In Focus: Spotted wing drosophila, Drosophila suzukii, across perspectives.

In August 2008, the first detection of the spotted wing drosophila, Drosophila suzukii, to the North America mainland in California caused great concern, as the fly was found infesting a variety of commercial fruits. Subsequent detections followed in Oregon, Washington, Florida and British Columbia in 2009; in Utah, North Carolina, South Carolina, Michigan, and Louisiana in 2010; and in Virginia, Montana, Wisconsin, Pennsylvania, New Jersey, Maryland and Mexico in 2011. In Europe, it has been detected in Italy and Spain in 2009 and in France in 2010. Economic costs to the grower from D. suzuki

Phototransduction in Drosophila.

The Drosophila visual transduction is the fastest known G protein-coupled signaling cascade and has been served as a model for understanding the molecular mechanisms of other G protein-coupled signaling cascades. Numbers of components in visual transduction machinery have been identified. Based on the functional characterization of these genes, a model for Drosophila phototransduction has been outlined, including rhodopsin activation, phosphoinoside signaling, and the opening of TRP and TRPL channels. Recently, the characterization of mutants, showing slow termination, revealed the physiologic

The Drosophila nephrocyte.

PURPOSE OF REVIEW: The functioning kidney requires proper organization in multiple cell types that mediate filtration and removal of wastes. Interest has increasingly focused on the podocyte as an important mediator of kidney function; defects in podocyte function likely mediate a broad palate of kidney dysfunctions. Here I explore recent work that establishes the Drosophila nephrocyte as a useful model for podocyte function and dysfunction. RECENT FINDINGS: Although described many decades in the past, recent evidence has emphasized important similarities in the molecules that construct the '

Drosophila TRP channels.

The transient receptor potential (TRP) superfamily comprises a large group of related cation channels that display surprising diversity in the specific modes of activation and cation selectivities. However, a unifying theme is that many TRP channels play important roles in sensory physiology. The superfamily includes 28 mammalian members, which are subdivided into multiple subfamilies. Each of these subfamilies is represented by at least one of the 13 members in Drosophila, suggesting common evolutionary relationships. In recent years it has become clear that TRP channels in flies and mammals

Response of Wild Spotted Wing Drosophila (Drosophila suzukii) to Microbial Volatiles.

The olfactory cues used by various animals to detect and identify food items often include volatile organic compounds (VOCs) produced by food-associated microorganisms. Microbial VOCs have potential as lures to trap animal pests, including insect crop pests. This study investigated microorganisms whose VOCs are attractive to natural populations of the spotted wing drosophila (SWD), an invasive insect pest of ripening fruits. The microorganisms readily cultured from wild SWD and SWD-infested fruits included yeasts, especially Hanseniaspora species, and various bacteria, including Proteobacteria

Mitochondrial DNA variation among the Drosophila athabasca semispecies and Drosophila affinis.

Morphology, allozymes, and levels of postreproductive isolation indicate that the semispecies of Drosophila athabasca are as recently diverged as typical populations within a Drosophila species. However, levels of behavioral isolation, divergence in male mating song, and divergence in X-chromosome inversions suggest a much more ancient divergence. In this article, we have examined mitochondrial DNA restriction site variation within and among the three semispecies. These data support the hypothesis that the three semispecies of Drosophila athabasca are at least as recently diverged as are typic

Vitellogenetic defects in hybrids of the species pair Drosophila virilis and Drosophila texana.

In interspecific matings between the species Drosophila virilis and Drosophila texana, female sterility can be observed in F2 backcross females and in F2 hybrid females. The results presented in this report show that the female sterility, whenever it exists, is due to prevention of vitellogenin synthesis in the fat body, but other abnormalities such as defects with the hybrid ovaries are not excluded. The observation that sterility appears among females from backcrosses suggests that incompatibilities between interspecific genes may cause female sterility even in the presence of a complete hab

Inheritance of cold shock tolerance in hybrids of Drosophila virilis and Drosophila lummei.

The genetic basis of the difference in cold shock tolerance between the southern temperate Drosophila virilis and its boreal relative D. lummei is studied. After adult eclosion, the parental stocks, reciprocal F1 and backcross hybrids were pretreated for eight days at 18 degrees C or at 6 degrees C. The cold shock used consisted of fast cooling to -10 degrees C and exposure to this temperature for varying lengths of time. D. lummei tolerated such exposure for 40-50% longer than did D. virilis (100-135% after acclimation). Reciprocal F1 females, differing only in their maternal cytoplasm deviat

Drosophila As a Cancer Model.

Over the last few decades, Drosophila cancer models have made great contributions to our understanding toward fundamental cancer processes. Particularly, the development of genetic mosaic technique in Drosophila has enabled us to recapitulate basic aspects of human cancers, including clonal evolution, tumor microenvironment, cancer cachexia, and anticancer drug resistance. The mosaic technique has also led to the discovery of important tumor-suppressor pathways such as the Hippo pathway and the elucidation of the mechanisms underlying tumor growth and metastasis via regulation of cell polarity

Hearing regulates Drosophila aggression.

Aggression is a universal social behavior important for the acquisition of food, mates, territory, and social status. Aggression in Drosophila is context-dependent and can thus be expected to involve inputs from multiple sensory modalities. Here, we use mechanical disruption and genetic approaches in Drosophila melanogaster to identify hearing as an important sensory modality in the context of intermale aggressive behavior. We demonstrate that neuronal silencing and targeted knockdown of hearing genes in the fly's auditory organ elicit abnormal aggression. Further, we show that exposure to cou

Nociception in Drosophila Larvae.

Nociception is the sensory modality by which animals sense stimuli associated with injury or potential tissue damage. When Drosophila larvae encounter a noxious thermal, chemical, or mechanical stimulus, they perform a stereotyped rolling behavior. These noxious stimuli are detected by polymodal nociceptor neurons that tile the larval epidermis. Although several types of sensory neurons feed into the nociceptive behavioral output, the highly branched class IV multidendritic arborization neurons are the most critical. At the molecular level, Drosophila nociception shares many conserved features

Mitochondrial encephalomyopathy in Drosophila.

Mitochondrial encephalomyopathies are common and devastating multisystem genetic disorders characterized by neuromuscular dysfunction and tissue degeneration. Point mutations in the human mitochondrial ATP6 gene are known to cause several related mitochondrial disorders: NARP (neuropathy, ataxia, and retinitis pigmentosa), MILS (maternally inherited Leigh's syndrome), and FBSN (familial bilateral striatal necrosis). We identified a pathogenic mutation in the Drosophila mitochondrial ATP6 gene that causes progressive, adult-onset neuromuscular dysfunction and myodegeneration. Our results demons

Transvection effects in Drosophila.

An unusual feature of the Diptera is that homologous chromosomes are intimately synapsed in somatic cells. At a number of loci in Drosophila, this pairing can significantly influence gene expression. Such influences were first detected within the bithorax complex (BX-C) by E.B. Lewis, who coined the term transvection to describe them. Most cases of transvection involve the action of enhancers in trans. At several loci deletion of the promoter greatly increases this action in trans, suggesting that enhancers are normally tethered in cis by the promoter region. Transvection can also occur by the

Postembryonic hematopoiesis in Drosophila.

We have investigated the blood cell types present in Drosophila at postembryonic stages and have analysed their modifications during development and under immune conditions. The anterior lobes of the larval hematopoietic organ or lymph gland contain numerous active secretory cells, plasmatocytes, few crystal cells, and a number of undifferentiated prohemocytes. The posterior lobes contain essentially prohemocytes. The blood cell population in larval hemolymph differs and consists mainly of plasmatocytes which are phagocytes, and of a low percentage of crystal cells which reportedly play a role

Genetics of a Drosophila phenoloxidase.

An electrophoretic mobility variant of phenoloxidase in a lz stock of Drosophila melanogaster was identified as the A3 component of the phenoloxidase complex by using two different activators to study enzyme activity-natural activator isolated from pupae and 50% 2-propanol. The structural gene for the A3 proenzyme, Dox-3, was not associated with lz on the X chromosome; it mapped to the right of rdo (53.1) and left of M(2)m in the second linkage group. The lz locus affects the differentiation of the crystal cell, the type of hemocyte that carries prophenoloxidase(s) in paracrystalline form. All

Loss of Drosophila UBE3A phenocopies Piezo dysfunction and drives hyperphagic feeding in Drosophila.

Angelman syndrome (AS) is a rare neurogenetic disorder characterized by developmental delay, speech impairment, ataxia, epilepsy, and in some cases hyperphagic feeding behavior. AS is caused by loss of function mutations, loss of expression, or maternal allele deletion of the E3 ubiquitin ligase UBE3A. Recent work has identified a connection between UBE3A and the mechanosensitive ion channel PIEZO2, raising the possibility that UBE3A may regulate PIEZO-dependent satiety signaling. In this study, we investigated the role of the Drosophila UBE3A ortholog, Dube3a, in Piezo-associated feeding beha

Baseline susceptibility of spotted wing Drosophila (Drosophila suzukii) to four key insecticide classes.

BACKGROUND: The invasive drosophilid pest, Drosophila suzukii Matsumura, is affecting berry production in most fruit-producing regions of the world. Chemical control is the dominant management approach, creating concern for insecticide resistance in this pest. We compared the insecticide susceptibility of D. suzukii populations collected from conventional, organic or insecticide-free blueberry sites. RESULTS: The sensitivity of D. suzukii to malathion and spinetoram declined slightly across the 3 years of monitoring, whereas it was more consistent for methomyl and zeta-cypermethrin. The sensi

An exploratory analysis of the effects of human FYN and its Drosophila ortholog in Drosophila tissues.

Many cancers are initiated by the activation of oncogenes, which cause cancer phenotypes through a variety of disruptive mechanisms. FYN is a human proto-oncogene implicated in many cancers, especially those of the central nervous system and ovary. FYN encodes a non-receptor tyrosine kinase from the Src tyrosine kinase family, and has been shown to promote proliferation and metastasis. Our previous publication demonstrated that introduction of human FYN in the Drosophila eye resulted in a deleterious effect on eye structure, suggesting potential conserved functions across species. This study u

Latitudinal Clines in Diapause Onset and Circadian Clock Characteristics in Drosophila littoralis and Drosophila montana.

Drosophila littoralis and Drosophila montana are two widespread species of the virilis group that occur at latitudes between approximately 40°N and 70°N. To survive the winter, they undergo a photoperiodically controlled diapause triggered by the decreasing photoperiod in late summer to autumn. Previous studies have shown that the critical photoperiod at which diapause is triggered depends on latitude. Fly populations living at high latitudes have a longer critical photoperiod than populations living at low latitudes, meaning that they enter diapause earlier. It is not yet fully understood how
