Parastatin (porcine chromogranin A347-419), a novel chromogranin A-derived peptide, inhibits parathyroid cell secretion.

Chromogranin A (CgA), previously referred to as secretory protein-I, is a 50-kilodalton protein present in secretory granules of many endocrine and neuroendocrine cells. In the parathyroid it is present and cosecreted with PTH in response to hypocalcemia. CgA appears to be a precursor of bioactive peptides including pancreastatin, beta-granin, vasostatin, and chromostatin. The presence of several highly conserved pairs of basic amino acids, putative cleavage sites, in the CgA molecule suggests that other yet unidentified bioactive peptides might exist within the molecule. We tested this specul

[Chromogranin A gene antisense transgenic mice and the tissue specific promoter of chromogranin A gene].

The vector pGAS1C-lacZ was constructed which contains cga promoter-lacZ and cga promoter-cga antisense cDNA two major parts. The pGAS1C-lacZ was integrated into genome of PC-12 cells and lacZ gene expressed in PC-12 cells. We cut the pGAS1C-lacZ with Xba I and Sal I to get rid of the framework of plasmid pGAS1C-lacZ, then microinjected the other part which contains antisense DNA into female pronucleus of the fertile mouse eggs. All injected fertile eggs were transplanted into the oviduct of foster so that the injected eggs developed normally. We selected transgenic mice by PCR, and got 14 tran

Plasma chromogranin-A in primary hyperparathyroidism.

We sought an explanation for prior findings of high plasma chromogranin-A (Chr-A) in primary hyperparathyroidism. Chr-A was measured in plasma samples from 55 controls and 73 patients with primary hyperparathyroidism caused by adenoma (n = 14), sporadic or familial hyperplasia (n = 10), or familial multiple endocrine neoplasia type 1 (FMEN1; n = 49). Serum or plasma samples were also tested for calcium, PTH, gastrin, pancreatic polypeptide, CG alpha, and PRL. Plasma Chr-A was 34 +/- 10 in parathyroid adenoma, 55 +/- 33 in parathyroid hyperplasia without FMEN1, 63 +/- 88 in FMEN1, and 25 +/- 8

Limitations of Chromogranin A in clinical practice.

CONTEXT: Usefulness of circulating Chromogranin A (CgA) for the diagnosis of neuroendocrine tumors (NEN) is controversial. The aim of the present study was to assess the actual role of this marker as diagnostic tool. METHODS: Serum blood samples were obtained from 42 subjects affected with NEN, 120 subjects affected with non-endocrine neoplasias (non-NEN) and 100 non-neoplastic subjects affected with benign nodular goitre (NNG). Determination of CgA was performed by means of immunoradiometric assay. RESULTS: The CgA levels among NEN-patients were not significantly different from NNG and non-

Altered chromogranin A circulating levels in Meniere's disease.

OBJECTIVES: Meniere's disease (MD) is an inner ear disorder characterized by episodic vertigo, ear fullness, and hearing loss; usually vertigo attacks cluster in specific period. We studied in MD patients the circulating levels of chromogranin A (CgA) and vasostatin-1 (VS-1), secreted by the neuroendocrine system and involved in the regulation of the endothelial barrier function. METHODS: Serum levels were assessed in 37 MD patients and 36 controls. The ratio between VS-1 and CgA was calculated. RESULTS: CgA was increased in patients compared to controls (1.46 versus 0.67 nM, p = 0.01) while

Chromogranin As a Biochemical Marker of Neuroendocrine Tumors.

Comparative study of plasma chromogranin A levels was carried out in 227 patients with neuroendocrine tumors of various locations and 66 normal subjects, men and women, by standard ELISA. In patients with tumors of all locations (pancreas, stomach, small and large intestine, and lungs), the glycoprotein levels were significantly (p<0.000001) higher than in controls. The patients demonstrated high variability of chromogranin A levels; the maximum concentrations were detected in patients with neuroendocrine tumors of the stomach and lung. The highest median values of chromogranin A were found in

Chromogranin A: a novel regulator of parathyroid gland secretion.

Chromogranin A (CgA) is a major protein of the parathyroid gland that is costored and cosecreted with parathyroid hormone (PTH). CgA, widely distributed in endocrine and neuroendocrine cells in addition to parathyroid cells, appears to be a precursor of biologically active autocrine or paracrine peptides that include pancreastatin (CgA240-288) and parastatatin (CgA347-419), compounds that strongly inhibit stimulated secretion of PTH and costored CgA. The biosynthesis of CgA and PTH via gene expression and mRNA translation are noncoordinately altered by agents that affect parathyroid cell secre

A Critical Approach to Clinical Biochemistry of Chromogranin A.

Chromogranin A (CGA) is a member of the granin family of proteins which are widespread in endocrine, neuroendocrine, peripheral, and central nervous tissues, where they are typically found in secretory granules. It is well accepted that CGA cooperates to regulate synthesis and secretion of these various granule signaling molecules.Because of its ubiquitous distribution within neuroendocrine tissues, CGA can be a useful diagnostic marker for neuroendocrine neoplasms, including carcinoids, pheochromocytomas, neuroblastomas, medullary thyroid carcinomas (MTC), some pituitary tumors, functioning a

Application of region-specific immunoassay for human chromogranin A: substantial clue for detection and measurement of chromogranin A in human plasma.

Chromogranin A (CgA), a secretory protein, is co-released with catecholamines from storage vesicles. It is known to be elevated in the circulation of patients with neuroendocrine and endocrine tumors. For further investigation of the protein, especially in humans, it is essential to facilitate quantitative analysis of the protein in human biological materials. In order to introduce novel immunological methodology for this purpose, we purposely selected human CgA(344-374) for the synthetic immunogen to produce region-specific CgA antibodies. The anti-synthetic peptide antibody thus obtained mad

Chromogranin A in the olfactory system of the rat.

The olfactory bulb of the rat contains chromogranin A at a similar level as the adrenal gland or the hypophysis as revealed by immunoblots. Olfactory chromogranin A also displays the same size as chromogranin A of endocrine cells. In the hippocampus and other brain regions, we could not detect chromogranin A by immunoblotting. In contrast, chromogranin A messenger ribonucleic acid (using S1 nuclease protection assays) was observed in all brain regions examined, including the olfactory bulb. By in situ hybridization histochemistry with a complementary ribonucleic acid probe (280 nucleotides), a

The sequence of porcine chromogranin A messenger RNA demonstrates chromogranin A can serve as the precursor for the biologically active hormone, pancreastatin.

Specific oligonucleotide priming of double-stranded DNA has been employed to sequence a porcine chromogranin A adrenomedullary cDNA. Porcine chromogranin A is more than 80% identical to human, bovine, and rat chromogranin A at its deduced N- and C-termini. A 49-amino acid region of the porcine molecule is 59-71% homologous to corresponding areas of rat, bovine, and human chromogranin A, and identical to the amino acid sequence of porcine pancreastatin. The sequence is preceded by an arginine at the N-terminus and followed by a GKR sequence at the C-terminus. Thus, porcine chromogranin A can se

Chromogranin A pathway: from pathogenic molecule to renal disease.

BACKGROUND: Chromogranin A (CHGA) is an index granin protein critical for biogenesis and exocytotic release of catecholamine storage granules. It is elevated in plasma of patients with sympathetic over-activity and kidney dysfunction. Several CHGA polymorphisms are associated with hypertensive kidney disease. Previously, we unraveled the molecular mechanism by which CHGA expression is regulated in African Americans carrying a genetic variation associated with hypertensive chronic kidney disease (CKD). METHOD: Experimental CKD mouse model were created by 5/6th nephrectomy (Npx) using wild-type

Chromogranin A-derived peptides are involved in innate immunity.

New endogenous antimicrobial peptides (AMPs) derived from chromogranin A (CgA) are secreted by nervous, endocrine and immune cells during stress. They display antimicrobial activities by lytic effects at micromolar range using a pore-forming mechanism against Gram-positive bacteria, filamentous fungi and yeasts. These AMPs can also penetrate quickly into neutrophils (without lytic effects), where, similarly to "cell penetrating peptides", they interact with cytoplasmic calmodulin, and induce calcium influx via Store Operated Channels therefore triggering neutrophils activation. Cerebrospinal fluid chromogranin A is unchanged in Alzheimer dementia.

Several lines of evidence link chromogranin A (CgA), the major soluble protein in catecholamine storage vesicles, with the cholinergic nervous system, abnormalities of which may play a central role in memory deficits in Alzheimer dementia. Because of reported elevations of CgA in Alzheimer brains and its presence in the senile plaque lesions of such brains, we evaluated the concentration of CgA in cerebrospinal fluid of Alzheimer dementia patients and matched controls. CgA was detectable in each sample, but the results in dementia showed substantial overlap with and no significant (p = 0.55) d

Ca2(+)-induced conformational change and aggregation of chromogranin A.

Chromogranin A, the most abundant protein in bovine adrenal chromaffin granules, bound calmodulin in a Ca2(+)-dependent manner, and the calmodulin-binding property was utilized to purify chromogranin A. Chromogranin A has been described in the past as a "random-coil polypeptide" with little alpha-helix or beta-sheet conformation. However, circular dichroism measurements with pure, native chromogranin A revealed relatively high alpha-helical contents (40% at the intravesicular pH of 5.5). Fluorescence studies confirmed previous observations that chromogranin A binds Ca2+ with low affinity. Cons

Chromogranin A-Associated Tubulopathy in a Patient With Neuroendocrine Tumor.

Chromogranin A (CGA) is a peptide secreted by neuroendocrine cells and filtered by the kidneys. Excessive CGA secretion is common in neuroendocrine tumors (NETs) and has been linked to organ dysfunction, though direct kidney injury remains rarely described. We present a woman in her mid-70s diagnosed with metastatic, well-differentiated pancreatic NET who developed progressive renal dysfunction. Initial CGA levels exceeded 86,000 ng/mL and later rose to >500,000 ng/mL. Despite the absence of nephrotoxic exposures, imaging abnormalities, or significant proteinuria, her serum creatinine increase
