Activation of Trk neurotrophin receptors by glucocorticoids provides a neuroprotective effect.

Glucocorticoids (GCs) display both protective and destructive effects in the nervous system. In excess, GCs produce neuronal damage after stress or brain injury; however, the neuroprotective effects of adrenal steroids also have been reported. The mechanisms that account for the positive actions are not well understood. Here we report that GCs can selectively activate Trk receptor tyrosine kinases after in vivo administration in the brain and in cultures of hippocampal and cortical neurons. Trk receptors are normally activated by neurotrophins, such as NGF and brain-derived neurotrophic factor

TRK Fusion Cancers in Children: A Clinical Review and Recommendations for Screening.

Chromosomal translocations involving the NTRK1, NTRK2, and NTRK3 genes (TRK fusions), which encode the neurotrophin tyrosine kinase receptors TRKA, TRKB, and TRKC, can result in constitutive activation and aberrant expression of TRK kinase. Certain cancers almost universally harbor TRK fusions, including infantile fibrosarcoma, cellular congenital mesoblastic nephroma, secretory breast cancer, and mammary analog secretory carcinoma of the salivary gland. TRK fusions have also been identified at lower frequencies across a broad range of other pediatric cancers, including undifferentiated sarcom

Secretory Carcinoma of the Thyroid: A Case Report and Update of Literature.

Primary secretory carcinoma (SC) of the thyroid gland is a rare neoplasm, characterized by the presence of oncogenic ETV6::NTRK3 fusions, which are amenable to tropomyosin receptor kinase (TRK) inhibitor therapy. Despite its morphologic, immunophenotypic, and genetic similarities to SC of the salivary and mammary glands, diagnostic pitfalls may arise in differentiating from papillary thyroid carcinoma due to overlapping features such as papillary growth, nuclear irregularity, and variable expression of PAX8. Tumor misclassification may lead to delayed consideration of molecular testing and tar

Contribution of RET, NTRK3 and EDN3 to the expression of Hirschsprung disease in a multiplex family.

BACKGROUND: Hirschsprung disease (HSCR) is a developmental disorder caused by a defect in the neural crest neuroblast migration process. It is considered to be a paradigm of complex disorders, with many loci contributing to manifestation of the disease. Although HSCR commonly appears as a sporadic trait, approximately 20% of HSCR cases are familial, with complex patterns of inheritance. METHOD: A multiplex HSCR family with an additive model of inheritance, in which the contribution of three genes (RET, NTRK3, EDN3) leads to the HSCR phenotype is reported. RESULTS AND DISCUSSION: The findings

Chemically diverse toxicants converge on Fyn and c-Cbl to disrupt precursor cell function.

Identification of common mechanistic principles that shed light on the action of the many chemically diverse toxicants to which we are exposed is of central importance in understanding how toxicants disrupt normal cellular function and in developing more effective means of protecting against such effects. Of particular importance is identifying mechanisms operative at environmentally relevant toxicant exposure levels. Chemically diverse toxicants exhibit striking convergence, at environmentally relevant exposure levels, on pathway-specific disruption of receptor tyrosine kinase (RTK) signaling

Congenital absence of corticospinal tract does not severely affect plastic changes of the developing postnatal spinal cord.

The arrival and refinement of corticospinal afferents are likely to influence the maturation of the spinal cord and sensory-motor networks. To understand this better, we studied the revision of monosynaptic muscle afferents, the expression of activity-related genes, neurotrophins and their receptors in the cervical spinal cord from postnatal day (P) 0 to 21. We compared control and Celsr3|Emx1 mice, in which corticospinal axons never develop. The corticospinal tract (CST), labeled by anti-protein kinase C gamma (PKCγ) antibody in the dorsal funiculus, increased gradually in the control, but wa

5-Aza-2'-deoxycytidine in the medial prefrontal cortex regulates alcohol-related behavior and Ntf3-TrkC expression in rats.

Recent studies have indicated that DNA methylation plays an important role in the development of alcohol abuse. 5-Aza-2'-deoxycytidine (5-Aza-dc), an inhibitor of DNA methyltransferases, was FDA approved for myelodysplastic syndrome treatment. However, it is unclear whether 5-Aza-dc is involved in alcohol abuse. In this study, using a chronic alcohol exposure model in rats, 5-Aza-dc was injected into the medial prefrontal cortex (mPFC). Alcohol-drinking behavior and the anxiety related behavior were evaluated by two-bottle choice and open field test. We found that 5-Aza-dc injection into the m

Real-World Evidence for Oncology Patients With Rare NTRK Gene Fusions: Final Results From the German Multicenter Patient Cohort Study REALTRK.

PURPOSE: TRK inhibitors (TRKis) have transformed the therapeutic landscape for patients with neurotrophic tyrosine receptor kinase (NTRK) gene fusion-positive tumors. However, approval of TRKis is based on evidence derived mainly from small, pooled, single-arm clinical trial cohorts. The REALTRK registry aims to describe real-world molecular diagnostic practices, treatment patterns, and clinical outcomes for adult patients with NTRK fusion-positive cancers. PATIENTS AND METHODS: The REALTRK registry was a multicenter cohort study that included adults with advanced solid tumors harboring NTRK1

Primary NTRK-rearranged spindle cell sarcoma of the colon with ETV6-NTRK3 fusion.

NTRK (neurotrophic receptor tyrosine kinase)-rearranged spindle cell tumours are rare sarcomas predominantly occurring in the extremities and trunk, with only rare gastrointestinal tract cases reported to date. A man in his late 30s presented with abdominal pain. Given the CT evidence of a large transmural tumour requiring radical resection irrespective of histological subtype, the multidisciplinary team proceeded directly to surgery without preoperative colonoscopy. Diagnosis was established through histopathology, immunohistochemistry (IHC) and next-generation sequencing (NGS). Right hemicol

Analgesic, anti-inflammatory and joint protective effects of ACD137, a selective negative allosteric modulator of TrkA, in models of chemotherapy-induced peripheral neuropathy and osteoarthritis.

OBJECTIVES: The neurotrophin receptor TrkA is a clinically and genetically validated target in pain signaling. Anti-nerve growth factor (NGF) monoclonal antibodies have shown clinical efficacy but with side effects such as rapidly progressing osteoarthritis limiting their use, potentially mediated via inhibition of NGF/p75NTR signaling. Therefore, we sought to identify novel and selective small molecule negative allosteric modulators of tropomyosin receptor kinase A (TrkA) and to verify their analgesic effect in vivo. METHODS: We have identified ACD137 as a potent and selective negative allos

Identification of NTRK3 fusions in plaque-like CD34-positive dermal fibroma.

AIMS: Plaque-like CD34-positive dermal fibroma (PDF), previously termed medallion-like dermal dendrocyte hamartoma, is a rare CD34-positive superficial spindle cell fibroblastic tumour that may closely mimic dermatofibrosarcoma protuberans. Recent studies have identified recurrent kinase gene fusions in a subset of congenital and paediatric CD34-positive plaque-like superficial spindle cell tumours with overlapping clinicopathologic features, highlighting an emerging molecular framework for this group. However, the molecular spectrum of lesions meeting classic histopathologic criteria for PDF

Central nervous system ganglioneuroblastoma harboring MYO5A-NTRK3 fusion.

Central nervous system (CNS) ganglioneuroblastoma is a rare neuroectodermal neoplasm and little is known about its clinical and biological features. Herein, we report a pediatric case of CNS ganglioneuroblastoma harboring MYO5A-NTRK3 fusion. The patient, a 4-year-old boy, underwent a partial resection of a supratentorial tumor that was histopathologically diagnosed as a CNS ganglioneuroblastoma. Treatment with radiotherapy was started per the St Jude Medulloblastoma 03 (SJMB03) protocol; however, the tumor progressed rapidly and radiotherapy was temporally discontinued. Meanwhile, the patient

Assessing targetable NTRK1/2/3 fusions in mesenchymal tumors via whole-exome RNA sequencing.

BACKGROUND: Neurotrophic receptor tyrosine kinase (NTRK) 1/2/3 fusions, though rare, are highly sought in cancer diagnostics due to tyrosine kinase inhibitors' histology-agnostic approval based on significant efficacy across tumor types. In mesenchymal tumors, these fusions are identified in five typical NTRK-rearranged mesenchymal tumors (NMT): infantile fibrosarcomas, lipofibromatosis-like neural tumors, NTRK-rearranged spindle cell neoplasms, and minority subsets of ALK-negative inflammatory myofibroblastic tumors and quadruple wild-type gastrointestinal stromal tumors. However, their occur

Nuclear staining for pan-Trk by immunohistochemistry is highly specific for secretory carcinoma of breast: pan-Trk in various subtypes of breast carcinoma.

AIMS: Secretory carcinoma of breast (SCB) typically harbours ETV6-NTRK3 gene fusion. Pan-Trk immunohistochemistry analysis (IHC) has been shown to be sensitive for SCB diagnosis. However, weak focal pan-Trk nuclear staining was previously found in 10% of non-secretory breast carcinomas. To further examine pan-Trk IHC specificity, we evaluated pan-Trk staining in various breast carcinoma subtypes. METHODS: The study cohort consisted of 346 invasive breast carcinomas (IBCs), including 8 SCBs and 48 triple-negative histological mimickers (36 metaplastic carcinomas, including 12 matrix-producing

NTRK Fusion Landscape in 195 Positive Cases of 17 Tumors: A Chinese Multicenter Retrospective Study With Optimized Pan-TRK Immunohistochemistry (IHC) Screening.

Neurotrophic tyrosine receptor kinase (NTRK) fusions are crucial in tumorigenesis and in guiding targeted therapy with TRK inhibitors. However, their rarity, fusion heterogeneity, and limitations of conventional pan-TRK immunohistochemistry (IHC) impede accurate clinical detection. This multicenter retrospective study analyzed 374 next-generation sequencing/fluorescence in situ hybridization-validated samples (195 NTRK positive and 179 NTRK negative) collected from 12 Chinese centers to investigate fusion heterogeneity and refine the interpretation of pan-TRK IHC. We developed an amplification
