The water extract of Aloe vera prevents fluoxetine-induced multiple-drug resistance of E. coli by inhibiting reactive oxygen species formation and membrane permeability.

BACKGROUND: The medication of synthetic chemical is one of the main treatments for depressive disorders. Different lines of evidence reveal that a long-term exposure to anti-depressants, e.g., fluoxetine, is causing multiple-drug resistance (MDR) of gut microbiomes. The MDR bacterial strains in gut pose a threat to intestinal balance and treatment of future microbial infection. Effective strategies are thus in urgent need to prevent the anti-depressant-mediated MDR of gut microbes. PURPOSE: We aimed to investigate the potential role of Aloe vera (L.) Burm. f. (aloe; Liliaceae family) to preve

Paris polyphylla saponin Ⅱ induces autophagy-associated ferroptosis by regulating the BECLIN1/SLC7A11 axis in cervical cancer.

Cervical cancer remains a lethal gynecologic malignancy requiring novel therapies. Here, we investigated the antitumor mechanism of Paris polyphylla saponin Ⅱ (PPSⅡ) in vitro and in vivo. PPSⅡ suppressed proliferation and induced cell death in SiHa, U14, and HeLa cells. The ferroptosis inhibitor ferrostatin-1 (Fer-1) and the autophagy inhibitor chloroquine (CQ) both partially reversed PPSⅡ-induced cell death, indicating involvement of both pathways. Mechanistically, SLC7A11 overexpression reversed the effects of PPSⅡ on GPX4 and ACSL4 expression, whereas BECLIN1 knockdown altered SLC7A11 expre

Comparison of anti-acetylcholinesterase activity of bulb and leaf extracts of Sternbergia candida Mathew & T. Baytop.

Studies on Alzheimer's disease have been highlighted due to increasing prevalence of this disease. Oldest hypothesis about the pathophysiology strengthens the research of cholinesterase inhibitors for treatment. Amaryllidaceae plants are well known for alkaloids showing cholinesterase inhibiting activity. Among them, Sternbergia species gained attention as a source of metabolites of these alkaloid contents. Studies have focused mainly on the bulbs of these plants. In this study the potential acetylcholinesterase (AChE) inhibitory activity of endemic Sternbergia candida Mathew & T. Baytop (Sc)

Sugar-binding proteins potently inhibit dendritic cell human immunodeficiency virus type 1 (HIV-1) infection and dendritic-cell-directed HIV-1 transfer.

Both endocytic uptake and viral fusion can lead to human immunodeficiency virus type 1 (HIV-1) transfer to CD4+ lymphocytes, either through directional regurgitation (infectious transfer in trans [I-IT]) or through de novo viral production in dendritic cells (DCs) resulting in a second-phase transfer to CD4+ lymphocytes (infectious second-phase transfer [I-SPT]). We have evaluated in immature monocyte-derived DCs both pathways of transfer with regard to their susceptibilities to being blocked by potential microbicidal compounds, including cyanovirin (CNV); the plant lectins Hippeastrum hybrid

Valorization of Paris polyphylla Byproducts: Integrated Multi-Omics and Molecular Docking Reveal the Anti-Melanogenic Mechanism of Plant-Derived Nanovesicles via the AKT/GSK3β/MITF Axis.

Valorizing agricultural byproducts into functional ingredients is highly desirable. Herein, plant-derived nanovesicles (PDNVs) from Paris polyphylla stems/leaves (SL-EXO) exhibited potent anti-melanogenic properties, whereas root-derived PDNVs were ineffective. In vitro, SL-EXO achieved 92.66% cell-free tyrosinase inhibition, while α-arbutin was 49.78%. In vivo, SL-EXO ameliorated the pigmentation-driving oxidative/senescent microenvironment in zebrafish and reduced macroscopic melanin by ~66%. Crucially, SL-EXO reversed α-MSH-induced hyperpigmentation in B16F10 cells while maintaining excelle

Effecting skin renewal: a multifaceted approach.

The skin undergoes intrinsic aging as a normal course, but exposure to ultraviolet (UV) light results in major cumulative damage that manifests as the typical aged photodamaged skin. UV irradiation produces a sequence of changes within the skin layers starting with signaling processes following DNA damage and culminating in nonabsorbed fragmentation of collagen and other proteins within the extracellular matrix. These fragments promote the synthesis of matrix metalloproteinases (MMPs) that further aggravate the damage to the ground substance and add to fragment accumulation. This study describ

Protection against experimental visceral leishmaniasis by immunostimulation with herbal drugs derived from Withania somnifera and Asparagus racemosus.

Visceral leishmaniasis (VL) is a vector-borne parasitic disease targeting tissue macrophages. It is among the most neglected infectious diseases. As available therapeutics for treatment of this disease have many side effects, there is a need for safer alternatives. One of the immunopathological consequences of active visceral leishmaniasis is suppression of protective T-helper (Th)-1 cells and induction of disease-promoting Th-2 cells, and thus the treatment of VL relies on immunomodulation. In the current study, herbal drugs derived as whole-plant extracts of Asparagus racemosus and Withania

Carbohydrate-binding agents act as potent trypanocidals that elicit modifications in VSG glycosylation and reduced virulence in Trypanosoma brucei.

The surface of Trypanosoma brucei is covered by a dense coat of glycosylphosphatidylinositol-anchored glycoproteins. The major component is the variant surface glycoprotein (VSG) which is glycosylated by both paucimannose and oligomannose N-glycans. Surface glycans are poorly accessible and killing mediated by peptide lectin-VSG complexes is hindered by active endocytosis. However, contrary to previous observations, here we show that high-affinity carbohydrate binding agents bind to surface glycoproteins and abrogate growth of T. brucei bloodstream forms. Specifically, binding of the mannose-s

[Biological activity of Ungernia victoris extract in the Escherichia coli CaCl2-transformation system in the presence of calcium channel modulators].

Furosemid and verapamil appear to have an effect on the yield of E. coli plasmid transformants according to their properties as calcium channel regulators in eukaryotes. It means that furosemid stimulates and verapamil inhibits transforming DNA penetration into competent cell. PHB/Ca2+ polyP complex does not function only as a channel for transforming DNA penetration but appears to be one of the targets for transformation blocking U. victoris extract.
