E3 ubiquitin ligase rififylin has yin and yang effects on rabbit cardiac transient outward potassium currents (Ito) and corresponding channel proteins.

Genome-wide association studies have reported a correlation between a SNP of the RING finger E3 ubiquitin protein ligase rififylin (RFFL) and QT interval variability in humans (Newton-Cheh et al., 2009). Previously, we have shown that RFFL downregulates expression and function of the human-like ether-a-go-go-related gene potassium channel and corresponding rapidly activating delayed rectifier potassium current (IKr) in adult rabbit ventricular cardiomyocytes. Here, we report that RFFL also affects the transient outward current (Ito), but in a peculiar way. RFFL overexpression in adult rabbit v

8-Chloro-cyclic AMP mediates growth inhibition of breast cancer cells via regulation of Yin-Yang 1 and estrogen receptor α levels.

As a derivative of cyclic AMP (cAMP), 8-Chloro-cyclic AMP (8-Cl-cAMP) is shown to induce growth inhibition, apoptosis, and differentiation in different cancer cells by activating AMP-activated protein kinase (AMPK) and p38 mitogen-activated protein kinase (p38 MAPK). The p38 MAPK is known as a downstream mediator of 8-Cl-cAMP and binds to a multifunctional transcription factor Yin-Yang 1 (YY1). Although cytotoxic effects of 8-Cl-cAMP on cancer cells have been studied over several decades, molecular details or interplays by which they allow selective cytotoxicity to certain cancer cells remain

The Role of MIF and IL-10 as Molecular Yin-Yang in the Modulation of the Host Immune Microenvironment During Infections: African Trypanosome Infections as a Paradigm.

African trypanosomes are extracellular flagellated unicellular protozoan parasites transmitted by tsetse flies and causing Sleeping Sickness disease in humans and Nagana disease in cattle and other livestock. These diseases are usually characterized by the development of a fatal chronic inflammatory disease if left untreated. During African trypanosome infection and many other infectious diseases, the immune response is mediating a see-saw balance between effective/protective immunity and excessive infection-induced inflammation that can cause collateral tissue damage. African trypanosomes are

Clonal expansion and functional exhaustion of monoclonal marginal zone B cells in mixed cryoglobulinemia: the yin and yang of HCV-driven lymphoproliferation and autoimmunity.

Monoclonal marginal zone (MZ) B cells expressing a V(H)1-69-encoded idiotype accumulate in HCV-associated mixed cryoglobulinemia (MC). These cells recognize the E2 protein of HCV and their massive clonal expansion reflects the propensity of MZ B cells to proliferate robustly upon antigenic stimulation by microorganisms, a property that makes them prone to neoplastic transformation. V(H)1-69(+) B cells of MC patients are phenotypically heterogeneous and resemble either mature MZ B cells (IgM(+)CD27(+)CD21(high)) or the unusual CD21(low) B cells that accumulate in other immunological disorders s

TACItly changing tunes: farewell to a yin and yang of BAFF receptor and TACI in humoral immunity? New genetic defects in common variable immunodeficiency.

PURPOSE OF REVIEW: The complex system of the tumour necrosis factor ligands BAFF and APRIL and their three receptors BCMA, TACI and BAFF receptor and its role in B-cell development and function is the objective of extensive research. Whereas the importance of BAFF/BAFF receptor interactions for B-cell survival could be clearly demonstrated, TACI is believed to counteract BAFF activity as a negative regulator in the murine model. The primarily immunodeficient phenotype of human TACI deficiency, however, claims a distinct function of this receptor in human peripheral B-cell development, class sw

Integrating "Yang transforming Qi and Yin constituting the body" with immune regulation: an evidence synthesis of multidimensional traditional chinese medicine therapy for immune thrombocytopenia.

Immune thrombocytopenia (ITP) is an acquired autoimmune hemorrhagic disorder with a substantial incidence globally across all age groups. Its pathogenesis involves the accelerated immune-mediated platelet destruction and impaired platelet production due to dysfunctional megakaryocyte maturation interactively. ITP is primarily treated by glucocorticoids and intravenous immunoglobulin in Western medicine conventionally. However, these therapies exhibit several limitations such as corticosteroid dependency, increased risk of infection, treatment resistance, and frequent relapse, despite its obvio

The yin and yang interplay between HDAC6 and KDM1 regulates the lysosomal degradation of mutp53 in colon cancer cells undergoing TG-induced ER stress.

Post-translational modifications (PTMs) play a key role in regulating protein/protein interactions and protein stability, thus influencing protein expression and function. HSPs and p53, including its wtp53 and mutp53 forms, make no exception to this rule, although the impact of PTMs on the regulation of these proteins has not yet been fully elucidated, particularly in the case of mutp53. These proteins, unlike wtp53, can behave as oncogenes, making their targeting an important step for successful anticancer therapy. We previously reported that mutp53 is degraded, preferentially via CMA, in col

Clinical Significance of Nuclear Yin-Yang Overexpression Evaluated by Immunohistochemistry in Tissue Microarrays and Digital Pathology Analysis: A Useful Prognostic Tool for Breast Cancer.

Yin Yang 1 (YY1) is a multifunctional transcription factor implicated in gene regulation, cell proliferation, and survival. While its role in breast cancer (BC) has been explored, its prognostic significance remains controversial. In this study, we evaluated nuclear YY1 expression in 276 BC tissue samples using immunohistochemistry (IHC), tissue microarrays (TMAs), and digital pathology (DP). Nuclear staining was quantified using Aperio ImageScope software, focusing on tumor regions to avoid confounding from stromal or non-tumor tissues. This selective and standardized approach enabled precise

Bidirectional Modulation of α-Glucosidase by Steroidal Saponins From Polygonatum kingianum var. grandifolium: Inhibitory and Activating Effects Align With the Yin-Yang Balance Theory.

Three new steroidal saponins, kingianoside L-N (1-3), whose structures were elucidated through comprehensive spectroscopic analysis, and 15 known compounds (4-18) were isolated from Polygonatum kingianum var. grandifolium, a source of the traditional antihyperglycemic medicine Polygonati rhizome. The effects of compounds 1-13 on α-glucosidase activity were evaluated in vitro. Compound 4 exhibited stronger α-glucosidase inhibitory activity than the positive control acarbose, with a half-maximal inhibitory concentration of 213.9 µM, suggesting its potential as an inhibitor with hypoglycemic effe

The Yin and Yang of support from significant others: Influence of general social support and partner support of avoidance in the context of treatment for social anxiety disorder.

Support from social networks is generally considered to protect against mental disorder but in some circumstances support for negative behaviours (such as avoidance) may be counterproductive. Given the critical interplay between social anxiety disorder and social interactions, it is surprising that the relationship of support from significant others to this disorder has received so little attention. The current study evaluated the reciprocal relationships between perceived social support and perceived partner support for avoidance behaviours (avoidance support) among a sample of 131 participan

Mechanism differences between typical yin and typical yang personality individuals assessed by Five-Pattern Personality Inventory (FPPI): Evidence from resting-state brain functional networks.

BACKGROUND: Most studies assessing brain-personality mechanisms have used Western personality questionnaires. However, Western personality questionnaires may not objectively reflect the personality characteristics of individuals in Eastern cultures such as China. Hence, we adopted the functional magnetic resonance imaging (fMRI) and the Chinese localized scale, FPPI, to explore the brain mechanisms differences of typical yin and typical yang personalities of individuals in China. METHODS: 30 typical yin personality participants (TYI) and 34 typical yang personality participants (TYA) were enr

Altered Resting-State Brain Activities in Drug-Naïve Major Depressive Disorder Assessed by fMRI: Associations With Somatic Symptoms Defined by Yin-Yang Theory of the Traditional Chinese Medicine.

Identification of biological markers for defining subtypes of major depressive disorder (MDD) is critical for better understanding MDD pathophysiology and finding effective treatment intervention. The "Yin and Yang" theory is a fundamental concept of traditional Chinese Medicine (TCM). The theory differentiates MDD patients into two subtypes, Yin and Yang, based on their somatic symptoms, which had empirically been used for the delivery of effective treatment in East Asia. Nonetheless, neural processes underlying Yin and Yang types in MDD are poorly understood. In this study, we aim to provide

Extracellular vesicles from Li-Qi-Yang-Yin formula attenuate LPS-stimulated inflammatory injury in human colonic epithelial cells in vitro by delivering bioactive metabolites and suppressing the oxidative stress-inflammation axis.

BACKGROUND: The Li-Qi-Yang-Yin (LQYY) formula, a traditional Chinese medicine (TCMs) compound, holds potential in managing gastrointestinal disorders. Extracellular vesicles (EVs) derived from medicinal plants are emerging as novel carriers of bioactive compounds, yet their role in multi-herb formulations like LQYY remains largely unexplored. This study aimed to isolate LEVs from the LQYY formula and investigate their protective effects against inflammation and oxidative stress in intestinal epithelial cells. METHODS: Li-Qi-Yang-Yin formula contains EVs were isolated from the LQYY decoction v
