Fetal proteins in ataxia-telangiectasia.

Ataxia-telangiectasia (AT) is a genetic disorder of unknown pathogenesis, with primary effects on the immune and nervous systems. The presence of a fetal-like thymus and elevated alpha-fetoprotein (alpha FP) levels in patients with AT suggests that suppressed mesodermal development may be a factor in the development of this disease. We investigated this hypothesis by using electrophoretic and quantitative analyses to test for the presence of other fetal proteins in mesodermal tissues. With the exceptions of alpha FP and carcinoembryonic antigen, all other proteins assessed in these patients we

Screening circulating proteins to identify biomarkers of fetal macrosomia.

OBJECTIVE: Fetal macrosomia is a major risk factor for shoulder dystocia, which can lead to birth asphyxia, maternal and neonatal traumatic injuries, and perinatal death. If macrosomia is diagnosed in the antenatal period, labour can be induced to decrease shoulder dystocia. But current clinical methods to diagnose fetal macrosomia antenatally perform with poor accuracy. Therefore, improved methods to accurately diagnose fetal macrosomia are required. Blood biomarkers that predict fetal macrosomia could be one such novel diagnostic strategy. We undertook a nested case-control study from a pros

Fetal proteins and chronic treatment with low-dose erythropoietin.

The potential stimulating effect of erythropoietin on the production of fetal proteins (FPs) has not been explored in human subjects. Therefore, the plasma levels of fetal fibrinogen (FF), carcinoembriogenic antigen (CEA), alpha-fetoprotein (AFP), and fetal hemoglobin (HbF) were measured in 12 uremic hemodialyzed patients before the first administration and after 1, 2, and 3 months of low-dose erythropoietin (r-Hu-EPO; 45 U/kg body wt I.V., thrice weekly). Such a treatment efficaciously increased total hemoglobin (Hb). CEA and AFP increased from 5.8 +/- 1.1 ng/ml and 2.9 +/- 0.9 ng/ml to the f

Derangement of hypothetical proteins in fetal Down's syndrome brain.

The success of the Human Genome Project (HGP) enables prediction of proteins by computer programs from nucleic acid sequences and for which there is no experimental evidence. Clues for function of hypothetical proteins are provided by sequence similarity with proteins of known function in model organisms. The availability of this bulk of new data is of immediate importance to Down's syndrome (DS) research. DS is the most common human chromosomal abnormality caused by an extra copy of chromosome 21 and is characterized by somatic anomalies and mental retardation. In addition, overexpression of

[The place of beta-fetoproteins among other fetal proteins].

Beta1-Fetoprotein (Fbeta1) and fetal hemoglobin (HbF) were compared with the help of immunodiffusion methods. They were shown to be non-identical although Fbeta1, as well as HbF, is stained by benzidine. Antisera against Fbeta1 do not form the line of precipitation with HbF. Besides, we succeeded to differentiate Fbeta1 from Fbeta2 (they are separated under chromatography on sephadex G-200). Fbeta1 differs from Fbeta2 by benzidine staining: Fbeta1 is benzidine-positive and Fbeta2 is benzidine-negative. The constant parallelism in the appearance of Fbeta2 and a monomeric subunit IgMs of the imm

IFITM proteins inhibit placental syncytiotrophoblast formation and promote fetal demise.

Elevated levels of type I interferon (IFN) during pregnancy are associated with intrauterine growth retardation, preterm birth, and fetal demise through mechanisms that are not well understood. A critical step of placental development is the fusion of trophoblast cells into a multinucleated syncytiotrophoblast (ST) layer. Fusion is mediated by syncytins, proteins deriving from ancestral endogenous retroviral envelopes. Using cultures of human trophoblasts or mouse cells, we show that IFN-induced transmembrane proteins (IFITMs), a family of restriction factors blocking the entry step of many vi

Expression of extracellular matrix proteins in the fetal rat gastric mucosa.

At gestational day 16 the epithelium of the rat stomach consists of a stratified layer of undifferentiated cells, and two days later glandular structures appear. The present study was carried out to identify extracellular matrix proteins that could be involved in the epithelial cell proliferation and differentiation processes that occur in the fetal rat stomach during this period. For comparative purposes the expression of the same components in the adult gastric mucosa was examined. Pregnant Sprague-Dawley rats received an intraperitoneal injection of 5-bromo-2'-deoxyuridine to label prolifer

Retinoic acid induction of stress proteins in fetal mouse limb buds.

Retinoic acid (RA) is teratogenic in rodent embryos. Several teratogens have been shown to induce the synthesis of a subset of heat shock proteins (stress proteins) in Drosophila. To determine if RA induces the synthesis of these proteins in rodent embryos, pregnant ICR mice were dosed with 100 mg/kg RA on Day 11 of gestation. Forelimb buds were removed from embryos 2.5 hr post-RA-treatment and nuclei were isolated, stained, and sorted from stages of the cell cycle. Nuclear proteins were extracted and analyzed by two-dimensional polyacrylamide gel electrophoresis. Nuclear proteins with molecul

Ret protein in the human fetal rectum.

A major gene for Hirschsprung's disease (HD) recently has been mapped in chromosome 10q11.2 and identified to be the RET proto-oncogene. Mutations of the RET gene have occurred in HD patients, and abnormalities of expression and function of Ret protein (a receptor tyrosine kinase, which is the product of the RET gene) have been found in their intestines. In vitro studies of the biological effects of HD mutations suggest a loss of function effect, which may be negative-dominant. However, the developmental role of the Ret protein in the organogenesis of the enteric nervous system (ENS) and its r

Review: Water channel proteins in the human placenta and fetal membranes.

It has been established that the permeability of the human placenta increases with advancing gestation. Indirect evidence has also proposed that aquaporins (AQPs) may be involved in the regulation of placental water flow but the mechanisms are poorly understood. Five AQPs have been found in the human placenta and fetal membranes [AQP1, 3, 4, 8 and 9]. However, the physiological function(s) and the regulation of these proteins remain unknown. Emerging evidence has shown that human fetal membrane AQPs may have a role in intramembranous amniotic fluid water regulation and that alterations in thei

Antimicrobial proteins and peptides in pregnancy: Guardians of the maternal-fetal frontier.

The success of human pregnancy relies on the precise synchronization and adaptation of the maternal immune system, which, together with fetal tissues, establishes a tolerogenic intrauterine environment while maintaining the capacity to mount effective immune responses. At the maternal-fetal interface, multiple physical and immune mechanisms coordinate the recognition and control of pathogens that could jeopardize pregnancy. Ascending infections from the lower genital tract can reach the uterine cavity, infect the fetal membranes (chorioamnionitis), and invade the amniotic fluid, triggering a p

[Role of maternal and fetal plasma proteins in transplacental transport of drugs].

One of the factors influencing the rate of drug transport through the placenta is their binding by maternal and fetal plasma proteins. Concentrations of the components binding the drug on both sides of the placenta influence significantly its distribution in the fetomaternal pair. It is emphasized that in plasma of a fetus or newborn there is observed the free fraction increase for most drugs--beta-adrenoblockers, local anesthetics, narcotic analgesics, antiarrthythmic and antiepileptic agents. As for salicylates, diazepam and valproic acid, one can note their accumulation in the fetal blood d

Assay of a placental protein to determine fetal risk.

Plasma concentrations and total amounts of pregnancy associated plasma protein A were determined in 272 patients at 34 weeks' gestation by immunoelectrophoretic assay. The mean plasma concentration and mean total amount of this protein were closely related (r = 0.9643) and were significantly raised in patients who subsequently developed pre-eclampsia (28 patients), went into premature labour (12), or suffered from antepartum haemorrhage (10). Mean values in all patients delivering growth-retarded babies were also raised, but when the results for such patients who also had other complications w

Association of proteins involved in fetal lung development with respiratory outcomes in infancy.

OBJECTIVE: We aimed to assess whether proteins which are involved in fetal lung development but also have an additional role in tissue or airway remodeling and oxidative stress response are associated with lung function and respiratory symptoms in infants. STUDY DESIGN: We performed hierarchical clustering to determine distinct profiles of nine cord blood proteins (growth factors, metalloproteinases, and autophagy markers) in 369 healthy infants born to non-asthmatic mothers from the Basel-Bern Infant Lung Development cohort (BILD). Using linear and negative binomial regression analysis, we e

Measurements of placental, decidual, and fetal proteins before and after chorionic villus sampling.

Circulating placental [human chorionic gonadotrophin (hCG), Schwangerschafts protein 1 (SP1), pregnancy-associated plasma protein A (PAPP-A), decidual (pregnancy protein 12 (PP12), and fetal alphafetoprotein (AFP)] proteins were measured immediately before and within 1 h in 18 women undergoing diagnostic chorionic villus sampling (CVS) in the first trimester. An elevation of serum AFP levels was consistently seen, while fluctuations in excess of 10 per cent of the pre-CVS levels of SP1 and PP12 were seen in the majority of patients. Fluctuations in hCG and PAPP-A were consistently less than 10

Uterine secretome: What do the proteins say about maternal-fetal communication in buffaloes?

The aim was to compare the UF proteomics of pregnant and non-pregnant buffalo during early pregnancy. Forty-four females were submitted to hormonal estrus synchronization and randomly divided into two groups: pregnant (n = 30) and non-pregnant (n = 14). The pregnant group was artificially inseminated and divided into a further two groups: P12 (n = 15) and P18 (n = 15). Conceptus and uterine fluid samples were collected during slaughter at, respectively, 12 and 18 days after insemination. Of all the inseminated females, only eight animals in each group were pregnant, which reduced the sample of
