Development of a model of carcinoma of the pancreas in rats was approached by attempting to identify chemicals that (a) behave as mutagens and (b) localize in the pancreas following systemic administration; and then to study the effects of long-term administration. Azaserine was selected because it behaves as a direct-acting mutagen in two bacterial test systems and because tissue distribution studies showed concentration especially in kidney and pancreas. Cholecystokinin (CCK) is a growth factor for normal pancreas. Numerous studies also suggest that CCK promotes pancreatic carcinogenesis in the rat. Our previous studies suggested that growth of preneoplastic pancreatic foci was stimulated by CCK more than that of normal pancreas. Pancreatic carcinomas have been induced in Wistar and W/LEW rats by administration of total azaserine doses of 150-520 mg/kg by injection or oral routes over periods of 5-52 weeks. The antineoplastic effects of thymoquinone (TQ) (which has antioxidant and anticarcinogenic effects) on neoplastic changes (atypical acinar cell foci, AACF) induced by azaserine were investigated, for the first time. Rats were randomly divided into five groups of 10 rats each (Cont, Tim, Az, AzTim1, and AzTim2). The Cont group was fed only with a standard diet. TQ (50 mg/L) was given orally (P.O.) to the Tim, AzTim1, and AzTim2 groups. TQ was given to Tim and AzTim1 after the first month, while AzTim2 group rats were given after the third month of azaserine injection, when AACFs began to form.

BACKGROUND: Atypical acinar cell foci (AACF) seen in pancreatic cancer are fatal and have been studied with some causative agents. However, for the first time, the effect of acetylsalicylic acid with nitric oxide (NO-ASA) on AACF was examined in this study. It was previously shown that Babesia-infected blood maintained in vitro incorporated high levels of 3H hypoxanthine and that the uptake could be suppressed by babesicidal drugs.
