Overexpression of the Global Transcriptional Regulator LaeA Leads to Production of Cyclic Lipopeptides in Marine-Derived Aspergillus niger L14.

Overexpression of a global transcriptional regulator LaeA was performed to awaken cryptic biosynthetic gene clusters (BGCs) in a marine sponge-derived fungus, Aspergillus niger L14, to obtain secondary metabolites (SMs), and resulted in the production of five previously undiscovered SMs in the wild-type strain, including three cyclic lipopeptides aspochracin (1), JBIR-15 (2) sclerotiotide C (3), kojic acid (4) and penicillic acid (5). Bioassays revealed that compound 2 displayed promising antimicrobial effects on Candida albicans with a minimum inhibitory concentration value of 32 µg/mL and 4

Dendritic Lipopeptide Nanovaccines Orchestrate Multi-Pattern Recognition Receptors Activation and Potentiate Antitumor Immunity.

Precise control over the spatial and temporal delivery of antigens and adjuvants is essential for eliciting effective and durable immune responses. Here, we report a self-assembling nanovaccine platform based on dendritic lipopeptides (DLPs) that integrate delivery and immune-stimulation through a structurally defined design. DLPs are constructed by conjugating structurally optimized lipid tails to second-generation lysine- or arginine-rich dendritic scaffolds. Systematic engineering of the lipid domain allows for precise fine-tuning of the amphiphilic structure, yielding molecular candidates

Design of potent, proteolytically stable stapled lipopeptide analogues of BimBH3 as PTP1B inhibitors for diabetes therapy.

Protein tyrosine phosphatase 1B (PTP1B) serves as a key negative regulator of both insulin and leptin signaling pathways, and its inhibition represents a potential dual-mechanism therapeutic strategy for type 2 diabetes and obesity by simultaneously improving insulin sensitivity and leptin signaling. However, the clinical translation of peptide-based therapeutics is often hindered by poor proteolytic stability and short in vivo half-life. To address these challenges, we implemented a rational dual-modification strategy combining fatty acid conjugation and lactam stapling. Specifically, N-termi

Dendritic lipopeptide-based transdermal siRNA delivery systems for effective non-invasive therapy in psoriasis.

Psoriasis is a prevalent chronic inflammatory skin disease characterized by immune cell activation and aberrant keratinocyte proliferation. Signal transducer and activator of transcription 3 (STAT3) plays a key role in the pathology of psoriasis, positioning it as a promising target for therapeutic strategies. However, current STAT3 inhibitors often lack specificity, leading to adverse effects. Here, we present the development of dendritic lipopeptides (DLPs) designed to facilitate the transdermal delivery of small interfering RNA (siRNA) to specifically inhibit STAT3 expression in psoriatic l

Cyclic lipopeptide antibiotics bind to the N-terminal domain of the prokaryotic Hsp90 to inhibit the chaperone activity.

Chemical arrays were employed to screen ligands for HtpG, the prokaryotic homologue of Hsp (heat-shock protein) 90. We found that colistins and the closely related polymyxin B interact physically with HtpG. They bind to the N-terminal domain of HtpG specifically without affecting its ATPase activity. The interaction caused inhibition of chaperone function of HtpG that suppresses thermal aggregation of substrate proteins. Further studies were performed with one of these cyclic lipopeptide antibiotics, colistin sulfate salt. It inhibited the chaperone function of the N-terminal domain of HtpG. H

The synthetic bacterial lipopeptide Pam3CSK4 modulates respiratory syncytial virus infection independent of TLR activation.

Respiratory syncytial virus (RSV) is an important cause of acute respiratory disease in infants, immunocompromised subjects and the elderly. However, it is unclear why most primary RSV infections are associated with relatively mild symptoms, whereas some result in severe lower respiratory tract infections and bronchiolitis. Since RSV hospitalization has been associated with respiratory bacterial co-infections, we have tested if bacterial Toll-like receptor (TLR) agonists influence RSV-A2-GFP infection in human primary cells or cell lines. The synthetic bacterial lipopeptide Pam3-Cys-Ser-Lys4 (

In vitro activities of a new lipopeptide antifungal agent, FK463, against a variety of clinically important fungi.

The in vitro antifungal activity and spectrum of FK463 were compared with those of amphotericin B, fluconazole, and itraconazole by using a broth microdilution method specified by National Committee for Clinical Laboratory Standards document M27-A (National Committee for Clinical Laboratory Standards, Wayne, Pa., 1997). FK463 exhibited broad-spectrum activity against clinically important pathogens including Candida species (MIC range, <==0.0039 to 2 microg/ml) and Aspergillus species (MIC range, <==0.0039 to 0.0313 microg/ml), and its MICs for such fungi were lower than those of the other anti

Effective immunization of mice against cutaneous leishmaniasis using an intrinsically adjuvanted synthetic lipopeptide vaccine.

Two peptides representing predicted T-cell epitopes of gp63, a major surface glycoprotein of the parasite Leishmania major, were used in vaccines tested in a murine model of cutaneous leishmaniasis. Either subcutaneous or intraperitoneal immunization in saline with a peptide representing gp63 amino acids 467-482 (p467) significantly protected CBA mice against the development of severe cutaneous lesions only when the peptide was intrinsically adjuvanted by covalently adding a lauryl-cysteine moiety (LC-p467) to its amino terminus during synthesis. In marked contrast, administration of p467 alon

Canine hydrophobic surfactant polypeptide SP-C. A lipopeptide with one thioester-linked palmitoyl group.

The amino acid sequence and the posttranslational modification of the hydrophobic surfactant polypeptide SP-C from canine, rabbit and bovine lungs were established by direct sequence analysis and plasma-desorption time-of-flight mass spectrometry. The results reveal that canine SP-C has only one cysteine residue which, however, is palmitoylated, like the two Cys residues in other characterized SP-C molecules. In addition, canine SP-C is N-terminally truncated, with only 34 amino acid residues in its longest form. Thus, SP-C molecules can apparently vary to some extent in the N-terminal lipid-m

Immunopotentiating activities of a low molecular weight lipopeptide, RP 56 142--studies in infectious models.

RP 56 142, N2-[N-(N-lauroyl-L-alanyl)-gamma-D glutamyl] L,L-2,6-diaminopimelamic acid belongs to a family of immunomodulating lipopeptides. Its structure is directly derived from that of lauroyltetrapeptide RP 40 639 which is a mixture of two stereoisomers, one of which (with D,D-2,6 diaminopimelamic acid) is totally devoid of in vivo activity. RP 56 142 displayed potent protective activities against bacterial infections such as K. pneumoniae, L. monocytogenes or S. typhimurium (at doses ranging between 0.03 and 100 mg/kg s.c., i.p., i.v.). In combined treatment protocols, suboptimal doses of

Unveiling the In Vitro Antibiofilm Efficacy of Antifungal Lipopeptides Purified from Bacillus sp. against Mixed-Species Biofilms of Candida.

Polymicrobial fungal infections are often associated with significant invasive mycosis. Candida non-albicans species such as Candida glabrata and Candida tropicalis are most frequent organisms after Candida albicans especially in biofilm-associated fungal infections with significant mortality and morbidity. In the present investigation, the antibiofilm efficacy of Bacillus-derived lipopeptides AF4 and AF5 was assessed against Candida dual species mixed biofilms at different stages such as adherence, developmental, and mature phase in vitro experiments. Interestingly, in this study, C. glabrata

Antimicrobial activity of synthetic cationic peptides and lipopeptides derived from human lactoferricin against Pseudomonas aeruginosa planktonic cultures and biofilms.

BACKGROUND: Infections by Pseudomonas aeruginosa constitute a serious health threat because this pathogen -particularly when it forms biofilms - can acquire resistance to the majority of conventional antibiotics. This study evaluated the antimicrobial activity of synthetic peptides based on LF11, an 11-mer peptide derived from human lactoferricin against P. aeruginosa planktonic and biofilm-forming cells. We included in this analysis selected N-acylated derivatives of the peptides to analyze the effect of acylation in antimicrobial activity. To assess the efficacy of compounds against plankton

A comparative analysis of cytokine production and tolerance induction by bacterial lipopeptides, lipopolysaccharides and Staphyloccous aureus in human monocytes.

Monocytes (MO) and macrophages (MAC) are important producers of cytokines involved in the pathophysiology of bacterial sepsis. Most studies concentrate on the effects of bacterial lipopolysaccharides (LPS) regarding the induction of cytokine gene expression and secretion in MO/MAC. Here we report that besides LPS, the synthetic lipoprotein analogue lipopeptide N-palmitoyl-S-(2,3-bis(palmitoyl)-(2RS)-propyl)-(R)-cysteinyl-alanyl- glycine (Pam3-Cys-Ala-Gly), another component of the outer membrane of Gram-negative bacteria, as well as heat-killed Staphyloccocus aureus (S. aureus/SAC) are potent

Immunoregulatory Dendritic Lipopeptide Nanoparticles for High-Capacity Cisplatin Delivery and Pancreatic Cancer Chemo-Immunotherapy.

Lipid nanoparticles (LNPs) are promising for cisplatin delivery but are hindered by low drug loading, instability, and poor efficacy. Here, multicarboxyl dendritic lipopeptides were developed to enhance cisplatin loading via platinum coordination and improve tumor targeting. Structure-activity relationship studies identified DG3 as the optimal lipopeptide, enabling high drug-loading LNPs (hDDLN) with good stability and delivery efficiency. Pt/hDDLN showed a superior therapeutic index over conventional liposomes in vitro and in vivo. Combining Pt/hDDLN with the stimulator of interferon genes (S

Deep-Penetrating Transdermal Lipopeptide Liposomes for Sustained IL-17 Inhibition and Prevention of Psoriatic Recurrence.

Psoriasis is a multisystemic, immune-mediated inflammatory disease characterized by high recurrence rates upon treatment cessation. Biotherapeutics are highly anticipated for treating psoriasis via transdermal delivery. However, the topical administration of potent immunomodulators is fundamentally hindered by the stratum corneum barrier, stringent molecular size constraints, and rapid proteolytic degradation within the skin microenvironment. Here, we report the development of transdermal lipopeptide liposomes (TLLs) designed for the site-specific delivery of a double-bridged cyclic tetradecap

Identification of a Pair of Linear or Cyclic Naturally Inspired Bifunctional Lipopeptide Antibiotics That Overcome Antimicrobial Resistance.

The emergence of multidrug-resistant pathogens presents a major clinical challenge worldwide. The underexplored bacterial genus Aquimarina harbors a large biosynthetic potential for the discovery of new classes of antibiotics with distinctive modes of action, which can be unearthed in a scalable manner using a synthetic bioinformatic natural product approach. Here, the discovery of a pair of linear or cyclic bifunctional cationic lipopeptide antibiotics, aquicidine L and aquicidine C4, with opposite antibacterial spectra, is reported, which are chemically synthesized on the basis of distinct s

New Cyclic Lipopeptide Compounds for Efficiently Inhibiting Cellular Inflammation and Collagen-Induced Arthritis in Mice.

Rheumatoid arthritis (RA) is a systemic autoimmune disease, and a large number of patients do not respond well to existing treatment strategies. Our previous report has discovered that the cyclic lipopeptide (CLP) daptomycin (DAP) has a good suppressive arthritis activity in mice. In this study, we have designed and synthesized five novel DAP-derived CLPs by structural optimization on the loop of DAP and further studied their anti-RA effects in vitro and in vivo. Especially, CLP-d2 has better anti-RA effect than DAP due to its better pharmacokinetic parameters and peptide stability. Mechanisti

Stalobacin: Discovery of Novel Lipopeptide Antibiotics with Potent Antibacterial Activity against Multidrug-Resistant Bacteria.

A novel lipopeptide antibiotic, stalobacin I (1), was discovered from a culture broth of an unidentified Gram-negative bacterium. Stalobacin I (1) had a unique chemical architecture composed of an upper and a lower half peptide sequence, which were linked via a hemiaminal methylene moiety. The sequence of 1 contained an unusual amino acid, carnosadine, 3,4-dihydroxyariginine, 3-hydroxyisoleucine, and 3-hydroxyaspartic acid, and a novel cyclopropyl fatty acid. The antibacterial activity of 1 against a broad range of drug-resistant Gram-positive bacteria was much stronger than those of "last res

Lipid composition in a strain of Bacillus subtilis, a producer of iturin A lipopeptides that are active against uropathogenic bacteria.

Urinary tract infections are a common disease in humans. Therefore, new methods are needed to destroy biofilms that are formed by uropathogens. Iturin A lipopeptides (LPs) C14 and C15 are potent biosurfactants synthetized by the Bacillus subtilis I'1a strain. The biological activity of extracted LPs was confirmed by examining extracts from I'1a cultures against uropathogenic bacteria that had been isolated from biofilms on urinary catheters. Compared with cultures of DSM 3257, which produce surfactin at a relatively low level, the extract obtained from strain I'1a exhibited a greater inhibitor
