly reduce the HSP72 expression of heat stress induced in fibroblasts. Hence, we attempted to understand the effects of MMI on MDMA induced changes in vivo. Male SD rats were randomly divided into four groups as follows:(a) water-saline (b) water-MDMA (c) MMI-sali

Physiologic and Perceptual Responses to Cold-Shower Cooling After Exercise-Induced Hyperthermia.

CONTEXT: Exercise conducted in hot, humid environments increases the risk for exertional heat stroke (EHS). The current recommended treatment of EHS is cold-water immersion; however, limitations may require the use of alternative resources such as a cold shower (CS) or dousing with a hose to cool EHS patients. OBJECTIVE: To investigate the cooling effectiveness of a CS after exercise-induced hyperthermia. DESIGN: Randomized, crossover controlled study. SETTING: Environmental chamber (temperature = 33.4°C ± 2.1°C; relative humidity = 27.1% ± 1.4%). PATIENTS OR OTHER PARTICIPANTS: Seventeen

Risperidone attenuates and reverses hyperthermia induced by 3,4-methylenedioxymethamphetamine (MDMA) in rats.

3,4-Methylenedioxymethamphetamine (MDMA, "ecstasy") is a widely used recreational drug. Despite an increase in the number of fatalities related to its use, no definite therapeutic method has been established thus far. In the present study, risperidone's ability to attenuate MDMA-induced hyperthermia and its mechanism of action were investigated in rats. The pharmacological effect of MDMA was evaluated using microdialysis. In the body temperature experiment, administration of risperidone before and after MDMA administration significantly suppressed MDMA-induced hyperthermia in a dose-dependent

Molecular mechanisms of hyperthermia- and cisplatin-induced cytotoxicity in T cell leukemia.

BACKGROUND: The prognosis of early and very early relapse in acute lymphoblastic leukemia of childhood is still very poor unless a hematopoietic stem cell transplant is performed if a second remission can be achieved by induction chemotherapy. Therefore an intensification of chemotherapy is required. MATERIALS AND METHODS: In the present study the molecular mechanisms of cisplatin- and/or hyperthermia-mediated cytotoxicity in CEM cells, a human T leukemia cell line, were investigated. RESULTS: Both hyperthermia and cisplatin induced the activation of the effector caspases-3 and -6. However,

Diltiazem inhibits halothane-induced contractions in malignant hyperthermia-susceptible muscles in vitro.

The ability of diltiazem to suppress halothane and halothane-caffeine induced contractures in malignant hyperthermia (MH) susceptible pig muscle, was tested in vitro. Muscle specimens were divided into two groups and tested with a modified halothane-caffeine contracture test. One group acted as the control; the other group was pretreated with diltiazem 20 mumol litre-1. The control muscles developed contractures attributable to halothane and halothane-caffeine, whereas the diltiazem-treated specimens did not. Increases in muscle twitch tension as a result of halothane or halothane-caffeine exp

Effects of microwave-induced local hyperthermia on mammary adenocarcinoma in C3H mice.

Microwave-induced hyperthermia was focused locally upon mammary adenocarcinoma implanted in C3H mice when the tumor reached 6 mm in diameter. In 54 treated mice, all tumors diminished in size after the first exposure (43 degrees, 45 min) and disappeared completely following the second treatment. No other effects from heat application were apparent. All the mice in the microwave-treated group survived the whole period of observation (4 months) without any evidence of tumor, while 18 nontreated controls died within 4 weeks after inoculation.

Second-Near-Infrared Light-Induced SnSe Nanosheets for Augmented Photonic Tumor Hyperthermia.

Triple-negative breast cancer (TNBC) poses a serious threat to the lives and health of patients, as traditional treatments are limited by psychological burden, systemic toxicity, or drug resistance. Photonic hyperthermia triggered by the second near-infrared (NIR-II) window is a promising minimally invasive strategy. Herein, we synthesized SnSe nanosheets via a simple, scalable method for NIR-II-triggered photonic tumor hyperthermia. SnSe nanosheets exhibited irregular lamellar structures with uniform Sn/Se distribution, favorable biocompatibility, and prominent photothermal performance under
