BACKGROUND: Despite the success of long-term antiretroviral therapy (ART), immune dysfunction-including incomplete T helper cell recovery, immune activation, and inflammation-persists and may affect the benefits of analytical ART treatment interruption (ATI) trials among people living with human immunodeficiency virus (HIV) (PLHIV) in sub-Saharan Africa (SSA). This narrative review summarizes evidence of incomplete immune function recovery among PLHIV on long-term ART and potential interventions to enhance their ability to participate in ATI trials with immunotherapies in the quest for an HIV

INTRODUCTION: Uridine diphosphate glucuronosyltransferase 1A1 (UGT1A1) is closely associated with the management of HIV and tuberculosis (TB) coinfection because it modulates the metabolism of antiretroviral (ARV) drugs. The frequency of UGT1A1 polymorphisms varies widely among sub-Saharan Africans. However, studies examining the frequency of UGT1A1 polymorphisms and their impact on drug response profiles, accounting for environmental factors, drug-drug and gene-drug interactions and non-compliance remain sparse. Given that HIV and TB treatments often involve complex drug regimens with a high

BACKGROUND: Glaucoma is one of the leading causes of visual impairment and blindness globally, mainly affecting older adults. Disproportionately affecting people of African descent, its high prevalence, combined with the region's limited healthcare infrastructure and access to eye care services, make it a major public health challenge deserving attention in sub-Saharan Africa.
OBJECTIVE: This paper aims to synthesise current literature on glaucoma prevalence and management options among older adults in sub-Saharan Africa.
MATERIALS AND METHODS: A systematic search was conducted using databas

Type 2 diabetes is rising globally, with sub-Saharan Africa facing the steepest projected increase. In low-resource settings, limited screening contributes to high rates of undiagnosed disease, delaying intervention. Traditional risk models often rely on single-factor thresholds or population averages, which can miss combinations of demographic, anthropometric, and lifestyle factors relevant in African contexts. However, emerging evidence suggests that interactions between multiple risk factors provide more accurate, region-specific characterizations of risk. We applied a multidimensional subg

BACKGROUND: The extant literature identifies low breast cancer literacy as a major reason for the low uptake of breast cancer screening among women in sub-Saharan Africa (SSA). Recognizing the potential of health education interventions to improve breast cancer literacy, researchers in SSA have implemented varied interventions. However, evidence on the effectiveness of these interventions in SSA is limited and fragmented, with variations in educational strategies, delivery methods, and cultural contexts. A systematic review is needed to synthesize existing evidence, assess the effectiveness of

Schistosomiasis is a neglected tropical disease, affecting 253 million people worldwide. This study aimed to describe the clinical and epidemiological characteristics of schistosomiasis in sub-Saharan African migrants evaluated through a general screening program for imported infectious diseases at a specialized international health unit in Barcelona, Spain. We performed a retrospective observational study (2014-2023) including confirmed (defined by egg detection in stool or urine) and probable cases (identified by positive serology against Schistosoma). Data on epidemiology, clinical presenta

BACKGROUND: Sub-Saharan Africa (SSA) continues to be the hub of the global HIV/AIDS pandemic, globally. Despite several attempts to meet their HIV prevention needs, men continue to have high rates of HIV infection. Pre-exposure prophylaxis (PrEP) is an effective treatment that, when taken as prescribed, can be very efficient in preventing and reducing the risk of HIV acquisition. HIV self-testing (HIVST) has been demonstrated to be acceptable among men in SSA. As such, this review aims to explore the existing literature on the integration of PrEP dispensing with HIVST among men in SSA.

Tuberculosis (TB) is a major health burden in Africa. Although TB is treatable, anti-TB drugs are associated with adverse drug reactions (ADRs), which are partly attributed to pharmacogenetic variation. The distribution of star alleles (haplotypes) influencing anti-TB drug metabolism is unknown in many African populations. This presents challenges in implementing genotype-guided therapy in Africa to decrease the occurrence of ADRs and enhance the efficacy of anti-TB drugs. In this study, we used StellarPGx to call variants and star alleles in NAT1, NAT2, GSTM1, GSTT1, GSTP1, and CYP2E1, from 1

This study explores, via a prospective cross-sectional online survey, sunscreen-use patterns and perceptions among digitally active people from sub-Saharan Africa. The survey reveals that 64% of respondents use sunscreen, primarily to prevent photoageing and hyperpigmentation, with broad-spectrum SPF >30 products the most commonly used. Fifty-six percent report reduced hyperpigmentation and sunburn with use. This study reports increased sunscreen use and knowledge of perceived benefits compared with previous African studies; however, knowledge gaps and cosmetic concerns (whitish casts and oili

BACKGROUND: Africans are underrepresented in Huntington's disease (HD) research. A European ancestor was postulated to have introduced the mutant Huntingtin (mHtt) gene to the continent; however, recent work has shown the existence of a unique Htt haplotype in South-Africa specific to indigenous Africans.
OBJECTIVE: We aimed to investigate the CAG trinucleotide repeats expansion in the Htt gene in a geographically diverse cohort of patients with chorea and unaffected controls from sub-Saharan Africa.
METHODS: We evaluated 99 participants: 43 patients with chorea, 21 asymptomatic first-degree

INTRODUCTION: The COVID-19 pandemic has posed unprecedented global public health challenges since its emergence in late 2019. Vaccination remains the most effective intervention for preventing severe disease and reducing transmission. However, disparities in vaccine distribution and access have been widely reported, particularly in low-income and middle-income countries and rural communities. Rural populations in sub-Saharan Africa (SSA) often face structural barriers, including weak health systems, limited infrastructure and an inadequate healthcare workforce, which may affect the availabilit

Oral pre-exposure prophylaxis (PrEP) is a valuable tool to help end HIV, but persistent PrEP use is low amongst adolescent girls and young women (AGYW) in sub-Saharan Africa (SSA), who are at high risk for HIV. To improve persistent PrEP use in AGYW, more research is needed to understand what drives their decisions to continue with PrEP. We conducted a thematic, comparative qualitative analysis of in-depth interviews with 32 AGYW who participated in PEPFAR's DREAMS program in western Kenya: 16 were randomly sampled among those who persisted with PrEP and 16 among those who discontinued. We com

INTRODUCTION: Previous Western studies have shown an association between neurodegenerative plasma biomarkers and geriatric depression. However, few studies have examined this relationship in sub-Saharan African countries. This article examines this association in older adults with or without clinical dementia in Kinshasa, Democratic Republic of Congo.
METHODS: This cross-sectional study included eighty-one people aged 65 years and older assessed for dementia using the Community Dementia Screening Instrument (CSID) and the Alzheimer's Disease Questionnaire. Depression and cognitive impairment

INTRODUCTION: Food hygiene practices are crucial to avoid foodborne illness and improve human well-being. Millions of people get sick, and many of them pass away due to eating unhealthy food. Foodborne diseases are still a public health problem in developing countries.
OBJECTIVE: This study aimed to determine the prevalence and factors associated with food hygiene practices among food handlers in sub-Saharan Africa.
METHODS: An extensive search was conducted using various databases including PubMed, Science Direct, African Journal Online, and Google Scholar. The search results were then extr

Human African trypanosomiasis (HAT), also known as sleeping sickness, is one of the Africa's 'neglected diseases' and is caused by infection with protozoan parasites of the Trypanosoma genus. Transmitted by the bite of the tsetse fly, it puts 70 million people at risk throughout sub-Saharan Africa and is usually fatal if untreated or inadequately treated. In this brief overview, some important recent developments in this disease are outlined. These cover various aspects including a reduction in disease incidence, newly recognised parasite reservoir sites in humans, disease outcome, novel diagn

BACKGROUND: Malaria control in African cities faces challenges mainly due to unplanned urbanization and the spread of Anopheles stephensi. Urbanization is changing malaria dynamics, driven by environmental changes and population growth, with nearly 70% of people projected to live in urban areas by 2050. This scoping review maps the epidemiology of urban malaria in sub-Saharan Africa, identifying research gaps and guiding strategies for control and elimination.
METHODS: A structured search across multiple databases was performed using predefined eligibility criteria to select articles. Accordi

Human African trypanosomiasis, caused by the kinetoplastid parasite Trypanosoma brucei, affects thousands of people across sub-Saharan Africa, and is fatal if left untreated. Treatment options for this disease, particularly stage 2 disease, which occurs after parasites have infected brain tissue, are limited due to inadequate efficacy, toxicity and the complexity of treatment regimens. We have discovered and optimized a series of benzoxaborole-6-carboxamides to provide trypanocidal compounds that are orally active in murine models of human African trypanosomiasis. A key feature of this series

BACKGROUND: Climate change is reshaping the populations health systems serve, yet health information systems (HIS) remain blind to one of the fastest-growing groups: people displaced or migrating in the context of climate change. In sub-Saharan Africa this mobility is now counted at scale, but mobility data and health data systems have developed as parallel infrastructures blind to each other's populations.
OBJECTIVE: To examine the health of populations moving in the context of climate change in sub-Saharan Africa, and to argue that a principal obstacle to climate-resilient health policy is
