A Bio-Hybrid ZnO Nanoplatform Functionalized with Antimicrobial Peptide/Protein Enhances Multimodal Bacterial Eradication and Accelerated Wound Healing.

The increasing prevalence of bacterial infections, along with the persistence of biofilms and impaired wound healing, underscores the urgent need for advanced antimicrobial strategies. In the current study, ZnO nanoparticles (NPs) were synthesized by the sol-gel method and functionalized with PEG to improve the colloidal stability and biocompatibility. The PEG-functionalized ZnO NPs were then conjugated with the antimicrobial peptide nisin and the enzyme lysozyme to obtain two distinct nanoconjugates: nisin-PEG-ZnO and lysozyme-PEG-ZnO. Extensive physicochemical analyses, including Fourier-tra

Characteristics of Biofilms Formed by Co-Culture of Listeria monocytogenes with Pseudomonas aeruginosa at Low Temperatures and Their Sensitivity to Antibacterial Substances.

We assessed the properties of biofilms (BFs) formed by mono- and co-cultures of Listeria monocytogenes and Pseudomonas aeruginosa (L+P-BF) at low temperatures and examined their sensitivity to several antibacterial substances. L. monocytogenes viable counts comprised only 1-10% of total L+P-BF viable counts at 10℃ and 15℃, indicating the significant prevalence of P. aeruginosa in co-cultures. L+P-BF formed at 10℃ and 15℃ showed very high resistance to antibiotics and NaClO. Examination of the effects of nattokinase and nisin, natural food additives with antibacterial properties, showed that th

Food-grade expression of Helicobacter pylori ureB subunit in Lactococcus lactis and its immunoreactivity.

Helicobacter pylori is the principal cause of chronic active gastritis, peptic ulcer, and gastric cancer. To develop an oral vaccine against H. pylori infection, we had expressed the H. pylori ureB gene (Genbank accession no. FJ436980) in nisin-controlled expression vectors using Lactococcus lactis NZ3900 as host. The ureB gene was amplified by PCR from a H.pylori strain MEL-Hp27. Then the ureB gene was fused translationally downstream of the nisin-inducible promoter nisA in a L. lactis plasmid pNZ8149. Lactose utilization based on the complementation of the lacF gene was used as a dominant se

Comparative genomic analysis shows that Streptococcus suis meningitis isolate SC070731 contains a unique 105K genomic island.

Streptococcus suis (SS) is an important swine pathogen worldwide that occasionally causes serious infections in humans. SS infection may result in meningitis in pigs and humans. The pathogenic mechanisms of SS are poorly understood. Here, we provide the complete genome sequence of S. suis serotype 2 (SS2) strain SC070731 isolated from a pig with meningitis. The chromosome is 2,138,568bp in length. There are 1933 predicted protein coding sequences and 96.7% (57/59) of the known virulence-associated genes are present in the genome. Strain SC070731 showed similar virulence with SS2 virulent strai

Pneumolysin-responsive liposomal platform for selective treatment of Streptococcus pneumoniae.

The bacterium Streptococcus pneumoniae has become a leading cause of meningitis, sepsis, and bacterial pneumonia worldwide, with increased prevalence of antibiotic-resistant serotypes serving to exacerbate the issue. The main factor responsible for colonization and immune response escape in pneumococcal infections is the secreted molecule pneumolysin, which is a subset within a family of related toxins that form transmembrane pores in biological membranes through cholesterol recognition and binding. The conserved activity and structure of pneumolysin between all observed S. pneumoniae serotype

Exposure and resistance to lantibiotics impact microbiota composition and function.

The intestinal microbiota is composed of hundreds of distinct microbial species that interact with each other and their mammalian host. Antibiotic exposure dramatically impacts microbiota compositions and leads to acquisition of antibiotic-resistance genes. Lantibiotics are ribosomally synthesized and post-translationally modified peptides produced by some bacterial strains to inhibit the growth of competing bacteria. Nisin A is a lantibiotic produced by Lactococcus lactis that is commonly added to food products to reduce contamination with Gram-positive pathogens. Little is known, however, ab

Antibacterial activity against porcine respiratory bacterial pathogens and in vitro biocompatibility of essential oils.

Bacterial respiratory infections affecting pigs such as pneumonia, pleuropneumonia, and pleurisy, are a major health concern in the swine industry and are associated with important economic losses. This study aimed to investigate the antibacterial activities of essential oils against major swine respiratory pathogens with a view to developing a potential alternative to antibiotics. Their synergistic interactions with the bacteriocin nisin was also examined. Lastly, we assessed the in vitro biocompatibility of the most efficient essential oils using a pig tracheal epithelial cell line. Of the n

Exploring the Activity of Bacteriocins Against Bacterial Biofilms Using a Simple and Inexpensive Assay.

There is an urgent need for novel antimicrobials that can combat antibiotic resistant bacteria. Biofilms constitute a particular niche where bacteria display an intrinsic higher resistance to antibiotics and that favors resistance appearance and spread among species. However, the susceptibility tests for sessile bacteria are not well standardized. This work provides a high-throughput, affordable and simple methodology to grow biofilms on a three-dimensional surface represented by glass beads placed in 96-well microtiter plates. Two Gram-positive and two Gram-negative bacterial species were tes

Absence of Synergism between a Dual-AMP Biogel and Antibiotics Used as Therapeutic Agents for Diabetic Foot Infections.

Diabetic foot infections (DFIs) are frequently linked to diabetic-related morbidity and death because of the ineffectiveness of conventional antibiotics against multidrug-resistant bacteria. Pexiganan and nisin A are antimicrobial peptides (AMPs), and their application may complement conventional antibiotics in DFI treatment. A collagen 3D model, previously established to mimic a soft-tissue collagen matrix, was used to evaluate the antibacterial efficacy of a guar gum gel containing pexiganan and nisin alone and combined with three antimicrobials toward the biofilms of Staphylococcus aureus a

Use of commercially available antimicrobial compounds for prevention of Listeria monocytogenes growth in ready-to-eat minced tuna and salmon roe during shelf life.

Listeria monocytogenes found in minced tuna and fish roe can cause listeriosis. These products are classified in category B according to the Codex Alimentarius Commission, i.e., ready-to-eat foods in which L. monocytogenes growth can occur. We investigated the effectiveness of nisin and other commercially available antimicrobial compounds (lysozyme, ε-polylysine, and chitosan) for prevention of L. monocytogenes growth during the expected shelf life of raw minced tuna and salmon roe products. Food samples inoculated with L. monocytogenes were incubated with each antimicrobial at 10°C for 7 days

The effect of MTADN on 10 Enterococcus faecalis isolates and biofilm: an in vitro study.

INTRODUCTION: Enterococcus faecalis is frequently isolated from both secondary and persistent root canal infections. MTAD is an effective intracanal irrigant, but its bactericidal activity requires improvement. The goal of this study was to investigate whether nisin, an antibacterial peptide, improves the antibacterial and antibiofilm activities of MTAD against E. faecalis clinical isolates. METHODS: Ten E. faecalis strains were isolated from root canals with persistent periapical lesions. The antibacterial activity of MTAD, MTADN (nisin in combination with doxycycline), and MTAN (nisin in pl

Isolation and Evaluation of Anti-Listeria Lactococcus lactis from Vegetal Sources.

This chapter describes methods used to isolate, identify, and partially characterize lactic acid bacteria (LAB) which exhibit inhibitory activity against Listeria monocytogenes from foods. Vegetal (plant based) sources are rich in naturally occurring LAB and therefore provide an easily accessible source of strains with potential antimicrobial activity for use in food-processing applications. From our previous work, the majority of LAB with inhibitory activity against L. monocytogenes were identified as generally recognized as safe (GRAS) Lactococcus lactis. Although these bacteria are most com

Synergy Assessment of Four Antimicrobial Bioactive Compounds for the Combinational Treatment of Bacterial Pathogens.

Antimicrobial resistance (AMR) has become a topic of great concern in recent years, with much effort being committed to developing alternative treatments for resistant bacterial pathogens. Drug combinational therapies have been a major area of research for several years, with modern iterations using combining well-established antibiotics and other antimicrobials with the aim of discovering complementary mechanisms. Previously, we characterised four GRAS antimicrobials that can withstand thermal polymer extrusion processes for novel medical device-based and therapeutic applications. In the pres

Inactivation of Bacteria Using Bioactive Nanoparticles and Alternating Magnetic Fields.

Foodborne pathogens are frequently associated with risks and outbreaks of many diseases; therefore, food safety and processing remain a priority to control and minimize these risks. In this work, nisin-loaded magnetic nanoparticles were used and activated by alternating 10 and 125 mT (peak to peak) magnetic fields (AMFs) for biocontrol of bacteria Listeria innocua, a suitable model to study the inactivation of common foodborne pathogen L. monocytogenes. It was shown that L. innocua features high resistance to nisin-based bioactive nanoparticles, however, application of AMFs (15 and 30 min expo

Microbiota-derived lantibiotic restores resistance against vancomycin-resistant Enterococcus.

Intestinal commensal bacteria can inhibit dense colonization of the gut by vancomycin-resistant Enterococcus faecium (VRE), a leading cause of hospital-acquired infections1,2. A four-strained consortium of commensal bacteria that contains Blautia producta BPSCSK can reverse antibiotic-induced susceptibility to VRE infection3. Here we show that BPSCSK reduces growth of VRE by secreting a lantibiotic that is similar to the nisin-A produced by Lactococcus lactis. Although the growth of VRE is inhibited by BPSCSK and L. lactis in vitro, only BPSCSK colonizes the colon and reduces VRE density in vi

A self-unfolding proximity enabling device for oral delivery of macromolecules.

Oral delivery of macromolecules remains highly challenging due to their rapid degradation in the gastrointestinal tract and poor absorption across the tight junctions of the epithelium. In the last decade, researchers have investigated several medical devices to overcome these challenges using various approaches, some of which involve piercing through the intestine using micro and macro needles. We have developed a new generation of medical devices called self-unfolding proximity enabling devices, which makes it possible to orally deliver macromolecules without perforating the intestine. These

Establishment and application of nano-liposomes system for targeted therapy of methicillin-resistant Staphylococcus aureus enteritis.

Inspired by the damage caused by bacterial pore-forming toxins to cell membranes, this study utilized the film-ultrasound dispersion method and polyethylene glycol (PEG) modification to prepare resveratrol and nisin-loaded nanoliposomes (RN-NPs) using soy lecithin as the main encapsulating material. These RN-NPs were designed for targeted therapy against Methicillin-resistant Staphylococcus aureus (MRSA) enteritis. The synergistic inhibitory effects of resveratrol and nisin (RN) on MRSA were confirmed through SEM, FCM, and gel electrophoresis. In addition to being highly stable and resistant t

Mycoidesin, a novel lantibiotic, exhibits potent bacteriostatic activity against Listeria monocytogenes and effectively controls its growth in beef.

UNLABELLED: Listeria monocytogenes can cause severe listeriosis, with the consumption of contaminated food being an important route of its transmission. Biopreservatives can be used for the prevention and control of L. monocytogenes in food. In this study, we identified a novel lantibiotic, mycoidesin, with potent bacteriostatic activity against L. monocytogenes. It exhibited 4- to 16-fold higher bacteriostatic activity against the L. monocytogenes strains than nisin A. Analysis of the mode of action of mycoidesin revealed that it exerted bacteriostatic activity against L. monocytogenes ATCC 1
