α-Amino bisphosphonate triazoles serve as GGDPS inhibitors.

Depletion of cellular levels of geranylgeranyl diphosphate by inhibition of the enzyme geranylgeranyl diphosphate synthase (GGDPS) is a potential strategy for disruption of protein transport by limiting the geranylgeranylation of the Rab proteins that regulate intracellular trafficking. As such, there is interest in the development of GGDPS inhibitors for the treatment of malignancies characterized by abnormal protein production, including multiple myeloma. Our previous work has explored the structure-function relationship of a series of isoprenoid triazole bisphosphonate-based GGDPS inhibitor

Renaissance of farnesyltransferase inhibitors in cancer.

BACKGROUND: Prenylation (geranylgeranylation or farnesylation) plays an important role in the regulation of RAS proteins. This observation led to development of farnesyltransferase inhibitors (FTI) which were extensively tested in clinical studies of various cancer types. Unfortunately, these studies failed and the development of these drugs stalled. MAIN BODY: The renaissance of FTIs started with the approval of lonafarnib in children's progeria syndromes and continued with the approval of tipifarnib in HRAS-mutant head and neck cancers. Although there are several trials running on various v

Targeting farnesyltransferase for cancer therapy and beyond.

Farnesyltransferase (FTase) mediates Ras prenylation, a crucial activation step. Historically, farnesyltransferase inhibitors (FTIs) failed in Ras-driven cancers because of geranylgeranyltransferase I (GGTase I)-mediated alternative prenylation of Ras. Recent advances have revitalized next-generation FTIs, developed via structure-guided design, phenotypic screening, or computational approaches, offering improved selectivity, dual FTase/GGTase I targeting, or enhanced peptidomimetic affinity to overcome resistance. Combinations with targeted agents or immunotherapies exert synergistic antitumor

Inhibition of geranylgeranyl diphosphate synthase induces apoptosis through multiple mechanisms and displays synergy with inhibition of other isoprenoid biosynthetic enzymes.

Inhibitors of isoprenoid synthesis are widely used for treatment of human diseases, including hypercholesterolemia and osteoporosis, and they have the potential to be useful for treatment of cancer. Statin drugs inhibit the enzyme HMG-CoA reductase, whereas nitrogenous bisphosphonates have more recently been shown to inhibit farnesyl disphosphate synthase. In addition, our laboratory has recently developed several potent and specific bisphosphonate inhibitors of geranylgeranyl diphosphate synthase, including digeranyl bisphosphonate. Because all three enzymes fall in the same biosynthetic path

The precursor paradox for monoterpene biosynthesis in angiosperms.

Monoterpenes are among the most widespread specialized metabolites in flowering plants and the main constituents of essential oils and aromas used in the food, cosmetic, and pharmaceutical industries. Their synthesis requires geranyl diphosphate (GPP), yet a dedicated GPP synthase (GPPS) is rare in angiosperms, though the geranylgeranyl diphosphate synthases (GGPPSs) that make C20 GGPP are universally conserved. How, then, is GPP supplied? Here we review four solutions: heterodimeric GGPPS/small-subunit (SSU) complexes; Leu-Val to Val-Ala substitution in the GGPPS active site; recruitment of G

Incorporation of an alpha-hydroxy substituent modulates activity of triazole bisphosphonate-based geranylgeranyl diphosphate synthase inhibitors.

Geranylgeranyl diphosphate synthase (GGDPS), the enzyme responsible for producing the isoprenoid substrate used in protein geranylgeranylation reactions, represents a novel therapeutic target in cancer, including malignancies such as multiple myeloma and osteosarcoma. Our work has focused on the development of a series of isoprenoid triazole bisphosphonate-based GGDPS inhibitors. Previous work has revealed that our lead GGDPS inhibitors do achieve substantial systemic distribution, in contrast to clinically used nitrogenous bisphosphonates which accumulate primarily in the bone. We have determ

Regulation of the Notch-ATM-abl axis by geranylgeranyl diphosphate synthase inhibition.

Notch proteins drive oncogenesis of many cancers, most prominently T-cell acute lymphoblastic leukemia (T-ALL). Because geranylgeranylated Rab proteins regulate Notch processing, we hypothesized that inhibition of geranylgeranyl diphosphate synthase (GGDPS) would impair Notch processing and reduce viability of T-ALL cells that express Notch. Here, we show that GGDPS inhibition reduces Notch1 expression and impairs the proliferation of T-ALL cells. GGDPS inhibition also reduces Rab7 membrane association and depletes Notch1 mRNA. GGDPS inhibition increases phosphorylation of histone H2A.X, and i

Pegylation of phenothiazine - A synthetic route towards potent anticancer drugs.

INTRODUCTION: Cancer is a big challenge of the 21 century, whose defeat requires efficient antitumor drugs. OBJECTIVES: The paper aims to investigate the synergistic effect of two structural building blocks, phenothiazine and poly(ethylene glycol), towards efficient antitumor drugs. METHODS: Two PEGylated phenothiazine derivatives were synthetized by attaching poly(ethylene glycol) of 550 Da to the nitrogen atom of phenothiazine by ether or ester linkage. Their antitumor activity has been investigated on five human tumour lines and a mouse tumor line as well, by determination of IC50. The in

Prenyl Transferases Regulate Secretory Protein Sorting and Parasite Morphology in Toxoplasma gondii.

Protein prenylation is an important protein modification that is responsible for diverse physiological activities in eukaryotic cells. This modification is generally catalyzed by three types of prenyl transferases, which include farnesyl transferase (FT), geranylgeranyl transferase (GGT-1) and Rab geranylgeranyl transferase (GGT-2). Studies in malaria parasites showed that these parasites contain prenylated proteins, which are proposed to play multiple functions in parasites. However, the prenyl transferases have not been functionally characterized in parasites of subphylum Apicomplexa. Here,

The Consideration of Pseudoxanthoma Elasticum as a Progeria Syndrome.

BACKGROUND: Pseudoxanthoma elasticum (PXE) is a rare autosomal recessive disorder caused by mutations in the ATP-binding cassette sub-family C member 6 (ABCC6) gene. Patients with PXE show molecular and clinical characteristics of known premature aging syndromes, such as Hutchinson-Gilford progeria syndrome (HGPS). Nevertheless, PXE has only barely been discussed against the background of premature aging, although a detailed characterization of aging processes in PXE could contribute to a better understanding of its pathogenesis. Thus, this study was performed to evaluate whether relevant fact

Farnesyltransferase Deficiency in Cardiomyocytes Initiates Senescence and Contributes to Cardiac Fibrosis.

Cardiomyocyte senescence contributes to cardiac fibrosis, yet the molecular mechanisms remain unclear. Farnesylation is a post-translational modification critical for cholesterol metabolism and is mediated by the farnesyltransferase beta subunit (FNTB). However, its specific role in cardiomyocyte senescence and cardiac fibrosis remains unclear. Cardiomyocyte-specific Fntb knockout mice were generated to assess cardiac remodeling. RNA sequencing, DNA damage assays, and senescence markers identified molecular pathways. Mechanistic studies included nuclear envelope ultrastructure analysis, laminA

Genome-Wide Analysis of the GGPS Gene Family in Phoebe zhennan and Analysis of the Role of PzGGPS12 in Terpenoid Biosynthesis.

Geranylgeranyl diphosphate synthase (GGPS) is a key enzyme in terpenoid biosynthesis, but its role in Phoebe zhennan remains largely unknown. Here, 14 PzGGPS genes were identified and classified into three groups with conserved gene structures and motifs. Among them, PzGGPS12 was identified as a key gene for terpenoid biosynthesis, wood fragrance formation, and drought tolerance. Functional analyses showed that PzGGPS12 catalyzes the production of terpene precursors and promotes the accumulation of mono-, sesqui-, di-, and triterpenoids, thereby enhancing drought resistance through metabolic f

Effect of Farnesyltransferase Inhibitor R115777 on Mitochondria of Plasmodium falciparum.

The parasite Plasmodium falciparum causes severe malaria and is the most dangerous to humans. However, it exhibits resistance to their drugs. Farnesyltransferase has been identified in pathogenic protozoa of the genera Plasmodium and the target of farnesyltransferase includes Ras family. Therefore, the inhibition of farnesyltransferase has been suggested as a new strategy for the treatment of malaria. However, the exact functional mechanism of this agent is still unknown. In addition, the effect of farnesyltransferase inhibitor (FTIs) on mitochondrial level of malaria parasites is not fully un

Cancer therapy. New strategies and treatment modalities for optimizing patient outcomes.

In the postgenome era, development of novel targeted therapeutics is anticipated to accelerate, with the promise of specifically tailored strategies to treat specific molecularly characterized disease entities. Greater understanding of disease pathophysiology and pharmacologic actions at the molecular, cellular, and tissue levels have provided the basis for expanding the clinical application of available therapies such as recombinant human erythropoietin (r-HuEPO, epoetin alfa). The role of epoetin alfa in anemic cancer patients receiving chemotherapy has grown as our understanding of the rela

Expanding the phenotypic and genotypic spectrum of GGPS1 related congenital muscular dystrophy.

Congenital muscular dystrophies are a group of progressive disorders with wide range of symptoms associated with diverse cellular mechanisms. Recently, biallelic variants in GGPS1 were linked to a distinct autosomal recessive form of muscular dystrophy associated with hearing loss and ovarian insufficiency. In this report, we present a case of a young patient with a homozygous variant in GGPS1. The patient presented with only proximal muscle weakness, and elevated liver transaminases with spared hearing function. The hepatic involvement in this patient caused by a novel deleterious variant in

Targeting protein geranylgeranylation slows tumor development in a murine model of prostate cancer metastasis.

The isoprenoid biosynthetic pathway (IBP) plays a critical role in providing substrates and enzymes necessary for the post-translational modification and thus activation of a number of proteins involved in prostate cancer metastasis. Previous work by our lab found novel compound disodium [(6Z,11E,15E)-9-[bis(sodiooxy)phosphoryl]-17-hydroxy-2,6,12,16-tetramethyheptadeca-2,6,11,15-tetraen-9-yl]phosphonate (GGOHBP), which inhibits the IBP enzyme geranylgeranyl diphosphate synthase (GGDPS), reduced protein geranylgeranylation without altering protein farnesylation. A farnesyltransferase inhibitor improves disease phenotypes in mice with a Hutchinson-Gilford progeria syndrome mutation.

Hutchinson-Gilford progeria syndrome (HGPS) is caused by the production of a truncated prelamin A, called progerin, which is farnesylated at its carboxyl terminus. Progerin is targeted to the nuclear envelope and causes misshapen nuclei. Protein farnesyltransferase inhibitors (FTI) mislocalize progerin away from the nuclear envelope and reduce the frequency of misshapen nuclei. To determine whether an FTI would ameliorate disease phenotypes in vivo, we created gene-targeted mice with an HGPS mutation (LmnaHG/+) and then examined the effect of an FTI on disease phenotypes. LmnaHG/+ mice exhibit

GGPS1-associated muscular dystrophy with and without hearing loss.

Ultra-rare biallelic pathogenic variants in geranylgeranyl diphosphate synthase 1 (GGPS1) have recently been associated with muscular dystrophy/hearing loss/ovarian insufficiency syndrome. Here, we describe 11 affected individuals from four unpublished families with ultra-rare missense variants in GGPS1 and provide follow-up details from a previously reported family. Our cohort replicated most of the previously described clinical features of GGPS1 deficiency; however, hearing loss was present in only 46% of the individuals. This report consolidates the disease-causing role of biallelic variant
