Molecular mechanisms of light-induced photoreceptor apoptosis and neuroprotection for retinal degeneration.

Human retinal dystrophies and degenerations and light-induced retinal degenerations in animal models are sharing an important feature: visual cell death by apoptosis. Studying apoptosis may thus provide an important handle to understand mechanisms of cell death and to develop potential rescue strategies for blinding retinal diseases. Apoptosis is the regulated elimination of individual cells and constitutes an almost universal principle in developmental histogenesis and organogenesis and in the maintenance of tissue homeostasis in mature organs. Here we present an overview on molecular and cel

[Oxygen-dependent processes in the irradiated organism].

Interaction of oxygen with the X-rayed tissues, chemical substances and elements is one of the main mechanism of radioactive damage. Radiosensibilization by oxygen (oxygen effect) is most highly exhibited under the action of high and moderate radiation doses. However there is only scanty investigations and information about oxygen-dependent processes under low doses of internal and external radiation due to their intricate character. Radiation exerts also a great influence on oxygen metabolism and oxygen consumption level. Oxygen takes active part in post-radiation recovery both at the level o

Association of malignancy with rapid growth in early lesions induced by irradiation of rat skin.

Epithelial lesions induced by irradiation of rat skin were studied to determine a) the relationship of malignancy to dose, b) the types of lesions and circumstances leading to overt malignancy, and c) the growth rates of lesions progressing to malignancy versus those of lesions remaining benign. High doses of radiation were shown to be associated with the production of epidermal cancers, the maximum yield being obtained at 6,400 rads. Conversely, a peak yield of noncancerous lesions was obtained at 1,600 rads. This association between malignancy and high dose was consistent for cancers evolvin

The impact of vitreous humor: a new perspective on radiation-induced cataractogenesis.

PURPOSE: The precise mechanisms underlying radiation-induced cataractogenesis remain incompletely understood. Increased oxidative stress is known to play a central role in cataract pathogenesis. The vitreous humor contributes to maintaining the hypoxic environment of the lens by regulating oxygen pressure and containing antioxidants. This study aims to explore the effect of radiation-induced changes in the vitreous humor on lens health, with a particular focus on its cataractogenic potential. METHODS: In this experimental study, 12 New Zealand rabbits were utilized. A single 20-Gy dose of rad

Suplatast tosilate reduces radiation-induced lung injury in mice through suppression of oxidative stress.

PURPOSE: Although radiotherapy is important in the treatment of malignant thoracic tumors, it has harmful effects on healthy tissues. We previously showed that suplatast tosilate, an anti-allergic agent, scavenged reactive oxygen species (ROS), including hydroxyl radicals. Because ROS-mediated oxidative stress is involved in radiation-induced lung injury, we hypothesized that suplatast tosilate could reduce radiation-induced lung injury via suppression of oxidative stress. METHODS AND MATERIALS: Murine alveolar epithelial cells were irradiated with or without a medium containing suplatast tos

Dose Optimization Study of AEOL 10150 as a Mitigator of Radiation-Induced Lung Injury in CBA/J Mice.

AEOL 10150 is a catalytic metalloporphyrin superoxide dismutase mimic being developed as a medical countermeasure for radiation-induced lung injury (RILI). The efficacy of AEOL 10150 against RILI through a reduction of oxidative stress, hypoxia and pro-apoptotic signals has been previously reported. The goal of this study was to determine the most effective dose of AEOL 10150 (daily subcutaneous injections, day 1-28) in improving 180-day survival in CBA/J mice after whole-thorax lung irradiation (WTLI) to a dose of 14.6 Gy. Functional and histopathological assessments were performed as seconda

Regenerative effects of myogenic gene transfected MSC derived exosomes on radiation esophagitis.

BACKGROUND: Radiation esophagitis is a common adverse effect of radiotherapy for head and neck cancers, and is marked by irreversible damage and fibrosis of esophageal muscle tissue. Although mesenchymal stem cell (MSC) therapy is emerging as a promising approach for tissue regeneration, clinical translation remains challenging due to issues with cell viability and differentiation in vivo. This study evaluates the regenerative efficacy of exosomes derived from MSCs transfected with myogenic genes (MyoD, Myogenin, Myf6, referred to as Myo-MIX) using a murine model of radiation-induced esophagea

Presymptomatic microRNA-based biomarker signatures for the prognosis of localized radiation injury in mice.

The threat of nuclear or radiological events requires early diagnostic tools for radiation induced health effects. Localized radiation injuries (LRI) are severe outcomes of such events, characterized by a latent presymptomatic phase followed by symptom onset ranging from erythema and edema to ulceration and tissue necrosis. Early diagnosis is crucial for effective triage and adapted treatment, potentially through minimally invasive biomarkers including circulating microRNAs (miRNAs), which have been correlated with tissue injuries and radiation exposure, suggesting their potential in diagnosin

Control of radiation-induced emesis with promethazine, cimetidine, thiethylperazine, or naloxone.

Promethazine (2 mg/kg), cimetidine (4 mg/kg), thiethylperazine (0.86 mg/kg), and naloxone (0.08 mg/kg) were each evaluated for their ability to increase the threshold of radiation-induced emesis in the dog. Each dog was fed a can of dog food (ca 0.4 kg) and then injected IM with the appropriate drug 1 hour before being irradiated by a 60Co teletherapy unit. The total radiation dose given an individual dog was determined by an up-and-down exposure schedule. Dogs were then observed continuously for 10 hours while the number, time of onset, and duration of each emetic episode were monitored. The

Cynaroside alleviates radiation-induced intestinal injury by inhibiting dynamin 2.

Cynaroside (Cyn) exhibits unique anti-inflammatory, anti-apoptotic properties and diverse bioactivities; however, its role in mitigating radiation-induced intestinal injury (RIII) remains unclear. This study aimed to investigate the protective effects of Cyn against RIII and explore the underlying mechanisms. C57BL/6 mice were subjected to a single 12 Gy X-ray total abdominal irradiation (TAI) followed by Cyn gavage. Cyn exhibited dose-dependent protection against RIII. Treatment with Cyn improved survival rates, attenuated body weight loss, preserved colon length and intestinal architecture (

NVP-AUY922 relieves radiation-induced intestinal injury via regulating EPHX1.

As a common side effect of radiotherapy, radiation-induced intestinal injury (RIII) greatly affects the prognosis of patients and the efficacy of radiotherapy. Current therapeutic strategies for RIII are still very limited. Thus, the identification of effective radioprotective agents is of great importance. NVP-AUY922 is an HSP90 inhibitor with favorable anti-inflammatory and antioxidant activities. It has been proven to mitigate radiation-induced lung injury. However, its effects on the alleviation of RIII remain unclear. In this study, our data indicated that NVP-AUY922 remarkably increased

Management of postirradiation infection: lessons learned from animal models.

Ionizing radiation depresses host defenses and enhances susceptibility to local and systemic infection due to endogenous or exogenous microorganisms. Exposure of mice to a lethal dose of ionizing 60Co-gamma radiation induces a dose-related reduction in the number of both aerobic and anaerobic bacteria from 10(10-12) to 10(4-6) per gram of stool within 4 days. The number of anaerobic bacteria stays low, but the number of Enterobacteriaceae per gram of stool increases significantly up to 10(9) by the 12th day after irradiation. This increase is associated with bacterial translocation of these or

Wound Trauma Exacerbates Acute, but not Delayed, Effects of Radiation in Rats: Mitigation by Lisinopril.

The goal of this study is to understand and mitigate the effects of wounds on acute radiation syndrome (ARS) and delayed effects of acute radiation exposure (DEARE), for preparedness against a radiological attack or accident. Combined injuries from concomitant trauma and radiation are likely in these scenarios. Either exacerbation or mitigation of radiation damage by wound trauma has been previously reported in preclinical studies. Female WAG/RijCmcr rats received 13 Gy X-rays, with partial-body shielding of one leg. Within 2 h, irradiated rats and non-irradiated controls were given full-thick

The microlesion concept in HZE particle dosimetry.

High energy, high-Z (HZE) particles are present in high-altitude and high-inclination satellite orbits. Most of the HZE dose above LET = 200 keV/micrometer is due to Fe nuclei. Individual HZE particles can damage several cells adjacent to one another along the particle track in tissue. The outcome has been described as a "microlesion" by D. Grahn. The present study attempts to define conditions for microlesions in specific tissues, to seek biological evidence that microlesions are produced, and to evaluate the microlesion as a potentially useful unit of dose in assessing hazards to spaceworker

[Pathogenesis and pathophysiology of injuries after radiotherapy of neoplasms].

The dose which can be given in radiotherapy and, thus, the chance of cancer control is primarily limited by the risk of producing severe chronic side effects in the uninvolved adjacent normal tissues. Only recently the challenge that chronic radiation injury poses to radiobiology has been fully acknowledged and experimental research in laboratory animals has started. Still, very little is known about the pathogenesis, the pathophysiology and the pathology of chronic normal tissue injury of most organs.
