Phylogeny and chemistry of biological mineral transport.

Three physiologically mineralizing tissues - teeth, cartilage and bone - have critical common elements and important evolutionary relationships. Phylogenetically the most ancient densely mineralized tissue is teeth. In jawless fishes without skeletons, tooth formation included epithelial transport of phosphates, a process echoed later in bone physiology. Cartilage and mineralized cartilage are skeletal elements separate from bone, but with metabolic features common to bone. Cartilage mineralization is coordinated with high expression of tissue nonspecific alkaline phosphatase and PHOSPHO1 to h

[Mechanisms of oxygen transport across biological membranes].

Using metallic microelectrodes, intracellular measurements were performed of changes in O2 concentration for spherical oocytes of hens by their replacement from normal (PO2=150 torr) to hyperoxic (to 600 torr) conditions, and vice versa. The curves of O2 influx and efflux were recorded in pulse regime of the working electrode polarization. The character of these curves can be expressed by the following equations: m(t)==c==e=y1t+y0 and m (t) =e-y1t+v0+c, where v1 is determined by the coefficient of cell membrane permeability for O2. Valid differences were shown between values of v1 for oxygen i

Anion transport in biologically relevant lipid mixtures.

A potent squaramide-based anion transporter was used to evaluate how vesicles prepared with tailored lipid compositions, which mimic organelle membranes, can impact transmembrane transport. Using HPTS and ISE assays, we show that the lipid environment and POPE flip-flop dynamics significantly influence transport efficiency.

Biological Functions and Clinical Significance of the ABCG1 Transporter.

ATP-binding cassette (ABC) transporters are a large family of proteins that transport various substances across cell membranes using energy from ATP hydrolysis. ATP-binding cassette sub-family G member 1 (ABCG1) is a member of the ABCG subfamily of transporters and performs many important functions, such as the export of cholesterol and some other lipids across the membranes of various cells. Cholesterol transport is the mechanism that links metabolism and the innate immune system. Due to its lipid transport function, ABCG1 may contribute to the prevention of atherosclerosis and is involved in

Drug & virus transport across biological barriers: interactions, diffusion, partitioning, permeability, and selectivity.

Biological barriers protect the human body by selectively blocking foreign material. Designing particles with coatings that efficiently transport across these barriers can increase the effectiveness and feasibility of advanced therapeutics. In particular, the mucus barrier protects the intestines, lungs, eyes, etc., complicating oral, inhaled, or ocular drug delivery. Heuristics for particle design are currently limited to the rate of diffusion within the barrier. Relying on first-principles theories for colloidal scale interactions, a cohesive model of the transport of particles through biolo

Rapid transport of deformation-tuned nanoparticles across biological hydrogels and cellular barriers.

To optimally penetrate biological hydrogels such as mucus and the tumor interstitial matrix, nanoparticles (NPs) require physicochemical properties that would typically preclude cellular uptake, resulting in inefficient drug delivery. Here, we demonstrate that (poly(lactic-co-glycolic acid) (PLGA) core)-(lipid shell) NPs with moderate rigidity display enhanced diffusivity through mucus compared with some synthetic mucus penetration particles (MPPs), achieving a mucosal and tumor penetrating capability superior to that of both their soft and hard counterparts. Orally administered semi-elastic N

Potential role of serotonin as a biological reductant associated with copper transportation.

Serotonin (5-HT) is a neurotransmitter that is derived from tryptophan. Owing to a hydroxyl group attached to the indole nucleus, 5-HT exhibits a considerably higher redox activity than tryptophan. To gain insight into the biological relevance of the redox activity of 5-HT, the effect of Cu(I)-binding ligands on the 5-HT-mediated copper reduction was investigated. The d-d transition band of Cu(II) complexed with glycine [Cu(II)-Gly2] was not affected by addition of 5-HT alone but was diminished when a thioether-containing compound coexists with 5-HT. Concomitant with disappearance of the d-d t

Structural Studies of the Taurine Transporter: A Potential Biological Target from the GABA Transporter Subfamily in Cancer Therapy.

The taurine transporter (TauT, SLC6A6) is a member of the solute carrier 6 (SLC6) family, which plays multiple physiological roles. The SLC6 family is divided into four subfamilies: GABA (γ-aminobutyric acid), monoamine, glycine and neutral amino acid transporters. Proteins from the GABA group, including the taurine transporter, are primarily considered therapeutic targets for treating central nervous system disorders. However, recent studies have suggested that inhibitors of SLC6A6 could also serve as anticancer agents. Overexpression of TauT has been associated with the progression of colon

Optoelectronic-Coupled-Driven Microrobot for Biological Cargo Transport in Conductive Isosmotic Glucose Solution.

Electric field-driven micro/nanorobots, as micro/nanodevices with autonomous motion capability, have emerged as promising candidates for targeted cargo delivery in biomedical applications due to their advantages of label-free operation, selectivity, and controllability. In biological systems, many biological cargos need to be operated in conductive isosmotic solutions to ensure their viability. However, in the conductive solution, electric field-driven micro/nanorobots exhibit significantly reduced propulsion performance, despite retaining the capability to manipulate cargos by the dielectroph

Sensitizing curium luminescence through an antenna protein to investigate biological actinide transport mechanisms.

Worldwide stocks of actinides and lanthanide fission products produced through conventional nuclear spent fuel are increasing continuously, resulting in a growing risk of environmental and human exposure to these toxic radioactive metal ions. Understanding the biomolecular pathways involved in mammalian uptake, transport and storage of these f-elements is crucial to the development of new decontamination strategies and could also be beneficial to the design of new containment and separation processes. To start unraveling these pathways, our approach takes advantage of the unique spectroscopic

COMPUTATIONAL MODELING OF DRUG TRANSPORT AND PERFUSION WITHIN COMPLEX BIOLOGICAL SYSTEMS AND GROWING TUMORS.

Mass transport is a fundamental process in living organisms, responsible for the delivery of oxygen, nutrients, and drugs from blood vessels to tissues, as well as the removal of waste products back to the vascular and lymphatic systems. This exchange occurs across various biological barriers, including blood vessel walls and cellular membranes, and involves complex mechanical and biochemical interactions. Traditional experimental and clinical methods have provided valuable insights into mass transport, but due to the complexity and heterogeneity of biological systems, computational models are

Novel electrospun chitosan/PEO membranes for more predictive nanoparticle transport studies at biological barriers.

The design of safe and effective nanoparticles (NPs) for commercial and medical applications requires a profound understanding of NP translocation and effects at biological barriers. To gain mechanistic insights, physiologically relevant and accurate human in vitro biobarrier models are indispensable. However, current transfer models largely rely on artificial porous polymer membranes for the cultivation of cells, which do not provide a close mimic of the natural basal membrane and intrinsically provide limited permeability for NPs. In this study, electrospinning is exploited to develop thin c
