Bovine Enteroids as an In Vitro Model for Infection with Bovine Coronavirus.

Bovine coronavirus (BCoV) is one of the major viral pathogens of cattle, responsible for economic losses and causing a substantial impact on animal welfare. Several in vitro 2D models have been used to investigate BCoV infection and its pathogenesis. However, 3D enteroids are likely to be a better model with which to investigate host-pathogen interactions. This study established bovine enteroids as an in vitro replication system for BCoV, and we compared the expression of selected genes during the BCoV infection of the enteroids with the expression previously described in HCT-8 cells. Genetic characterization of bovine coronavirus in Vietnam.

A maximum clade credibility tree constructed using the full-length spike (S) and hemagglutinin-esterase genes revealed that Vietnamese Bovine coronavirus (BCoV) strains belong to a single cluster (C1); therefore, they might share a common origin with Cuban and Chinese BCoV strains. The omega values of cluster 1 (C1) and cluster 2 (C2) were 0.15734 and 0.11613, respectively, and naive empirical bayes analysis identified two amino acid positions (179 and 501) in the S protein in C1 and three amino acid positions (113, 501, and 525) in that of C2 that underwent positive selection (p > 99%). Bovine coronavirus presence in domestic bovine and antelopes sub-Saharan Africa: evidence from Namibia.

BACKGROUND: Bovine coronavirus (BoCV) causes significant economic losses to cattle farming due to mortality in calves, reduced growth performances and milk production in feedlots and dairy cattle. Worldwide distribution of BoCV has been demonstrated, although knowledge of its epidemiology in Africa, especially in the sub-Saharan region, is limited. RESULTS: In the present study, a total of 208 swab samples of wild ruminants and 435 bovines from different regions of Namibia were obtained and tested by a BoCV-specific qRT-PCR. Twenty-six bovine samples tested positive [26/435 (5.98%; 95CI: 3.94

Evidence of respiratory infection by bovine coronavirus in a neonatal calf.

BACKGROUND: Bovine coronavirus (BCoV) is a major pneumoenteric pathogen that causes diarrhea in calves and respiratory disease in cattle of all ages. While BCoV-associated diarrhea is well-documented, reports of respiratory infection in neonatal calves remain limited, particularly in the Republic of Korea (ROK). Here, we describe a case of BCoV-induced pulmonary infection in a calf that presented without clinical respiratory signs. CASE PRESENTATION: A 7-day-old Holstein calf developed white, watery diarrhea and dehydration that subsequently progressed to astasia, showing no respiratory signs

Study on the susceptibility of bovine coronavirus to BALB/c mice.

There are no other bovine coronavirus (BCoV) infection models except calves, which makes efficacy evaluation of vaccines and pathogenic mechanism research of BCoV inconvenient owing to their high value and inconvenient operation. This study aimed to establish a mouse model of BCoV infection. BCoV was used to infect 4-week-old male BALB/c mice and the optimal infection conditions were screened, including the following infection routes: gavage, intraperitoneal injection, and tail vein injection at doses of 1 × 108 TCID50, 2 × 108 TCID50 and 4 × 108 TCID50. Using the optimal infection conditions,

Detection of Bovine Coronavirus in Healthy and Diarrheic Dairy Calves.

BACKGROUND: BCoV is identified in both healthy and diarrheic calves, complicating its assessment as a primary pathogen. OBJECTIVES: To investigate the detection rates of bovine coronavirus (BCoV) in feces of healthy and diarrheic calves and to describe the usefulness of a pancoronavirus reverse transcriptase (RT) PCR (PanCoV-RT-PCR) assay to identify BCoV in samples of diarrheic calves. ANIMALS: Two hundred and eighty-six calves <21 days. Calves with liquid or semiliquid feces, temperature >39.5°C, and inappetence were considered as cases, and those that had pasty or firm feces and normal ph

A Review: Bovine Coronavirus Evolution, Molecular Epidemiology, and Genetic Variation.

Bovine coronavirus (BCoV) is a key pathogen causing calf diarrhea and bovine respiratory diseases, bringing sustained economic losses to the cattle industry. As an RNA virus, BCoV possesses high mutation and recombination capacities, leading to prominent genomic genetic diversity. The genome contains hypervariable and conserved regions, with the S (especially S1), HE and Open Reading Frame (ORF4) genes serving as major variation hotspots linked to viral antigenicity, tissue tropism shift and immune evasion. Host immune pressure drives strong positive selection on S protein antigenic variation.

Screening of Bovine Coronavirus Multiepitope Vaccine Candidates: An Immunoinformatics Approach.

Bovine coronavirus (BCoV) is a causative agent of enteric and respiratory disease in cattle. BCoV has been reported to cause a variety of animal diseases and is closely related to human coronaviruses; moreover, it has attracted extensive attention from both cattle farmers and researchers. With the rise of BCoV, a vaccine that is prophylactic and immunotherapeutic has to be utilized for a preemptive and adroit therapeutic approach. The aim of this study was to develop a novel multiepitope-based BCoV vaccine that can induce an immune response using a silicon reverse vaccinology approach. In this

Experimental intranasal infection reveals broad tissue tropism of bovine coronavirus.

Bovine coronavirus (BCoV) causes neonatal calf diarrhea, winter dysentery, and respiratory disease worldwide. However, its primary replication site remains uncertain, and systemic studies on tissue tropism are limited. We, therefore, intranasally inoculated colostrum-fed Holstein calves (6-13-day-old) with a BCoV isolate derived from diarrheic calf, and monitored clinical signs, gross and histopathological lesions, antigen distribution, and viral replication in multiple tissues. All calves were seropositive before challenge. Infected animals developed diarrhea without respiratory manifestation

Infection of calves with in-vivo passaged bovine parainfluenza-3 virus, alone or in combination with bovine respiratory syncytial virus and bovine coronavirus.

Bovine respiratory disease complex is etiologically complex and usually involves co-infection by several agents, including bovine parainfluenza virus-3 (BPIV-3), bovine respiratory syncytial virus (BRSV), and bovine coronavirus (BCoV). Traditionally, vaccines have been tested in seronegative calves infected with a single in vitro-passaged agent, often with little disease, resulting in unvaccinated subjects. To overcome the potential problem of attenuation coincident with in vitro culture of the viruses, cocktails of field isolates of BPIV-3s and BCoVs were passaged in the lungs of neonatal col

Detection and genomic characterization of an emerging bovine coronavirus in Southwest China.

Bovine coronavirus (BCoV) poses significant economic threats to the cattle industry, yet molecular epidemiological data from Southwest China remain scarce. In this study, we investigated the molecular epidemiology, genomic features, and antigenic characteristics of BCoV circulating in this region. Molecular detection showed that BCoV was frequently detected among clinically affected cattle, with farm-level clinical detection rates exceeding 75% in several major cattle-producing areas. Phylogenetic analysis showed that all identified strains belonged to the GII subclade, among which 11 formed a

Detection of bovine coronavirus in myocardial tissue of a naturally infected calf.

Coronaviruses are RNA viruses with high genomic plasticity and with the ability to infect a wide range of animal hosts, including humans. Among them, the highly pathogenic SARS-CoV-2, a betacoronavirus, is the causative agent of severe respiratory infection, and has also been associated with extrapulmonary manifestations, including cardiac involvement. BCoV, another betacoronavirus, is a major pathogen of cattle, primarily affecting the respiratory and enteric systems. Herein, a case of death in a calf with clinical signs consistent with BCoV infection is described. Potential cardiac involveme

Evaluation of inactivation of bovine coronavirus by low-level radiofrequency irradiation.

Inactivation of influenza A virus by radiofrequency (RF) energy exposure at levels near Institute of Electrical and Electronics Engineers (IEEE) safety thresholds has been reported. The authors hypothesized that this inactivation was through a structure-resonant energy transfer mechanism. If this hypothesis is confirmed, such a technology could be used to prevent transmission of virus in occupied public spaces where RF irradiation of surfaces could be performed at scale. The present study aims to both replicate and expand the previous work by investigating the neutralization of bovine coronavi

Photodynamic Inactivation of Bovine Coronavirus with the Photosensitizer Toluidine Blue O.

Coronaviruses (CoVs) belong to the group of enveloped positive-sense single-strand RNA viruses and are causative agents of respiratory, gastro-intestinal, and central nervous systems diseases in many host species, i.e., birds, mammals, and humans. Beta-CoVs revealed a great potential to cross the barrier between species by causing three epidemics/pandemics among humans in the 21st century. Considering the urgent need for powerful antiviral agents for decontamination, prevention, and treatment of BCoV infections, we turned our attention to the possibility of photodynamic inactivation with photo

Epidemiological survey and genetic diversity of bovine coronavirus in Northeast China.

In 2020, to trace the prevalence and evolution of bovine coronavirus (BCoV) in China, a total of 1383 samples (1016 fecal samples and 367 nasal swab samples) were collected from 1016 cattle exhibiting diarrhea symptoms on dairy farms and beef cattle farms in Heilongjiang Province, Northeast China. All samples were subjected to reverse transcription-polymerase chain reaction (RT-PCR) detection of the BCoV N gene, followed by an analysis of its epidemiology and genetic evolution. The results indicated that of the 1016 diarrhea-affected cattle, 15.45% (157/1016) were positive for BCoV, in which p

Bovine respiratory coronavirus enhances bacterial adherence by upregulating expression of cellular receptors on bovine respiratory epithelial cells.

Bovine coronavirus (BCoV) is one of the agents causing bovine respiratory disease complex (BRDC), with single infection tending to be mild to moderate; the probability of developing pneumonia in BRDC may be affected by viral and bacterial combinations. Previously, we reported that bovine respiratory syncytial virus (BRSV) infection enhances adherence of Pasteurella multocida (PM) to cells derived from the bovine lower respiratory tract but that BRSV infection in cells derived from the upper respiratory tract reduces PM adherence. In this study, we sought to clarify whether the modulation of ba

Investigating bovine coronavirus in Indian calves: Incidence, molecular evidence, and pathological role in bovine respiratory disease complex (BRDC).

BACKGROUND: Bovine coronavirus (BCoV) and bacterial pathogens contribute to bovine respiratory disease complex (BRDC) in young calves. However, the role of BCoV in BRDC occurrence and site-specific respiratory pathology in India remains poorly explored. AIMS: This study aimed to assess BCoV prevalence in BRDC cases. METHODS: We investigated 406 weaner calves (166 cattle, 240 buffaloes) up to ≤1 year, with respiratory distress and pulmonary lesions. RESULTS: BRDC cases exhibited 0.98% BCoV occurrence, confirmed by partial N gene amplification (172 bp) via RT-PCR and immunohistochemistry (IHC

Non-major histocompatibility complex-restricted cytotoxicity of bovine coronavirus-infected target cells mediated by bovine intestinal intraepithelial leukocytes.

Non-specific cellular mechanisms of defence against intestinal virus infections of cattle were investigated using bovine coronavirus (BCV) as a representative enteric virus. Since BCV infection is limited to the epithelial cells of the intestinal tract, defence mechanisms must be capable of acting at that site to be effective. Therefore, the intraepithelial leukocyte (IEL) population of the intestinal mucosa was chosen for initial study. Treatment of intestinal samples with DTT and EDTA in calcium- and magnesium-free buffers allowed recovery of viable IEL populations appropriate for further fu
