Expression of MUM1/IRF4 selectively clusters with primary effusion lymphoma among lymphomatous effusions: implications for disease histogenesis and pathogenesis.

Primary effusion lymphoma (PEL) is a peculiar B-cell lymphoma characterized by infection by human herpesvirus type-8/Kaposi sarcoma-associated herpesvirus (HHV-8/KSHV) and by preferential growth in the serous body cavities. Histogenetic studies have suggested that PEL originates from B cells at a late stage of differentiation. In this study, we have investigated PEL for the expression status of MUM1/IRF4 (multiple myeloma 1/interferon regulatory factor 4) protein, which is involved in physiological B-cell maturation and represents a histogenetic marker of late B-cell differentiation. Using mul

Circulating plasma cells in multiple myeloma: characterization and correlation with disease stage.

The aim of this study was to develop a flow cytometric test to quantitate low levels of circulating myeloma plasma cells, and to determine the relationship of these cells with disease stage. Cells were characterized using five-parameter flow cytometric analysis with a panel of antibodies, and results were evaluated by comparison with fluorescent consensus-primer IgH-PCR. Bone marrow myeloma plasma cells, defined by high CD38 and Syndecan-1 expression, did not express CD10, 23, 30, 34 or 45RO, and demonstrated weak expression of CD37 and CD45. 65% of patients had CD19- 56+ plasma cells, 30% CD1

IgA nephropathy-associated breast milk B cell alterations.

BACKGROUND: IgA nephropathy (IgAN) is a mucosal immune-associated disease characterized by the overproduction of galactose-deficient IgA1 (Gd-IgA1) and formation of nephritogenic immune complexes. Epstein-Barr virus (EBV), which preferentially infects IgA+ B cells, has been implicated in IgAN pathogenesis, although its role remains unclear. Given mucosa involvement, we characterized breast milk B cells as available representatives of mucosal tissue to better understand the pathogenesis of IgAN. METHODS: We performed a phenotypic analysis of B cells (CD19+), plasmablasts (CD38+), and CD138+ ce

Spotlight on the Transglutaminase 2-Heparan Sulfate Interaction.

Heparan sulfate proteoglycans (HSPGs), syndecan-4 (Sdc4) especially, have been suggested as potential partners of transglutaminase-2 (TG2) in kidney and cardiac fibrosis, metastatic cancer, neurodegeneration and coeliac disease. The proposed role for HSPGs in the trafficking of TG2 at the cell surface and in the extracellular matrix (ECM) has been linked to the fibrogenic action of TG2 in experimental models of kidney fibrosis. As the TG2-HSPG interaction is largely mediated by the heparan sulfate (HS) chains of proteoglycans, in the past few years a number of studies have investigated the aff

LTBP-2 has multiple heparin/heparan sulfate binding sites.

Latent transforming growth factor-beta-1 binding protein-2 (LTBP-2) is a protein of poorly understood function associated with fibrillin-1-containing microfibrils during elastinogenesis. In this study we investigated the molecular interactions of LTBP-2 with heparin and heparan sulfate proteoglycans (HSPGs) since unidentified cell surface HSPGs are critical for normal fiber assembly. In solid phase assays, heparin conjugated to albumin (HAC) bound strongly to recombinant full-length human LTBP-2. This interaction was completely blocked by addition of excess heparin, but not chondroitin sulfate

Heparan sulfate proteoglycan specificity during axon pathway formation in the Drosophila embryo.

Axon guidance is influenced by the presence of heparan sulfate (HS) proteoglycans (HSPGs) on the surface of axons and growth cones (Hu, [2001]: Nat Neurosci 4:695-701; Irie et al. [2002]: Development 129:61-70; Inatani et al. [2003]: Science 302:1044-1046; Johnson et al. [2004]: Curr Biol 14:499-504; Steigemann et al. [2004]: Curr Biol 14:225-230). Multiple HSPGs, including Syndecans, Glypicans and Perlecans, carry the same carbohydrate polymer backbones, raising the question of how these molecules display functional specificity during nervous system development. Here we use the Drosophila cen

THBS1+ Macrophages Exacerbate Modic Changes via SDC4-Dependent Activation of NLRP3 Inflammasome.

Modic changes (MCs) in the lumbar vertebral endplates are a common imaging finding in patients with low back pain. Their pathogenesis and progression are closely linked to the local immune microenvironment, underscoring the need to investigate the immunomodulatory mechanisms involved in MCs. Here, we identified the THBS1+ macrophages displaying a strongly pro-inflammatory phenotype. The THBS1+ macrophages were enriched at lesion sites in both human lumbar MCs specimens and the C. acnes-induced mouse model of MC. In vitro, C. acnes and its key metabolites promoted THBS1 expression in macrophage

Multifunctionalization of Cells with a Self-Assembling Molecule to Enhance Cell Engraftment.

Cell-based therapy is a promising approach to restoring lost functions to compromised organs. However, the issue of inefficient cell engraftment remains to be resolved. Herein, we take a chemical approach to facilitate cell engraftment by using self-assembling molecules which modify two cellular traits: cell survival and invasiveness. In this system, the self-assembling molecule induces syndecan-4 clusters on the cellular surface, leading to enhanced cell viability. Further integration with Halo-tag technology provided this self-assembly structure with matrix metalloproteinase-2 to functionali

A low androgenic state inhibits erectile function by suppressing endothelial glycosides in the penile cavernous tissue of rats.

BACKGROUND: The endothelial glycocalyx is an important barrier that protects the structure and function of endothelial cells. Androgen deficiency is a common factor that causes structural and functional impairment of endothelial cells. AIM: To investigate changes in the endothelial glycocalyx in the penile corpus cavernosum of the rat with low androgen status and its relationship with erection function. METHODS: Eighteen 10-week-old Sprague-Dawley male rats were randomly divided into 3 groups (n = 6 each): sham operation, castration, and castration + testosterone replacement. The maximum int

A Synthetic Lethal Screen Identifies a Role for Lin-44/Wnt in C. elegans Embryogenesis.

BACKGROUND: The C. elegans proteins PTP-3/LAR-RPTP and SDN-1/Syndecan are conserved cell adhesion molecules. Loss-of-function (LOF) mutations in either ptp-3 or sdn-1 result in low penetrance embryonic developmental defects. Work from other systems has shown that syndecans can function as ligands for LAR receptors in vivo. We used double mutant analysis to test whether ptp-3 and sdn-1 function in a linear genetic pathway during C. elegans embryogenesis. RESULTS: We found animals with LOF in both sdn-1 and ptp-3 exhibited a highly penetrant synthetic lethality (SynLet), with only a small perce

Molecular histogenesis of plasmablastic lymphoma of the oral cavity.

PURPOSE: Transurethral lithotripsy (TUL) is a major modality for the endoscopic management of ureteral stones. Ureteral spasm makes access for ureters difficult, which causes impaction of the ureteroscope, ureteral dislodge, and a low success rate of endoscopic surgeries. This study described the outcomes of a new endoscopic surgical experience by use of 40-degree warm saline irrigation during TUL compaired with routine ambient air irrigation in TUL. MATERIALS AND METHODS: In this randomized clinical trial from 2014 to 2015, 150 patients with ureteral stone with balanced randomization were di

Extracellular matrix alterations after blood instillation in tunica albuginea of rats.

The cause of Peyronie's disease (PD) is still not completely understood. The objective of this study, therefore, was to analyze the histological and biochemical alterations that occur after the instillation of blood in the tunica albuginea (TA) of rats with an emphasis on the remodeling process of ECM. Thirty male Wistar rats were divided into 4 groups: two control groups with instillation of distilled water in TA followed by penectomy after 15 days or 45 days, respectively and two experimental groups with instillation of blood in TA followed by penectomy after 15 days or 45 days, respectively

SDCBP competes with CBL to stabilize PES1 and promote c-Myc translation in ESCC tumorigenesis.

Syndecan-binding protein (SDCBP) has been identified as a potential prognostic biomarker in esophageal squamous cell carcinoma (ESCC) in our previous study. However, its specific interacting partners and oncogenic mechanisms remain incompletely understood. Here, we identify PES1 as a direct binding partner of SDCBP and demonstrate that SDCBP stabilizes PES1 by competitively disrupting its interaction with the E3 ubiquitin ligase CBL, thereby inhibiting PES1 ubiquitination and degradation. Stabilized PES1 enhances pre-rRNA processing and promotes c-Myc protein synthesis. In a 4-nitroquinoline 1

Morphometrical and immunopathological dissection of the hepatic Schistosoma haematobium granuloma in the murine host.

This paper presents the results of a study aimed at identifying the immunopathological and morphometrical characteristics of liver granulomas induced by the parasite Schistosoma haematobium. The OF1 mouse is a susceptible host for experimental infection with the Egyptian NAMRU-3 strain of S. haematobium. Formation of liver granulomas started after 13-14 weeks of infection. These hepatic granulomas reached a maximum volume after 20 weeks of infection (MGV: 1.82 +/- 0.15 x 10(7) micron 3) and regressed thereafter (1.55 +/- 0.16 x 10(7) micron 3 after 24 weeks).
