Bioactive venoms and toxins are emerging as a promising source of drug leads. Optimized through evolution, these compounds display remarkable selectivity and ligand affinity toward a range of relevant pharmacological targets. The successful development of new drugs from toxins is hampered in some areas by the chemical complexity of the active compounds, which limits the possibility of using chemical synthesis or recombinant strategies for drug lead generation. Marine paralytic shellfish toxins produced by marine microalgae is one such family of compounds. These compounds are highly potent bloc

The present study compares the CMFDA/FDA + motility- and the Most Probable Number (MPN) Dilution Culture + Motility methods for testing the viability of ≥10-<50 μm organisms in chlorine treated ballast water. The results of both methods were within the regulatory compliance criterion <10 organisms/mL, but the MPN-method revealed that growth-outs did occur. While the CMFDA/FDA method showed <0.5 organisms/mL, the MPN-method gave approx. 6 organisms/mL. This demonstrated that false negatives, i.e. living but not stained organisms, may occur when using the CMFDA/FDA-method for compliance testing

Two new macrolides, iriomoteolides-14a (1) and 14b (2) have been isolated from the marine dinoflagellate Amphidinium species (strain KCA09057). Compounds 1 and 2 are 15-membered macrolides, which are structural analogs of amphidinolides O (3) and P (4). The structures of 1 and 2 were assigned on the basis of detailed NMR analyses and chemical conversion studies. Compounds 1 and 2 showed moderate cytotoxic activity against human cervix adenocarcinoma HeLa cells.

Short flashes of blue light (bioluminescence) from dinoflagellates can reduce copepod grazing of light-emitting cells. Other protective strategies against grazing are toxicity, reduced cell chain length and altered swimming patterns in different phytoplankton. Both toxicity and bioluminescence capacity in dinoflagellates decrease in copepod-free cultures, but toxin production can be restored in response to chemical alarm signals from copepods, copepodamides. Here we show that strains of the dinoflagellates Lingulodinium polyedra and Alexandrium tamarense, kept in culture for 14 and 9 years res

An amphidinol-prioritized fractioning approach was for the first time developed to isolate multiple specialty metabolites such as amphidinols, carotenoids and fatty acids using the biomass of the marine microalgae Amphidinium carterae. The biomass was produced in a raceway photobioreactor and the exhausted culture media were reused, thus fulfilling sustainability criteria employing a circular economy concept. The integrated bioactive compounds-targeted approach presented here consisted of four steps with which recovery percentages of carotenoids, fatty acids and amphidinols of 97%, 82% and 99

Phosphorus (P) acts as a crucial limiting nutrient for the growth of marine phytoplankton cells and the formation of algal blooms. The dinoflagellate Alexandrium pacificum is known for causing frequent and intense blooms in specific estuarine and coastal regions. In this study, we investigated the growth and physiological transformations under conditions characterized by P-deficiency, NaH2PO4, and ATP. For the first time, an integrated comparative analysis of the secretome and proteome was performed to investigate the global protein expression profile of A. pacificum, with 355 and 2308 differe

During a survey of the production of goniodomin A (GDA) by Alexandrium pseudogonyaulax in Danish coastal waters, Krock et al. (2018) obtained mass spectral evidence for the presence of a truncated congener, herein termed GD754, having a molecular weight 14 Da lower than GDA and assigned it as goniodomin B (GDB). An erroneous structure of GDB involving deletion of a methylene group between rings B and D had previously been reported by Espiña et al. (2016) but without experimental details. HPLC properties reported by Krock for GD754 point to it being a homolog of GDA. Comparison of mass spectral

Dinoflagellate algae are a diverse group of single-celled eukaryotes, often living in marine environments. The majority of species are entirely free-living, but many can become symbionts with corals, jellyfish and other marine organisms. With rising sea temperatures, the function of the dinoflagellate photosynthetic machinery, and the redox state of the photosynthetic electron transport chain are impaired. This photosynthetic impairment is likely to be an important cause of coral bleaching. In the chloroplasts of plants and many algae, disturbance of the chloroplast redox state can be in part

The marine toxigenic dinoflagellate family Amphidomataceae, comprising Amphidoma and Azadinium, has been primarily characterized by thecal morphology examined by SEM. To further clarify the detailed morphology, the intracellular ultrastructure of five Azadinium species, encompassing two toxigenic species (Azadinium poporum and Az. spinosum) and three non-toxigenic species (Az. inconspicuum, Az. dalianense, and Az. zhuanum), was examined using TEM. The ultrastructure of two Amphidoma species was also examined for comparison. All species exhibited typical dinoflagellate organelles, i.e., nucleus

The expansion of the Atlantic blue crab, Callinectes sapidus, along the northern Adriatic coast of Italy is threatening local ecosystems and bivalves aquaculture. This invasive species can host the parasitic dinoflagellate Hematodinium perezi, but no records of this infection in blue crabs exist for the northern Adriatic Sea. To investigate the presence and prevalence of H. perezi in C. sapidus in this region, 209 commercial size specimens collected from seven lagoons were surveyed in spring 2024 using a combination of cytological and histological analyses alongside a specific quantitative PCR

Zinc is an essential metal to living organisms, including corals and their symbiotic microalgae (Symbiodiniaceae). Both Zn(II) deprivation and overload are capable of leading to dysfunctional metabolism, coral bleaching, and even organism death. The present work investigated the effects of chemically defined Zn species (free Zn, ZnO nanoparticles, and the complexes Zn-histidinate and Zn-EDTA) over the growth of the dinoflagellates Symbiodinium microadriaticum, Breviolum minutum, and Effrenium voratum, and on the trypsin-like proteolytic activity of the hydrocoral Millepora alcicornis. Amphidinolides C, F, and U, including C2-C4 analogs, are highly cytotoxic marine macrolides, mainly isolated from dinoflagellates of the genus Amphidinium. All these polyketides share a 75 % or more similar structure, highlighted by a macrolactone ring, at least one trans-2,5-substituted-THF motif and a characteristic polyenic side chain. From their isolation and absolute configurational assignment, the total synthesis of these marine macrolides represented an intense challenge to the organic synthesis community over the last 15 years, with around 14 research groups engaged in this inspiring t

Epizootiologists recurrently encounter symbionts and pathobionts in the haemolymph (blood equivalent) of shellfish. One such group is the dinoflagellate genus Hematodinium, which contains several species that cause debilitating disease in decapod crustaceans. The shore crab Carcinus maenas acts as a mobile reservoir of microparasites, including Hematodinium sp., thereby posing a risk to other co-located commercially important species, e.g. velvet crabs (Necora puber). Despite the widespread prevalence and documented seasonality of Hematodinium infection dynamics, there is a knowledge gap regar

Phosphorus (P) is an essential macronutrient impacting bloom formation of marine dinoflagellates. The dinoflagellate Alexandrium pacificum is a cosmopolitan species known to frequently cause dense blooms in estuarine and coastal waters worldwide, while the physiological and molecular responses of A. pacificum to P utilization are still not well understood. Herein, the growth, P utilization, toxin production and transcriptomes of A. pacificum grown under P-deficient, inorganic P-replete, and organic P-replete conditions were compared. The results indicated that P-deficient adversely affected th

Amphidinolides are a family of more than forty macrolides of varying sizes and complex structures isolated from dinoflagellates of the genus Amphidinium. Although all of them display potent-to-moderate cytotoxicity, their full bioactivity profile and mode of action have not been fully investigated. Access to enough material is needed for these studies, but samples of these compounds are limited due to the minute amounts that can only be obtained by either large-scale cultivation of the organism that produces them or by total synthesis. Of all the amphidinolides known to date, only the targets

BACKGROUND: Unicellular dinoflagellates are an important group of primary producers within the marine plankton community. Many of these species are capable of forming harmful algae blooms (HABs) and of producing potent phycotoxins, thereby causing deleterious impacts on their environment and posing a threat to human health. The recently discovered toxigenic dinoflagellate Azadinium spinosum is known to produce azaspiracid toxins. These toxins are most likely produced by polyketide synthases (PKS). Recently, PKS I-like transcripts have been identified in a number of dinoflagellate species. Desp

Ciguatera poisoning (CP) is the most prevalent marine toxin illness globally and disproportionately affects people from circumtropical regions due to the occurrence of the causative microalgae, Gambierdiscus. Increased reports of CP cases globally are linked to changing ocean temperatures, localized outbreaks in new regions and importation of ciguatoxin-contaminated fish into non-endemic regions. Gambierdiscus spp. produce several classes of bioactive polyether metabolites in addition to ciguatoxins, including gambierones, maitotoxins, gambieroxide, gambierol and gambieric acids. The role of t

Gambierdiscus spp. is mainly responsible for the ciguatera fish poisoning (CFP) around the world. The gambiertoxin produced by Gambierdiscus can be passed through the food chain to form ciguatoxins (CTXs) that cause ciguatoxins poisoning. However, the toxic effects of Gambierdiscus on fish through the food chain and related mechanism remains unclear. In this study, the toxicity of Gambierdiscus caribaeus on the marine medaka (Oryzias melastigma) was investigated, where the simulated food chain toxic algae-food organism-fish (G. caribaeus-Artemia metanauplii-O. melastigma) was set. The results

In 1968 Burkholder and associates (J. Antibiot. (Tokyo)1968, 21, 659-664) isolated the antifungal toxin goniodomin from an unidentified Puerto Rican dinoflagellate and partially characterized its structure. Subsequently, a metabolite of Alexandrium hiranoi was isolated by Murakami et al. from a bloom in Japan and its structure was established (Tetrahedron Lett.1988, 29, 1149-1152). The Japanese substance had strong similarities to Burkholder's but due to uncertainty as to whether it was identical or only similar, Murakami named his toxin goniodomin A. A detailed study of this question now prov
