Immunogenicity of plant-produced African horse sickness virus-like particles: implications for a novel vaccine.

African horse sickness (AHS) is a debilitating and often fatal viral disease affecting horses in much of Africa, caused by the dsRNA orbivirus African horse sickness virus (AHSV). Vaccination remains the single most effective weapon in combatting AHS, as there is no treatment for the disease apart from good animal husbandry. However, the only commercially available vaccine is a live-attenuated version of the virus (LAV). The threat of outbreaks of the disease outside its endemic region and the fact that the LAV is not licensed for use elsewhere in the world, have spurred attempts to develop an

Immunogenic virus-like particles continuously expressed in mammalian cells as a veterinary rabies vaccine candidate.

Rabies is one of the most lethal infectious diseases in the world, with a mortality approaching 100%. There are between 60,000 and 70,000 reported annual deaths, but this is probably an underestimation. Despite the fact that there are vaccines available for rabies, there is a real need of developing more efficacious and cheaper vaccines. This is particularly true for veterinary vaccines because dogs are still the main vector for rabies transmission to human beings. In a previous work, we described the development and characterization of rabies virus-like particles (RV-VLPs) expressed in HEK293

Prevention of cervical cancer: journey to develop the first human papillomavirus virus-like particle vaccine and the next generation vaccine.

In 2006, the first human papillomavirus (HPV) virus-like particle (VLP) vaccine was licensed. Gardasil(®), the quadrivalent HPV 6, 11, 16 and 18 recombinant VLP vaccine (4vHPV), developed by Merck demonstrated remarkable efficacy in prevention of important clinical pre-cursors to cervical cancer and genital warts. The vaccine was designed to protect against HPV 16 and 18 that cause ∼70% of cervical cancers and HPV 6 and 11 that cause ∼90% of genital warts. Initially, Gardasil(®) was indicated in the United States for women 9-26 years of age for the prevention of HPV 16 and 18-related cervical,

Detection of a New Recombinant Rabbit Hemorrhagic Disease Virus 2 in China and Development of Virus-like Particle-Based Vaccine.

Rabbit hemorrhagic disease virus (RHDV) is a very virulent virus of the genus Lagovirus causing severe and fatal hepatitis in the European rabbit (Oryctolagus cuniculus). RHDV has two distinct genotypes: GI.1 (RHDV) and GI.2 (RHDV2). The first RHDV2/GI.2 strain was identified as a recombinant virus between a non-pathogenic (GI.3P) and a pathogenic (GI.2) lagovirus, and the recombination is thought to have been a key mechanism in the emergence and evolution of RHDV2. Here, a new variant of RHDV2 was identified affecting domestic rabbits on Chinese farms, with a mortality rate of 70-80%. Phyloge

Balanced T and B cell responses are required for immune protection against Powassan virus in virus-like particle vaccination.

Powassan virus (POWV) is a tick-borne pathogen for which humans are an incidental host. POWV infection can be fatal or result in long-term neurological sequelae; however, there are no approved vaccinations for POWV. Integral to efficacious vaccine development is the identification of correlates of protection, which we accomplished in this study by utilizing a murine model of POWV infection. Using POWV lethal and sub-lethal challenge models, we show that (1) robust B and T cell responses are necessary for immune protection, (2) POWV lethality can be attributed to both viral- and host-mediated d

Virus Like Particles (VLP) as multivalent vaccine candidate against Chikungunya, Japanese Encephalitis, Yellow Fever and Zika Virus.

Mosquito borne viral diseases are an emerging threat as evident from the recent outbreak of Zika virus (ZIKV) as well as repeated outbreaks of Chikungunya (CHIKV), Yellow fever (YFV) and Japanese encephalitis (JEV) virus in different geographical regions. These four arboviruses are endemic in overlapping regions due to the co-prevalence of the transmitting mosquito vector species Aedes and Culex. Thus, a multivalent vaccine that targets all four viruses would be of benefit to regions of the world where these diseases are endemic. We developed a potential Virus Like Particle (VLP) based multiva

Vaccine Efficacy Induced by 2009 Pandemic H1N1 Virus-Like Particles Differs from that Induced by Split Influenza Virus.

Influenza virus-like particles (VLPs) vaccines are highly immunogenic, showing strong protective efficacy against homologous virus infection compared to split vaccine. However, a comparative efficacy study against heterosubtypic virus infection between VLPs and split vaccine has yet to been reported. In this study, we generated VLPs vaccine containing hemagglutinin (HA) and matrix protein (M1) of the 2009 pandemic H1N1, and investigated the protective efficacies induced by VLPs vaccine and commercial monovalent H1N1 pandemic split vaccine from Sanofi-Pasteur. Mice were intramuscularly immunize

Antibody Persistence in Adults Two Years after Vaccination with an H1N1 2009 Pandemic Influenza Virus-Like Particle Vaccine.

The influenza virus is a human pathogen that causes epidemics every year, as well as potential pandemic outbreaks, as occurred in 2009. Vaccination has proven to be sufficient in the prevention and containment of viral spreading. In addition to the current egg-based vaccines, new and promising vaccine platforms, such as cell culture-derived vaccines that include virus-like particles (VLPs), have been developed. VLPs have been shown to be both safe and immunogenic against influenza infections. Although antibody persistence has been studied in traditional egg-based influenza vaccines, studies on

Virus-like particle vaccine with authentic quaternary epitopes protects against Zika virus-induced viremia and testicular damage.

Zika virus (ZIKV) caused unprecedented outbreaks in South America and the Caribbean in 2015-2016, leading primarily to a series of abnormalities in neonates termed congenital Zika syndrome. The threat of ZIKV reemergence has seen the development of multiple ZIKV vaccines that are at the preclinical stage or in early-stage clinical trials. Herein, we describe a pathway to the development of ZIKV vaccines generated using a baculovirus-insect cell expression system, which is widely applied for the manufacture of biologics for human use. A virus-like-particle-producing DNA vaccine triggers specific humoral immunity against hepatitis C virus in mice.

Worldwide, 50 million people are chronically infected with hepatitis C virus (HCV), with 1 million new infections annually. The limitations of direct-acting antiviral drugs, including high costs, limited access, reinfections, and emergence of drug resistance, have highlighted the urgent need for a prophylactic vaccine to achieve global elimination. However, the development of an HCV vaccine remains challenging due to the genetic diversity of the virus, evasion mechanisms, and our limited understanding of protective immunity. The use of DNA-based vaccines is a promising strategy for HCV vaccine

A modular plug-and-play vaccine construction platform using bio-click chemistry and virus-like particles.

Virus-like particles (VLPs) are a convenient platform for the presentation of antigens to enhance their immunogenic properties. In the present study, we combined sortase-based labelling of the outer surface protein C (OspC) of Borrelia burgdorferi (as an antigen) with click chemistry to couple it to functionalised VLPs of the hepatitis B core antigen (HBc) expressed in Escherichia coli. The first step involved p-azido-phenylalanine (pAzF) being bioorthogonally incorporated into HBc. The major immunodominant region (MIR) was chosen as the incorporation site because HBc is known to form dimers t

Foot-and-mouth disease virus-like particle mRNA vaccine induces strong humoral and cellular immunity.

Foot-and-mouth disease (FMD) is a major animal infectious disease that has garnered significant international attention. Currently, conventional inactivated vaccines and virus-like particle (VLP) vaccines primarily induce protective immune responses through the generation of neutralizing antibodies. However, these vaccines exhibit limited activation of cellular immunity and offer short-term immune persistence. Furthermore, inactivated vaccines raise certain biosafety concerns. In this study, an FMD mRNA vaccine capable of self-assembling into non-infectious VLP was developed. Immunization of m

Development and Evaluation of a Multi-Epitope Vaccine Based on P22 Virus-Like Particles Targeting Helicobacter pylori.

BACKGROUND: The current first-line treatment for Helicobacter pylori (H. pylori) infection is bismuth-based quadruple therapy. However, the widespread use of antibiotics has contributed to the emergence of antibiotic-resistant strains, thereby increasing the likelihood of treatment failure. This study aimed to develop a multi-epitope H. pylori vaccine, designated P22-C3, based on P22 Virus-like Particles, with the objective of reducing infection rates and preventing transmission. MATERIALS AND METHODS: The structural characteristics of P22-C3 were analyzed using transmission electron microsco

Pre-clinical safety studies of intranasal virus-like particles based vaccine for prevention of COVID-19.

INTRODUCTION: The large-scale and prolonged pandemic of the novel coronavirus disease (COVID-19) has demonstrated the need for effective vaccination. Along with immunogenicity, safety is a critical issue for vaccines, as public trust can contribute to the success or failure of immunization programs. In preclinical studies, we assessed the safety of an intranasal Virus-like particle (VLP)-based vaccine in mice and rats. The aim of the study is to conduct preclinical acute and subchronic toxicity studies assessing local tolerability of an intranasal VLP vaccine against COVID-19 in accordance wit

Safety and Efficacy of Plant-Produced Trivalent Virus-Like Particle Vaccine Candidate Against Bluetongue Disease.

Bluetongue (BT) is a non-contagious, insect-transmitted disease of wild and domestic ruminants caused by bluetongue virus (BTV). Effective control of BT disease relies on vaccination against prevalent or seasonal serotypes using live attenuated or inactivated vaccines. Limitations of these vaccines offer opportunities for improvement. Transient protein production in plants has evolved as a platform that offers a unique ability to express multiple antigens and complex protein assemblies. In this study, a trivalent virus-like particle (VLP) vaccine candidate against BT was produced in Nicotiana

ESCRT recruitment to SARS-CoV-2 spike induces virus-like particles that improve mRNA vaccines.

Prime-boost regimens for COVID-19 vaccines elicit poor antibody responses against Omicron-based variants and employ frequent boosters to maintain antibody levels. We present a natural infection-mimicking technology that combines features of mRNA- and protein nanoparticle-based vaccines through encoding self-assembling enveloped virus-like particles (eVLPs). eVLP assembly is achieved by inserting an ESCRT- and ALIX-binding region (EABR) into the SARS-CoV-2 spike cytoplasmic tail, which recruits ESCRT proteins to induce eVLP budding from cells. Purified spike-EABR eVLPs presented densely arrayed

Efficacy of a plant-produced infectious bronchitis virus-like particle vaccine in specific pathogen-free chickens.

Infectious bronchitis (IB) Gammacoronavirus causes a highly contagious respiratory disease in chickens that is listed by the World Organisation for Animal Health (WOAH). Its high mutation ability has resulted in numerous variants against which the commercially available live or recombinant vaccines singly offer limited protection. Agrobacterium-mediated transient expression in Nicotiana benthamiana (tobacco) plants was used here to produce a virus-like particle (VLP) vaccine expressing a modified full-length IBV spike (S) protein of a QX-like IB variant. In a challenge study with the homologou

Production of Multi-Subtype Influenza Virus-Like Particles by Molecular Fusion with BAFF or APRIL for Vaccine Development.

Virus-like particle (VLP) technology is an alternative platform for developing vaccines to combat seasonal and pandemic influenza. Influenza VLPs are non-infectious nanoparticles that can elicit effective vaccine immunogenicity in hosts. B-cell-activating factor (BAFF, or BLyS) and a proliferation-inducing ligand (APRIL) are members of the tumor necrosis factor (TNF) superfamily of cytokines. Both BAFF and APRIL are homotrimers that interact with homotrimeric receptors. Here, we report a method of the production of influenza VLPs by molecular incorporation with BAFF or APRIL homotrimers to int

Virus-like particle-based vaccine against coxsackievirus A6 protects mice against lethal infections.

Coxsackievirus A6 (CA6) is emerging as one of the major causative agents of hand, foot, and mouth disease (HFMD) worldwide. However, no vaccine is currently available for preventing CA6 infection. Here, we report the development of a virus-like particle (VLP)-based recombinant vaccine for CA6. We produced CA6 VLPs in insect cells by infecting the cells with a baculovirus coexpressing the genes encoding CA6 P1 and 3CD. Biochemical analyses showed that the produced VLPs consisted of VP0, VP1, and VP3 capsid subunit proteins generated by the cleavage of P1 by 3CD. Mice immunized with these VLPs p
