Ursodeoxycholic acid (UDCA) is a safe and effective medical therapy for most patients with primary biliary cirrhosis (PBC), but some patients show an incomplete response. Silymarin is a potent antioxidant with immunomodulatory and antifibrotic properties. The aim of this study was to evaluate the safety and assess the efficacy of silymarin in patients with PBC who had shown a suboptimal response to UDCA. Twenty-seven patients with PBC who had been on UDCA (13-15 mg/kg/day) therapy for 7 to 221 months and had shown a persistent elevation of alkaline phosphatase activity at least 2 times the upp

We explored the possibility that silymarin (SY), a fraction from Silybum marianum, might protect against the effects of in utero exposure to ethanol upon subsequent social memory function. Three groups of 8 pregnant female Sprague-Dawley rats each were provided with a liquid diet containing 35% ethanol derived calories (EDC). One experimental group received a daily subcutaneous injection of 400 mg/kg SY, the second, a 400 mg/kg oral dose of SY, a third group was maintained on the 35% EDC diet. A fourth group served as the pair-fed control group. The liquid diet regimen was maintained throughou

A randomized double-blind study about the therapy of the cirrhosis of the liver shows a significant higher surviving rate of the alcoholic cirrhosis in the group treated with Silymarin. This result can be well explained by the protective influence of this substance against toxic injuries. The etiology of many chronic liver diseases is uncertain and therefore it is advisable to try the therapy also in other cases. The conditions of the study are exactly reported. The influence on the clinic of the disease and on the laboratory data obtained in many controls will be published later, because the

Alcoholic liver disease (ALD), caused by excessive alcohol abuse, encompasses a battery of liver conditions including fatty liver to more severe conditions such as alcoholic hepatitis and cirrhosis. Silymarin derived from the dried fruits of Silybum marianum can shield the liver cells from alcohol damage and prevent toxins from penetrating and damaging the liver. Overcoming the challenges of silymarin's poor solubility, low bioavailability, and limited absorption is crucial to fully realize its health benefits. Herein, spirulina platensis (SP) was used as a natural carrier for silymarin delive

Aluminum (Al) is abundant in the environment, and its toxicity is attributed to free radical formation and subsequent oxidative stress. While silymarin is a well-known antioxidant, its low water solubility and bioavailability limit its therapeutic effects. This study was designated to formulate silymarin chitosan nanoparticles (SM-CS-NPs) and evaluate its ameliorative effect against hepatotoxicity induced by aluminum chloride (AlCl3). SM-CS-NPs were prepared by ionotropic gelation method and characterized using different techniques. Rats were distributed into six groups (n=7/group), control, s

Bilirubin is considered to be one of the most potent endogenous antioxidants in humans. Its serum concentrations are predominantly affected by the activity of hepatic bilirubin UDP-glucuronosyl transferase (UGT1A1). Our objective was to analyze the potential bilirubin-modulating effects of natural polyphenols from milk thistle (Silybum marianum), a hepatoprotective herb. Human hepatoblastoma HepG2 cells were exposed to major polyphenolic compounds isolated from milk thistle. Based on in vitro studies, 2,3-dehydrosilybins A and B were selected as the most efficient compounds and applied either

It has been well established that silymarin has hepatoprotective and anti-fibrotic effects. But the mechanisms are poorly understood. In recent years, the role of Ly6Chi monocytes in liver fibrosis has been well demonstrated. Thus, in present study we aimed to investigate whether silymarin can relieve liver fibrosis by reducing Ly6Chi monocytes infiltration. The mouse model of liver fibrosis was established by injected with carbon tetrachloride (CCl4) via intraperitoneal repeatedly. Mice in silymarin group received silymarin treatment by gavage. Silymarin significantly reduced liver inflammati

OBJECTIVES: Renal ischemia/reperfusion (I/R) injury is associated with increased mortality and morbidity rates due to acute renal failure (ARF). Oxidative stress induced with renal I/R injury directly affects glomerular and tubular epithelium through reactive oxygen species. Several studies have been directed to the treatment of renal I/R injury. The aim of this study was to test the attenuation with silymarin (SM) treatment of renal I/R injury-induced morphological changes in the rat kidney.
METHODS: A total of 32 adult male Sprague-Dawley rats were evaluated in four groups. Group I (sham),

Methotrexate (MTX) is a medication that is frequently prescribed for the treatment of both malignant disorders and inflammatory pathologies. However, its use is limited by dose-dependent pulmonary toxicity. The present study investigated the protective effects of silymarin (SLM) against MTX-induced lung injury by evaluating apoptosis, oxidative stress, autophagy, inflammation and histopathological changes in rats. Twenty-eight Wistar albino rats were assigned to the Control, SLM (50 mg/kg, p. o.), MTX (20 mg/kg, i. p.), and MTX + SLM groups. MTX treatment led to a significant elevation in malo

Lead is a widespread environmental contaminant that induces multi-organ toxicity primarily through oxidative damage and inflammatory responses. Salivary glands particularly the parotid are increasingly recognized as vulnerable targets of metal-induced injury, with significant implications for oral and digestive health. This study investigated the protective effect of silymarin, a flavonolignan antioxidant extracted from milk thistle seeds, against lead acetate-induced parotid gland damage in rats. Forty adult male Wistar rats were allocated into four groups: untreated controls, silymarin only

Sodium valproate (SV) is a well-known anti-epileptic drug, also used to control convulsions, bipolar disorders and migraines. SV has been shown to induce liver toxicity in clinical subjects. Syringic acid (SA), a natural polyphenolic compound has potential antioxidant, anti-inflammatory and several beneficial effects. Therefore, in this study, we evaluated hepatoprotective effect of SA against SV-induced liver injury in rats. Wistar rats were treated with SV orally at a dose of 500 mg/kg, once daily, for 14 days. Another three groups of rats were administered with SV and concurrently treated w
