The effect of chalone-containing ethanol extract of rat skin (CCE) on the growth in mice of transplanted uterine cervix and skin carcinomas, hepatoma-22a, sarcoma-180 and leukemia L-1210 was studied. When CCE is added to the suspension of tumour cells (10 mg CCE/100 mg tumour tissue/ml saline) the most obvious retention of tumour growth is observed on squamous-cell carcinoma of uterine cervix (72.6%; p less than 0.01). For many years the existence of an antral inhibitory hormone or chalone has been controversial. Recent experimental findings however have suggested that there is an antral inhibitory mechanism, active against gastrin release and gastrin stimulated acid secretion, hat is suppressed in an alkaline milieu and stimulated in an acid milieu. Antral somatostatin possesses the characteristics necessary to fulfill this role, and these findings have prompted the hypothesis that somatostatin plays a vital role in the homeostasis of gastric acid secretion and is indeed the long lost antral chalone.

The ethanolic extract of rat skin contains epidermal G1- and G2-chalones which having been added to cell suspension of transplantable squamous-cell cornified carcinoma of mice cervix, inhibits its growth in recipients by 72,6 per cent. The same extract failed to inhibit the growth of transplantable mouse tumours of other histogenesis (hepatoma 22a, leukemia L-1210 and sarcoma 180). When added to cell suspension of transplantable carcinoma of mouse skin which had been anaplized during a long period of transplantation (over 10 years) this extract inhibits its growth by 39,2 per cent only.

In the course of purification of chalone deriving from the subcellular fractions of bovine spleens, the butanol extraction of smooth and rough microsomes supernatants was performed. After extraction the lower layer was treated with SDS, buffered at pH 5,8. The homogeneity of obtained fractions was investigated by means of polyacrylamide electrophoresis. The effect of inhibitory extracts of human granulocytes (chalone) on the circadian rhythm of myelopoietic colony formation in soft agar (CFUc) was studied over one year. Normally, the number of bone marrow CFUc shows a strong circadian rhythm. Preincubation of the cells with the extracts before plating modulated this rhythm with phase shifts and modifications of amplitude. The effects of the extracts oscillated between significant inhibition and stimulation over a 24 h period. No significant seasonal variations in the inhibitory effects were found.

We have previously shown that the growth response of the rat thyroid to a sustained elevation of the serum level of TSH, induced by goitrogen administration, is self-limiting. This study investigated the possibility that this limitation of growth is due to the inhibitory action of a chalone secreted by the thyroid follicular cells, the serum concentration of which increases as the gland grows. A study was made of the effect of an aqueous extract of human gastric mucosa on the mitotic activity of the surface epithelium of the mouse stomach. The extract was found to exert a statistically significant inhibitory action.
