From fungal diversity to antimicrobial innovation: the potential of biotransformation in the era of resistance.

Antimicrobial resistance (AMR) continues to outpace the development of new anti-infective agents, particularly against priority bacterial pathogens such as Acinetobacter baumannii, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Staphylococcus aureus, as well as clinically relevant fungi including Candida auris. In this scenario, biotransformation has emerged as a complementary innovation strategy for antimicrobial discovery because it expands the chemical space around bioactive scaffolds through selective enzymatic or whole-cell modification. Among the available biocatalysts, fungi are esp

Acetohydroxyacid Synthase (AHAS) as a Promising and Underexplored Target for the Development of New Antifungal Agents.

INTRODUCTION: Fungi are ubiquitous and play crucial ecological roles, but fungal infections pose serious threats to human, animal, and environmental health, with a significant economic and social burden. Current antifungal therapies face challenges, such as limited drugs, toxicity, and resistance, highlighting the urgent need for drugs with new mechanisms of action. The enzyme acetohydroxyacid synthase (AHAS) is a promising target, as it is involved in branched-chain amino acid biosynthesis, a pathway lacking in animals and already explored in herbicide development. METHODS: We conducted an i

Dual action electrochemical bandage operated by a programmable multimodal wearable potentiostat.

We have developed electrochemical bandage (e-bandage) prototypes that generate the reactive oxygen species hypochlorous acid (HOCl) or hydrogen peroxide (H2O2) for potential use to treat biofilm-infected wounds in humans. We have shown that both e-bandage-generated HOCl and H2O2 kill biofilms in vitro and in infected wounds on mice, with the former being more active in vitro. The H2O2-generating e-bandage, more so than the HOCl-generating e-bandage, was associated with improved healing of infected wounds. Here, a strategy in which H2O2 and HOCl are alternately generated-for dual action-was exp

Evaluation of an automated far ultraviolet-C light technology for decontamination of surfaces and aerosolized viruses in bathrooms.

BACKGROUND: Aerosols generated during toilet flushing are a potential source for transmission of viral and bacterial pathogens in bathrooms. However, manual decontamination of bathrooms after each use is not feasible. METHODS: We tested the efficacy of a wall-mounted far ultraviolet-C (UV-C) light technology that only delivers far UV-C when people are not present for decontamination of surfaces and aerosolized viral particles in an unoccupied hospital bathroom. A quantitative disk carrier test method was used to test efficacy against organisms on steel disk carriers placed in 9 sites in the b

Evaluation of Resistance Development to the Gwt1 Inhibitor Manogepix (APX001A) in Candida Species.

Manogepix (MGX) targets the conserved fungal Gwt1 enzyme required for acylation of inositol early in the glycosylphosphatidylinositol biosynthesis pathway. The prodrug fosmanogepix is currently in clinical development for the treatment of invasive fungal infections. We determined that the median frequencies of spontaneous mutations conferring reduced susceptibility to MGX in Candida albicans, C. glabrata, and C. parapsilosis ranged from 3 × 10-8 to <1.85 × 10-8 Serial passage on agar identified mutants of C. albicans and C. parapsilosis with reduced susceptibility to MGX; however, this methodo

Wastewater and environmental sampling holds potential for antimicrobial resistance surveillance in food-producing animals - a pilot study in South African abattoirs.

Antimicrobial resistance (AMR) poses a significant global One Health challenge that causes increased mortality and a high financial burden. Animal production contributes to AMR, as more than half of antimicrobials are used in food-producing animals globally. There is a growing body of literature on AMR in food-producing animals in African countries, but the surveillance practices across countries vary considerably. This pilot study aims to explore the potential of wastewater and environmental surveillance (WES) of AMR and its extension to the veterinary field. Floor drainage swab (n = 18, 3/ab

Wild-type MIC distributions and epidemiological cut-off values for ibrexafungerp, manogepix, and rezafungin against WHO fungal priority pathogens.

OBJECTIVE: This study gathered public data regarding the in vitro activity of three novel antifungal agents, ibrexafungerp, manogepix, and rezafungin, against the fungi listed in the WHO fungal priority pathogens list. METHODS: Epidemiological cut-off values (ECVs/ECOFFs) were established for species with sufficient data (≥100 MIC values) and when distributions met further essential criteria. For pathogens in which data did not fulfil one or more essential criteria, tentative ECVs (T-ECVs/T-ECOFFs) were proposed. The values were determined by a comprehensive analysis that included visual insp

Epidemiological shifts and advances in research on early diagnosis of invasive fungal infection in critically ill patients.

Invasive fungal infections (IFI) primarily occur in immunocompromised patients, particularly those in intensive care units (ICU). Due to the use of immunosuppressive agents, invasive therapeutic procedures, and advancements in diagnostic technologies, the detection rate of IFI has shown a significant upward trend. This review aims to explore epidemiological changes in the field of IFI, early detection techniques, and the application of artificial intelligence (AI) technologies. We conducted a literature review using PubMed data up to April 2025, focusing on studies related to IFI. Specifically

Machine learning enables identification of an alternative yeast galactose utilization pathway.

How genomic differences contribute to phenotypic differences is a major question in biology. The recently characterized genomes, isolation environments, and qualitative patterns of growth on 122 sources and conditions of 1,154 strains from 1,049 fungal species (nearly all known) in the yeast subphylum Saccharomycotina provide a powerful, yet complex, dataset for addressing this question. We used a random forest algorithm trained on these genomic, metabolic, and environmental data to predict growth on several carbon sources with high accuracy. Known structural genes involved in assimilation of

Antimicrobial properties and biocompatibility of semi-synthetic carbohydrate-based ionic hydrogels.

Hydrogels have gained significant interest in the last decades, especially in the medical sector, due to their versatile properties. While hydrogels from naturally occurring polysaccharides (e.g. cellulose) are well-known, those produced from polymerizable carbohydrate-based monomers remain underexplored. However, these semi-synthetic hydrogels offer the great advantage of having adjustable properties for customization depending on their application. The objective of this study was to characterize semi-synthetic carbohydrate-based ionic hydrogels produced from GVIM-I (glucosyl vinyl imidazoliu

Identification of Essential Genes and Fluconazole Susceptibility Genes in Candida glabrata by Profiling Hermes Transposon Insertions.

Within the budding yeasts, the opportunistic pathogen Candida glabrata and other members of the Nakaseomyces clade have developed virulence traits independently from C. albicans and C. auris To begin exploring the genetic basis of C. glabrata virulence and its innate resistance to antifungals, we launched the Hermes transposon from a plasmid and sequenced more than 500,000 different semi-random insertions throughout the genome. With machine learning, we identified 1278 protein-encoding genes (25% of total) that could not tolerate transposon insertions and are likely essential for C. glabrata f
