Regulation of sterol metabolism by gut microbiota and its relevance to disease.

Sterols play an indispensable role in maintaining cell membrane stability, regulating hormone synthesis, and preserving physiological homeostasis. Recently, the function of the gut microbiota in modulating host sterol metabolism has become the focus of extensive research. Nonetheless, the specific functions carried out by the gut microbiota in sterol metabolism and their health implications remain unclear due to a lack of comprehensive synthesis and analysis. This review aims to consolidate current perspectives regarding the sources and metabolic mechanisms of sterols, with an emphasis on the

Molecular Characterization of Sterol C4-Methyl Oxidase in Leishmania major.

Sterol biosynthesis requires the oxidative removal of two methyl groups from the C-4 position by sterol C-4-demethylase and one methyl group from the C-14 position by sterol C-14-demethylase. In Leishmania donovani, a CYP5122A1 (Cytochrome P450 family 5122A1) protein was recently identified as the bona fide sterol C-4 methyl oxidase catalyzing the initial steps of C-4-demethylation. Besides CYP5122A1, Leishmania parasites possess orthologs to ERG25 (ergosterol pathway gene 25), the canonical sterol C-4 methyl oxidase in Saccharomyces cerevisiae. To determine the contribution of CYP5122A1 and E

Sterol Composition Modulates the Response of Saccharomyces cerevisiae to Iron Deficiency.

Iron is a vital micronutrient that functions as an essential cofactor in multiple biological processes, including oxygen transport, cellular respiration, and metabolic pathways, such as sterol biosynthesis. However, its low bioavailability at physiological pH frequently leads to nutritional iron deficiency. The yeast Saccharomyces cerevisiae is extensively used to study iron and lipid metabolisms, as well as in multiple biotechnological applications. Despite iron being indispensable for yeast ergosterol biosynthesis and growth, little is known about their interconnections. Here, we used lipid

Use of Plant Sterol and Stanol Fortified Foods in Clinical Practice.

Plant sterols and stanols (PS) are natural, non-nutritive molecules that play a structural role in plant membranes similar to that of cholesterol in animal membranes and abound in seeds and derived oils. PS exert their physical effect of interference with micellar solubilization of cholesterol within the intestinal lumen and are marginally absorbed by enterocytes, with negiglible increases in circulating levels. The physiological role of PS in plants and their natural origin and non-systemic action, together with their cholesterol-lowering effect, make them an attractive option as non-pharmaco

The CAP protein superfamily: function in sterol export and fungal virulence.

CAP superfamily proteins, also known as sperm-coating proteins, are found in all kingdoms of life and have been implicated in a variety of physiological contexts, including immune defense in plants and mammals, sperm maturation and fertilization, fungal virulence, and toxicity of insect and reptile venoms as well as prostate and brain cancer. CAP family members are mostly secreted glycoproteins that are highly stable in the extracellular fluid. All members of the superfamily share a common CAP domain of approximately 150 amino acids, which adopts a unique α-β-α sandwich fold. The conserved str

Role of a disordered steroid metabolome in the elucidation of sterol and steroid biosynthesis.

In 1937 Butler and Marrian found large amounts of the steroid pregnanetriol in urine from a patient with the adrenogenital syndrome, a virilizing condition known to be caused by compromised adrenal secretion even in this pre-cortisol era. This introduced the concept of the study of altered excretion of metabolites as an in vivo tool for understanding sterol and steroid biosynthesis. This approach is still viable and has experienced renewed significance as the field of metabolomics. From the first cyclized sterol lanosterol to the most downstream product estradiol, there are probably greater th

Prevention of endotoxin-induced uveitis in rats by plant sterol guggulsterone.

PURPOSE: To investigate the anti-inflammatory effects of guggulsterone, an antioxidant and antitumor agent, in endotoxin-induced uveitis (EIU) in rats and to elucidate the underlying molecular mechanism or mechanisms related to ocular inflammation. METHODS: EIU was induced by subcutaneous injection of lipopolysaccharide (LPS; 150 μg) into Lewis rats treated with guggulsterone (30 mg/kg body weight, intraperitoneally) or its carrier. After 24 hours the rats were killed, eyes were enucleated, and aqueous humor (AqH) was collected. Numbers of infiltrating cells and levels of matrix metalloprotei

Factors regulating sterol biosynthesis in the rat pineal gland in vitro.

The effects of hormones, feeding and photoperiod on pineal sterol biosynthesis were studied in rat pineal glands incubated with 1-14C acetate. The rate of sterol biosynthesis was 7.1 nmol/g/2 h. A high synthesis rate was observed during mid-dark of a 12:12 LD cycle and under constant light. Melatonin, testosterone, and glucagon had no effect on sterol synthesis whereas estradiol increased it and insulin and norepinephrine treatments decreased it. Increased cAMP levels resulting from dibutyryl cAMP and theophylline were also found to be inhibitory. Dietary cholesterol did not alter the rate of

A case of cerebrotendinous xanthomatosis. II: The sterol content of a cataractous lens.

The cholestanol content of a cataractous lens nucleus from a patient with cerebrotendinous xanthomatosis (CTX) was quantified by gas chromatography-mass spectrometry and found to be 0.27 micrograms per mg freeze-dried lens tissue. The cholestanol-cholesterol ratio of 1.7% in the lens nucleus was similar to that in the serum of the CTX patient. The cholestanol content and cholestanol-cholesterol ratio in the CTX lens were approximately four-fold and six-fold greater respectively than the mean levels found in three senile cataractous lens nuclei analysed simultaneously for comparative purposes.

Oxidative demethylation in sterol metabolism. Inhibition by NADH, a required cofactor.

Oxidative demethylation of 4-methyl sterols by rat liver microsomes has been shown to be inhibited by low levels of NADH. Using snake venom-treated microsomes to remove endogenous NADH-utilizing activities, we observed inhibition of overall demethylation at concentrations of NADH as low as 2.5 microM. At 20 microM NADH, overall demethylation had been reduced to 60% of optimal activity. In the presence of NADH, oxidative metabolism of [30,31-14C]4,4-dimethyl-5 alpha-cholest-7-en-3 beta-ol resulted in formation of an intermediate which was metabolized anaerobically to 14CO2 in the presence of NA

Stoichiometry of 4-methyl sterol oxidase of rat liver microsomes.

The stoichiometry of 4-methyl sterol oxidase has been investigated by concurrent assays of rates of oxygen consumption, oxidation of reduced pyridine nucleotide, and formation of steroid 4alpha-oic acid, which is the oxidized product of attack of 4-methyl sterol precursors of cholesterol. The basal, steroid-independent rates of oxidation of alpha-NADH and alpha-NADH-dependent oxygen consumption by rat liver microsomes are about 10 to 15% of the rates observed with beta-NADH. Thus, alpha-NADH is substituted for beta-NADH; alpha-NADH oxidation is observed spectrophotometrically. The slow rate of

HIV protease inhibitor induces fatty acid and sterol biosynthesis in liver and adipose tissues due to the accumulation of activated sterol regulatory element-binding proteins in the nucleus.

The mechanism by which human immunodeficiency virus (HIV) protease inhibitor therapy adversely induces lipodystrophy and hyperlipidemia has not been defined. This study explored the mechanism associated with the adverse effects of the prototype protease inhibitor ritonavir in mice. Ritonavir treatment increased plasma triglyceride and cholesterol levels through increased fatty acid and cholesterol biosynthesis in adipose and liver. Ritonavir treatment also resulted in hepatic steatosis and hepatomegaly. These abnormalities, which were especially pronounced after feeding a Western type high fat

Streamlined LC-HRMS-based determination of sterols and triterpenic alcohols in edible oils for authentication purposes.

Sterol analysis is a key parameter for assessing the authenticity and quality of olive oil, as defined by official methods. Nevertheless, these protocols are labor-intensive and require multiple preparation steps, including sample fractionation and derivatization. In the present work, a simplified analytical methodology is proposed for the determination of free sterols and triterpenic alcohols, eliminating these time-consuming procedures. The method is based on the direct analysis of saponified extracts, prepared according to the official protocol, followed by a single dilution step and analys

Physiological Functions of Side-Chain-Retaining Sterols in the Brain and Their Roles in Neurodegenerative Diseases.

Although the brain comprises only 2% of total body weight, it contains approximately 23% of the total cholesterol of the body. In the brain, cholesterol plays a critical role as a structural component of cell membranes and myelin sheaths. However, the blood-brain barrier restricts cholesterol influx from the systemic circulation into the brain. As a result, the brain synthesizes cholesterol de novo and regulates its metabolism independently. Desmosterol, a cholesterol precursor produced during cholesterol biosynthesis, and cholesterol metabolites, 24S-hydroxycholesterol and chenodeoxycholic ac

Response of different PTH assays to therapy with sevelamer or CaCO3 and active vitamin D sterols.

Amino-terminally truncated parathyroid hormone (PTH) fragments are detected to differing degrees by first- and second-generation immunometric PTH assays (PTH-IMAs), and acute changes in serum calcium affect the proportion of these fragments in circulation. However, the effect of chronic calcium changes and different vitamin D doses on these PTH measurements remains to be defined. In this study, 60 pediatric dialysis patients, aged 13.9 +/- 0.7 years, with secondary hyperparathyroidism were randomized to 8 months of therapy with oral vitamin D combined with either calcium carbonate (CaCO(3)) or

Use of active vitamin D sterols in patients with chronic kidney disease, stages 3 and 4.

This paper reviews randomized controlled trials and other reported data on the use of the active vitamin D sterols, such as calcitriol, alfacalcidol, and doxercalciferol, in the management of secondary hyperparathyroidism in patients with mild-to-moderate renal insufficiency (stage 3 or 4 chronic kidney disease). Data on potential benefits, including improved histologic abnormalities of bone from secondary hyperparathyroidism, increased bone mineral density, and a reduction of elevated parathyroid hormone levels, are documented. Consideration is given to the risks of such therapy, which includ

Mass Spectral Analysis of Sterols and Other Steroids in Different Ionization Modes: Sensitivity and Oxidation Artifacts.

In the course of synthetic work and mass spectrometry (MS) with hydroxy steroids, we observed not only the loss of H2O but prominent 2 and 4 amu losses using atmospheric pressure chemical ionization (APCI), leading to confusion of the structural assignments. This loss of 2 amu, which we attributed mainly to oxidation of hydroxyls, varied among 44 steroids and sterols analyzed; 36 showed losses of 2n amu in APCI MS analysis (17/22 Δ5 steroids, 17/19 Δ4 steroids, and 2/3 estrogens). With the Δ5 steroids and sterols, the precursor MH+ was either observed as a minor ion or (more frequently) not de

Effect of vitamin D sterols on bone histology in pre-dialysis patients: A prospective controlled study.

BACKGROUND: Abnormalities related to mineral and bone metabolism are a common finding in chronic kidney disease (CKD). Vitamin D compounds are often prescribed to CKD patients with the purpose to control secondary hyperparathyroidism and reduce the risk of high-turnover bone disease. However, data on the effect of vitamin D sterols on bone histology in non-dialysis CKD is limited. MATERIALS AND METHODS: A prospective controlled study was conducted on a cohort of 56 patients with CKD stages 3 and 4. 19 patients on calcitriol and 12 patients on cholecalciferol were compared to a group of 25 age
