The aim of this study was to develop an efficient exogenous gene delivery system using cultured marrow cells and porous hydroxyapatite ceramics. Bone marrow cells were obtained from the femoral shaft of a Fischer 344 rat and cultured in a medium containing 15% fetal bovine serum until confluent. After trypsinization, cells were subcultured at a cell density of 1 x 10(4) cells/cm2 in the presence of fetal bovine serum. The subcultured bone marrow cells were infected with recombinant retroviruses carrying the lacZ gene. The retrovirus infection was performed seven times from day 1 to day 7 durin

The Spiegelberg brain pressure catheter is a low cost implantable intracranial pressure measuring system which has the unique ability to perform regular automatic zeroing. A new version of the catheter has become available with a subdural bolt fixation to allow insertion of the device into the brain parenchyma. The accuracy of this system has been evaluated in comparison with a ventricular fluid pressure method in a series of patients to determine its accuracy and utility in the clinical environment. Hourly readings from the Spiegelberg system have been compared with those obtained using a sta

The influence of transducer position and angulation upon the mitral systolic echo was studied in 100 presumably healthy females. Echocardiographic studies were performed from the second, third, fourth and fifth intercostal spaces (ICS). The role of the sound beam's path relative to cardiac motion was assessed by analyzing the recorded mitral valve pattern as a function of transducer orientation, independent of the absolute ICS used. With the transducer directed caudally when both mitral leaflets and left atrium were recorded, holosystolic or midsystolic posterior motion of the mitral valve lea

Eighty patients with various forms of heart disease were studied with the use of a newly developed ultrasonic system having 20 transducers arranged in a linear array. This system allows visualization of the heart in two dimensions in real time. All 15 patients with the mitral valve prolapse syndrome, 13 patients with mitral stenosis, five patients with pericardial effusion, four patients with atrial septal defect, and one patient with left ventricular dyssynergy were properly recognized with this system. One of five patients with hypertrophic myopathy and one of four patients with congestive m

Immunoglobulin E (IgE) is a critical biomarker for the diagnosis and therapeutic monitoring of allergic diseases. Conventional IgE detection methods (e.g., ELISA, ImmunoCAP) generally offer satisfactory sensitivity but require hours to days, rely on specialized equipment and trained personnel, and are unsuitable for point-of-care or home-based testing. Here, we developed a portable point-of-care testing platform termed Aptamer-CRISPR Glucose Transducer (ACGT), based on aptamer competition and CRISPR-Cas12a trans-cleavage. The platform employs the D17.4 aptamer as the molecular recognition elem

Cavitation driven by low-frequency ultrasound has been widely recognized as an effective method of transdermal drug delivery, but challenges remain regarding the size and efficiency of the sonicator. The cymbal transducer is a promising candidate for such applications; however, the conventional design is limited by its relatively low power output and difficulty in manufacturing with consistency, limiting its success in arrays and for extended treatment times. This study introduces a modified cymbal transducer, operating at 66.2 kHz, designed to offer higher output pressure and an easier fabric

The pituitary adenylate cyclase-activating polypeptide receptor (PAC1R) is a class B G protein-coupled receptor (GPCR) that engages both G proteins and β-arrestins to mediate diverse signaling responses, yet how PAC1R adopts distinct intracellular conformations to achieve this transducer selectivity remains poorly understood. Here, we report the cryo-electron microscopy structure of PAC1R in complex with β-arrestin 1 (βarr1), revealing a core-engaged conformation. Comparison with the Gs-bound PAC1R structure shows that βarr1 engagement is associated with remodeling of the intracellular transme

Safety of hearing devices attached to the tympanic membrane depends on applied forces and resulting movements of the ossicles when pulling off the device. A robotized temporal bone measurement setup was designed to apply and measure defined pull-off forces and as well as the corresponding displacements of the round-window membrane via Laser-Doppler-vibrometry. Here, we investigate in five temporal bones the pull-off dynamics of the Vibrosonic Hearing Contact Lens® the silicone membrane of which being formed to match the tympanic-membrane shape of individual temporal bones. Acute respiratory distress syndrome (ARDS) is marked by damage to the capillary endothelium and alveolar epithelium following edema formation and cell infiltration. Currently, there are no effective treatments for severe ARDS. Pathologies such as sepsis, pneumonia, fat embolism, and severe trauma may cause ARDS with respiratory failure. The primary mechanism of edema clearance is the epithelial cells' Na/K-ATPase (NKA) activity. NKA is an enzyme that maintains the electrochemical gradient and cell homeostasis by transporting Na+ and K+ ions across the cell membrane. Direct injury on alveolar c

Chronic inflammation is a hallmark of atherosclerosis and results from an imbalance between proinflammatory and proresolving signaling. The human GPR32 receptor, together with the ALX/FPR2 receptor, transduces biological actions of several proresolving mediators that stimulate resolution of inflammation. However, since no murine homologs of the human GPR32 receptor exist, comprehensive in vivo studies are lacking. Using human atherosclerotic lesions from carotid endarterectomies and creating a transgenic mouse model expressing human GPR32 on a Fpr2×ApoE double-KO background (hGPR32myc×Fpr2-/-×

Infusion of ex vivo transduced haematopoietic stem cells (HSC) has emerged as a promising new treatment of certain monogenetic disorders. Since early clinical studies on patients with severe combined immune deficiency were halted due to de novo leukaemia, the technology has matured. Thus, treatment of transfusion-dependent thalassaemia and adenosine deaminase deficient severe combined immunodeficiency by using lentiviral vectors for gene correction of autologous HSC can induce expression of the deficient protein and thus potentially cure the patients. The review summarises recent advances allo

INTRODUCTION: Ultrasound has been increasingly utilized for the identification of endotracheal tube (ETT) location after an intubation attempt, particularly among patients in cardiac arrest. However, prior studies have varied with respect to the choice of transducer and no studies have directly compared the accuracy between transducer types. Our study is the first to directly compare the accuracy of ETT confirmation between the linear and curvilinear transducer.
METHODS: This study was performed in a cadaver lab using three different cadavers chosen to represent varying neck circumferences. C

How cells sense their mechanical environment and transduce forces into biochemical signals is a crucial yet unresolved question in mechanobiology. Platelets use receptor glycoprotein Ib (GPIb), specifically its α subunit (GPIbα), to signal as they tether and translocate on von Willebrand factor (VWF) of injured arterial surfaces against blood flow. Force elicits catch bonds to slow VWF-GPIbα dissociation and unfolds the GPIbα leucine-rich repeat domain (LRRD) and juxtamembrane mechanosensitive domain (MSD). How these mechanical processes trigger biochemical signals remains unknown. Here we ana

HYPOTHESIS: Changes to the angular position of the vibrating floating mass transducer (FMT) coupled to the long process of the incus will not affect stapes velocity.
OBJECTIVE: The MED-EL Vibrant Soundbridge is an active middle ear implantable device, which constitutes an effective alternative to acoustic hearing aids for the rehabilitation of patients with sensorineural and mixed hearing loss. Because of varied anatomy, it is not always possible to position the FMT in line with the vibrating axis of the stapes. Changes in stapes velocity after angulation of the FMT are measured using laser D

The surgical rehabilitation of mixed hearing losses can be performed by coupling the floating mass transducer of the Vibrant Soundbridge to the round window. The quality of coupling the floating mass transducer to the round window is crucial for the audiological outcome. It was the aim of this study to further observe the different patterns of floating mass transducer position at the round window. We compared twenty patients with mixed hearing loss implanted with a floating mass transducer attached to the round window and 24 surgeries between 5/2007 and 6/2010. An evaluation of the chronologic

AIM: To discuss existing disparity of practice and clinical implications of measuring cerebral perfusion pressure (CPP) from differing reference points thus highlighting the need for standardized benchmarks.
BACKGROUND: When managing traumatic brain injury (TBI), the arterial transducer level is a key to an accurate CPP reading; however, there is a lack of national standards about where to zero arterial transducers when monitoring CPP.
METHODS: A systematized search using the Cochrane library database, Pubmed database, Medline, British Library on line, CINAHL and PROQUEST using key search te

The pericellular matrix (PCM) is a narrow tissue region surrounding chondrocytes in articular cartilage, which together with the enclosed cell(s) has been termed the "chondron." While the function of this region is not fully understood, it is hypothesized to have important biological and biomechanical functions. In this article, we review a number of studies that have investigated the structure, composition, mechanical properties, and biomechanical role of the chondrocyte PCM. This region has been shown to be rich in proteoglycans (e.g., aggrecan, hyaluronan, and decorin), collagen (types II,

We have selectively targeted monocyte/macrophages overexpressing an immunomodulatory molecule, latency-associated peptide (LAP), a naturally occurring antagonist for transforming growth factor-beta1, to murine skin utilizing UV light to produce a cutaneous influx of transduced monocyte/macrophages. Bone marrow (BM) cells from BALB/c mice were transduced in vitro with a retroviral construct containing green fluorescent protein (GFP) for detection and human LAP (hLAP) as a test molecule. The transduced BM cells were then cultured in vitro with granulocyte-macrophage colony-stimulating factor (GM

As a result of underlying pathological diseases, such as osteoporosis, osteopenia, or due to altered loading after joint replacements, bones become more susceptible to microdamage accumulation than those of normal human beings, as are those of athletes who undertake strenuous exercise [Stromsoe, 2004. Fracture fixation problems in osteoporosis. Injury 35, 107-113]. Experimental evidence has linked bone adaptation to microdamage, and to increased cell activity. In this work, we investigated whether microcrack detection is related to rupturing of the cellular material itself due to crack face di

Expression of the adipocyte-derived protein resistin, which is thought to play a key role in the development of insulin resistance in vivo, is regulated by a variety of hormones and mediators, including insulin and TNFalpha. Here we describe our use of adenovirus-mediated gene transfer to determine which transcription factors and signaling pathways affect resistin expression in 3T3-L1 adipocytes. We found that resistin expression was enhanced by overexpression of C/EBPalpha and suppressed by C/EBPzeta, a negative regulator of C/EBPalpha. Additionally, C/EBPalpha induced resistin even in L6 myo
