EDC-2: The Endocrine Society's Second Scientific Statement on Endocrine-Disrupting Chemicals.

The Endocrine Society's first Scientific Statement in 2009 provided a wake-up call to the scientific community about how environmental endocrine-disrupting chemicals (EDCs) affect health and disease. Five years later, a substantially larger body of literature has solidified our understanding of plausible mechanisms underlying EDC actions and how exposures in animals and humans-especially during development-may lay the foundations for disease later in life. At this point in history, we have much stronger knowledge about how EDCs alter gene-environment interactions via physiological, cellular, m

The influence of endocrine disruptors on growth and development of children.

PURPOSE OF REVIEW: This review describes the most recent data about the effects of endocrine disrupting compounds (EDCs) on infant and early childhood growth and reproductive tract development as well as controversies in the field. RECENT FINDINGS: EDCs are present in pregnant women, young children and adolescents. Whether the level of exposure contributes to disease is an ongoing debate. Epidemiological studies suggest associations between prenatal EDC exposure and disease outcome, but animal studies using controlled EDC exposure have varying results with underlying mechanisms largely unknow

[Endocrine disruptors and obesity: obesogens].

Incidence and prevalence of owerweight and obesity have greatly increased over the past three decades in almost all countries around the world. This phenomenon is not easily explained by lifestyle changes in populations with very different initial habits. This has led to consider the influence of other factors, the so-called endocrine disruptors, and more specifically obesogens. This study reviewed the available evidence about polluting chemical substances which may potentially be obesogens in humans: DES, genistein, bisphenol A, organotins (TBT, TPT), and phthalates. The first three groups of

Cumulative effects of in utero administration of mixtures of "antiandrogens" on male rat reproductive development.

Although risk assessments are typically conducted on a chemical-by-chemical basis, the 1996 Food Quality Protection Act (FQPA) required the Environmental Protection Agency (EPA) to consider cumulative risk of chemicals that act via a common mechanism of toxicity. To this end, we are conducting studies with mixtures to provide a framework for assessing the cumulative effects of "antiandrogenic" chemicals. Rats were dosed during pregnancy with antiandrogens singly or in pairs at dosage levels equivalent to about one half of the ED50 for hypospadias or epididymal agenesis. The pairs include: AR a

Effects of inhaled municipal refuse incinerator fly ash in the guinea pig.

Fly ash was collected from two municipal refuse incinerators. It was analyzed for heavy metals, elements, and a wide range of toxic organics. It was resuspended in air for inhalation exposure of guinea pigs. These animals were exposed at high concentrations of each ash 6 h/d for 5 d, and tissues were taken 45 d after the exposure. Following the first exposure and after each daily exposure the ventilatory response of these animals upon challenge with CO2 was found to be depressed. Recovery occurred following exposure. Heavy metals, cadmium, lead, zinc, and mercury were elevated in the lungs of

[Simultaneous determination of eight additives in polyethylene food contact materials by ultrahigh-performance liquid chromatography].

Measurement of additive residues in food contact materials is important for safety monitoring at the initial stage. Most of the current studies focus on the determination of the migration amounts of chemical hazards from food contact materials into food simulants. Studies on chemical hazard residues in food contact materials are limited to monomers, oligomers, heavy metals, phthalic acid esters, and biphenols, which are known environmental pollutants. Only a few studies have investigated analysis methods for additive residues in food contact materials. In this study, the main factors (monitori

Hepatic and renal tissue damage in Balb/c mice exposed to diisodecyl phthalate: The role of oxidative stress pathways.

Diisodecyl phthalate (DIDP) is commonly used as a plasticizer in industrial and consumer products, however, its toxicity remains unclear. This study investigated the possible involvement of oxidative stress in DIDP-induced liver and kidney toxicity. Liver function and kidney function, tissue lesions, oxidative stress biomarkers, inflammatory mediators and apoptosis factors were investigated in this study. The results showed that oral exposure to DIDP induced a marked increase in lever of alanine aminotransferase (ALT), aspartate aminotransferase (AST), urinary nitrogen (UREA) and creatinine (C

Phthalates modulate steroid 5-reductase transcripts in the Western clawed frog embryo.

Phthalates are used worldwide in the manufacturing of plastics, added to cosmetic products, personal care products, pharmaceuticals, medical devices, and paints; and are widely detected in soil, surface water, and organism tissues. Phthalate esters have been previously shown to interfere with the endocrine system in vertebrates. However, few studies have investigated the effects of phthalates on testosterone-converting enzymes that affect hormone levels and reproduction. In the present study, we exposed the Western clawed frog (Silurana tropicalis) to 0.1, 1, and 10 μM diethylhexyl phthalate (

[Endocrine disruptor compounds and their role in the developmental programming of the reproductive axis].

Different perturbations during fetal and postnatal development unleash endocrine adaptations that permanently alter metabolism, increasing the susceptibility to develop later disease, process known as "developmental programming." Endocrine disruptor compounds (EDC) are widely spread in the environment and display estrogenic, anti-estrogenic or anti-androgenic activity; they are lipophilic and stored for long periods in the adipose tissue. Maternal exposure to EDC during pregnancy and lactation produces the exposure of the fetus and neonate through placenta and breast milk. Epidemiological and

Adverse effects on development of the reproductive system in male offspring of rats given monobutyl phthalate, a metabolite of dibutyl phthalate, during late pregnancy.

The objective of this study was to determine the adverse effects of monobutyl phthalate (MBuP), a major metabolite of dibutyl phthalate (DBP), on development of the reproductive system in offspring following maternal administration during late pregnancy, and to assess the role of MBuP in the antiandrogenic effects of DBP. Pregnant rats were given MBuP by gastric intubation at 250, 500, or 750 mg/kg on days 15 through 17 of pregnancy. Maternal body weight gain and food consumption during the administration period were significantly decreased at 500 mg/kg and higher and at 750 mg/kg, respectivel

Platelet mitochondrial DNA methylation mediates the association of bisphenol, phthalate, and paraben exposures with type 2 diabetes mellitus: An exploratory nested case-control study.

Exposure to environmental pollutants has been found to be associated with epigenetic modifications of platelet mitochondria, which may influence the risk of type 2 diabetes mellitus (T2DM). However, research on the relationship between exposure to environmental endocrine disrupting chemicals (EDCs) and T2DM remains very limited at the molecular level of mitochondrial epigenetic alterations. This study aims to investigate the impact of mixed exposure to bisphenols (BPs), phthalates (PAEs), and parabens (PBs) on T2DM and platelet mitochondrial DNA (mtDNA) methylation, using a nested case-control

Oxidative stress as a pathway linking maternal phthalate and bisphenol exposure to childhood allergy and lung function: Mediation and interaction analyses.

Phthalates and bisphenols may contribute to childhood allergies through oxidative stress (OS). We examined OS as a potential pathway using two approaches: maternal OS biomarkers to evaluate mediation, and an OS genetic pathway function score reflecting child-level susceptibility to evaluate gene-environment interactions. In an Australian population-based birth cohort (n = 797), nine phthalate metabolites, three bisphenols, and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were measured from spot urine samples at 36 weeks' gestation, with allergic and lung function outcomes assessed up to age four. Breastfeeding Duration and Anogenital Distance in 2-Year-Old Infants.

BACKGROUND: The anogenital distance (AGD) is an anthropometric marker determined by exposures to androgens in utero and throughout the first few months of life. Early exposures to endocrine-disrupting chemicals such as phthalates have been significantly associated with shortened AGD in boys. Limited studies have explored phthalate concentrations in breast milk and infant formula. OBJECTIVE: To explore the associations between breastfeeding duration and AGD measures in infants. MATERIALS AND METHODS: MALAMA (Medio Ambiente y Lactancia Materna) is a follow-up study of 430 mother-child pairs, f

Prenatal phthalate, triclosan, and bisphenol A exposures and child visual-spatial abilities.

INTRODUCTION: During fetal development, sex steroids influence sexually dimorphic behaviors, such as visual-spatial abilities. Thus, endocrine disrupting chemicals that impact sex steroids during gestation may affect these behaviors. OBJECTIVE: We investigated the relationship between prenatal urinary phthalate metabolite, triclosan, and BPA concentrations and visual-spatial abilities in a prospective cohort of 198 mother-child dyads. METHODS: Data are from a prospective cohort in Cincinnati, OH (HOME Study). We measured nine phthalate metabolites, triclosan, and BPA in maternal urine sample

Prenatal phthalate causes cryptorchidism postnatally by inducing transabdominal ascent of the testis in fetal rats.

Phathalate esters, which are commonly used as plasticizers for polyvinyl chloride, are also well known to disturb Sertoli cells. This study aims to show the effect of prenatally administered phthalate on testicular descent in pre- and postnatal rats. Pregnant rats were exposed to mono-n-butyl phthalate (MBP) by gavage from the 15th to the 18th gestational days. Rats administered with solvent only were used as controls. In 20-day-old fetuses (n = 15), the degree of transabdominal testicular ascent in relation to the bladder neck was thus found to be significantly higher in MBP-treated rats than

Associations between exposure to environmental pollutants, metabolic syndrome risk, and obesity-related anthropometric indices.

Environmental pollutants-including heavy metals, phthalates, bisphenol A (BPA), polycyclic aromatic hydrocarbons (assessed via urinary 1-hydroxypyrene), and volatile organic compounds (assessed via benzene metabolites)-have been linked to metabolic dysfunction. Yet, population-based evidence across age groups remains limited. This study examined associations between exposure to multiple pollutants and metabolic syndrome (MetS) and its components, focusing on novel obesity-related anthropometric indices that capture subtle metabolic alterations. Originally designed as a case-control study, it w
