Unoccluded retinol penetrates human skin in vivo more effectively than unoccluded retinyl palmitate or retinoic acid.

The formation of all-trans retinoic acid is an oxidative process whereby retinol is converted to retinaldehyde and then to retinoic acid. Because retinol causes qualitative molecular changes similar to those produced by retinoic acid, we compared potency of retinol, retinaldehyde, and retinyl palmitate to retinoic acid and assessed the effects of occlusion. Retinoids were prepared in an experimental vehicle of 95% ethanol:propylene glycol (7:3) with anti-oxidant. Induction of retinoic acid 4-hydroxylase activity was the end point for comparison. Retinoic acid concentrations from 0.001% to 0.05

Titration of numbers of human-derived Mycobacterium leprae required to progressively oxidize 14C-palmitic acid and release 14CO2.

Mycobacterium leprae was isolated from skin-punch biopsies of 2 untreated lepromatous leprosy patients. The bacteria were enumerated, diluted 10-fold and cultured in Middlebrook 7H9 medium supplemented with albumin, dextrose, catalase and 14C-palmitic acid. The cultures were incubated at 33 degrees C in a modified Buddemeyer radiorespiratory detection vessel. Those cultures containing at least 10(7) mycobacteria demonstrated a progressive evolution of 14CO2.

Adipocyte-Derived Palmitic Acid Promotes Breast Cancer Malignancy via ZDHHC15-Mediated S-Palmitoylation of PPARγ.

Peroxisome proliferator-activated receptor gamma (PPARγ), a well-characterized regulator of adipogenesis, also functions as a tumor suppressor in breast cancer. However, the PPARγ agonist rosiglitazone has shown limited clinical efficacy, highlighting the importance of understanding PPARγ regulation within the tumor microenvironment. Here, we demonstrate that adipocyte-derived palmitic acid promotes PPARγ S-palmitoylation via zinc finger DHHC-domain containing protein 15 (ZDHHC15), as confirmed by acyl-biotin exchange (ABE) assay. Immunoblot, RT-qPCR, and immunoprecipitation further revealed t

Evaluation of absorption kinetics and bioavailability of palmitic acid and 2-palmitoylglycerol, a metabolite of interesterified fat, radiolabeled with 99m-technetium.

Interesterified fat (IF) has replaced partially hydrogenated fats because it does not generate trans isomers, but it may affect lipid metabolism due to structural changes in triacylglycerols (TAGs). Since chemical interesterification alters TAG configuration and influences digestion and absorption, methodological approaches capable of tracking lipid metabolites are needed. In this study, we investigated the absorption kinetics and systemic distribution of IF metabolites using a radiolabeling strategy. Eight-week-old Swiss mice received via gavage: PA group-3.7 MBq of 99mTc-palmitic acid (99mTc

CEBPG-Mediated Palmitic Acid Adaptation of Cancer-Associated Fibroblasts Drives Metastasis of Oral Squamous Cell Carcinoma.

Fatty acids (FAs) create a pro-metastatic niche in multiple cancers, but how the tumor microenvironment (TME) counteracts FA stress remains unclear. Here, we found that the FA transporter CD36 was upregulated in myofibroblastic cancer-associated fibroblasts (myoCAFs), where it correlated with the metastasis of oral squamous cell carcinoma (OSCC). Among five predominant FA species enriched in OSCC tissues, palmitic acid (PA) potently activated myoCAF phenotypes across 2D, 3D, and organoid co-culture models. In vivo, PA promoted lymph node metastasis in orthotopic tumors comprising OSCC cells an

Released palmitic acid-mediated TLR4/NF-κB activation enhances the virulence of Bordetella pertussis MT28 lineage.

Whooping cough has resurged globally despite high vaccination coverage. In China, a macrolide-resistant (MR) Bordetella pertussis lineage carrying the high-virulence ptxP3 allele, termed ptxP3 MR-MT28 (MT28), has been increasingly reported as a predominant circulating lineage, although the factors underlying its expansion remain unclear. By integrating epidemiological surveillance with genomic, phenotypic, and in vitro and in vivo infection analyses of representative clinical isolates, we demonstrate that MT28 strains exhibit enhanced colonization capacity and heightened inflammatory potential

Unveiling the Mechanism of Action of Palmitic Acid, a Human Topoisomerase 1B Inhibitor from the Antarctic Sponge Artemisina plumosa.

Cancer remains a leading cause of death worldwide, highlighting the urgent need for novel and more effective treatments. Natural products, with their structural diversity, represent a valuable source for the discovery of anticancer compounds. In this study, we screened 750 Antarctic extracts to identify potential inhibitors of human topoisomerase 1 (hTOP1), a key enzyme in DNA replication and repair, and a target of cancer therapies. Bioassay-guided fractionation led to the identification of palmitic acid (PA) as the active compound from the Antarctic sponge Artemisina plumosa, selectively inh

BI-3231, an enzymatic inhibitor of HSD17B13, reduces lipotoxic effects induced by palmitic acid in murine and human hepatocytes.

17-β-hydroxysteroid dehydrogenase 13 (HSD17B13), a lipid droplet-associated enzyme, is primarily expressed in the liver and plays an important role in lipid metabolism. Targeted inhibition of enzymatic function is a potential therapeutic strategy for treating steatotic liver disease (SLD). The present study is aimed at investigating the effects of the first selective HSD17B13 inhibitor, BI-3231, in a model of hepatocellular lipotoxicity using human cell lines and primary mouse hepatocytes in vitro. Lipotoxicity was induced with palmitic acid in HepG2 cells and freshly isolated mouse hepatocyte

GPAT1 Activity and Abundant Palmitic Acid Impair Insulin Suppression of Hepatic Glucose Production in Primary Mouse Hepatocytes.

BACKGROUND: Glycerol-3-phosphate acyltransferase (GPAT) activity is correlated with obesity and insulin resistance in mice and humans. However, insulin resistance exists in people with normal body weight, and individuals with obesity may be metabolically healthy, implying the presence of complex pathophysiologic mechanisms underpinning insulin resistance. OBJECTIVE: We asked what conditions related to GPAT1 must be met concurrently for hepatic insulin resistance to occur. METHODS: Mouse hepatocytes were overexpressed with GPATs via adenoviral infection or exposed to high or low concentration

Period circadian clock 3 inhibits palmitic acid-induced oxidative stress and inflammatory factor secretion in podocytes.

OBJECTIVES: High fat-induced podocyte injury is one of the important factors leading to obesity related nephropathy (ORG), but the mechanism is not clear. This study aims to explore the mechanism of period circadian clock 3 (PER3) in the oxidative stress and inflammation induced by palmitic acid (PA) in podocytes. METHODS: The C57BL/6J mice were fed with chow and high-fat diet for 16 weeks. The PER3 expression in kidney tissues were detected in the normal body weight group and the obesity group. The PER3 mRNA and protein expression were detected after the podocytes were induced with different

Differential response of liver sinusoidal endothelial cells and hepatocytes to oleic and palmitic acid revealed by Raman and CARS imaging.

Excess circulating fatty acids contribute to endothelial dysfunction that subsequently aggravates the metabolic conditions such as fatty liver diseases. However, the exact mechanism of this event is not fully understood, and the investigation on the effect of a direct exposure to fatty acids together with their subsequent fate is of interest. In this work we employed a chemically specific and label-free techniques such as Raman and CARS microscopies, to investigate the process of lipid droplets (LDs) formation in endothelial cells and hepatocytes after exposure to oleic and palmitic acid. Forsythiae Fructus polysaccharides mitigate DSS colitis by modulating gut microbiota-derived palmitic acid and PI3K/AKT pathway.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder with limited therapeutic options, and the contribution of microbiota-associated metabolites to disease progression remains incompletely understood. Here, we demonstrate that Forsythia polysaccharides (FPs) exert protective effects against dextran sulfate sodium (DSS)-induced colitis through coordinated regulation of gut microbiota, microbial metabolism, and inflammatory signaling. FPs treatment markedly alleviated colonic injury and inflammation, restored intestinal barrier integrity, and remodeled the gut microbial community.

Investigating the Protective Effects of Platycodin D on Non-Alcoholic Fatty Liver Disease in a Palmitic Acid-Induced In Vitro Model.

The occurrence of non-alcoholic fatty liver disease (NAFLD) has been increasing at an alarming rate worldwide. Platycodon grandiflorum is widely used as a traditional ethnomedicine for the treatment of various diseases and is a typical functional food that can be incorporated into the everyday diet. Studies have suggested that platycodin D (PD), one of the main active ingredients in Platycodon grandiflorum, has high bioavailability and significantly mitigates the progress of NAFLD, but the underlying mechanism of this is still unclear. This study aims to investigate the therapeutic effect of P

Advanced Liver Fibrosis Is Independently Associated with Palmitic Acid and Insulin Levels in Patients with Non-Alcoholic Fatty Liver Disease.

Changes in lipid metabolism occur during the development and progression non-alcoholic fatty liver disease (NAFLD). However, the fatty acid (FA) profile in red blood cells (RBC) from patients with liver fibrosis remains unexplored. Thus, the goal of this study was to evaluate the fatty acid profile in RBC, dietary lipid intake and insulin resistance indicators in patients with NAFLD, according to the degree of hepatic fibrosis. Using elastography, patients were classified with (n = 52) and without (n = 37) advanced liver fibrosis. The fatty acid profile in RBC was analyzed using gas chromatogr

Effect of sulfo-N-succinimidyl palmitate on the rat heart: myocardial long-chain fatty acid uptake and cardiac hypertrophy.

Abnormal long-chain fatty acid metabolism has been suggested as having a role in the genesis of certain cardiac diseases, and depressed myocardial long-chain fatty acid uptake has been clinically demonstrated in some patients with hypertrophic cardiomyopathy. However, the site where long-chain fatty acid metabolism is affected in cardiomyopathy remains unclear. Although cardiac hypertrophy is reported to be induced in rats by a fat-free diet, little is known of the consequences of depressed myocardial long-chain fatty acid uptake. Sulfo-N-succinimidyl derivatives of long-chain fatty acids have

The effects of consumption of interesterified fats rich in palmitic acid compared with stearic acid on intermediary markers of cardiometabolic disease risk: a randomized controlled trial in healthy adults.

BACKGROUND: Interesterified (IE) fats rich in palmitic acid (16:0) and stearic acid (18:0) are widely used hard fats. Comparative effects of commercially relevant palmitic or stearic acid-rich IE fat blends on cardiometabolic disease (CMD) risk markers remain unclear. It was hypothesized that functionally matched palmitic and stearic acid-rich fats would have equivalent effects on plasma total:high-density lipoprotein (HDL) cholesterol ratio within a predefined equivalence margin of ±0.09. OBJECTIVES: This study aimed to compare the effects of diets enriched in palmitic or stearic acid, deliv
