Biochemical characterization of the chloroplastic β-carbonic anhydrase from Flaveria bidentis (L.) "Kuntze".

C3 and C4 plant carbonic anhydrases (CAs) are zinc-enzymes that catalyze the reversible hydration of CO2. They are sub-divided in three classes: α, β and γ, being distributed between both photosynthetic subtypes. The C4 dicotyledon species Flaveria bidentis (L.) "Kuntze" contains a small gene family encoding three distinct β-CAs, named FbiCA1, FbiCA2 and FbiCA3. We have expressed and purified recombinant FbiCA1, which is localized in the chloroplast where it is thought to play a role in lipid biosynthesis and antioxidant activity, and biochemically characterized it by spectroscopic and inhibit

Differential response of photosynthetic light reactions to phosphate starvation in C3 and C4  Flaveria.

C4 photosynthesis is a CO2-concentration mechanism that separates CO2 fixation between two cell types, thereby reducing photorespiration and making C4 plants more efficient than their C3 counterparts. While the C4 cycle has evolved multiple times across different genera, this study evaluates very closely related C3 and C4 species within the genus Flaveria. Apart from their carbon metabolism, C4 plants also possess adaptations in their mineral nutrition. One key nutrient which is also directly involved in photosynthesis is phosphorus. It is absorbed by the plant in the form of inorganic phospha

The effects of allelochemicals from root exudates of Flaveria bidentis on two Bacillus species.

To determine the allelopathic effects of root exudates from Flaveria bidentis on function of Bacillus, pot experiment was used to collect root exudates from living plants and test its allelopathic effects on function of Bacillus frigoritolerans and Bacillus megaterium, which were two dominant bacteria in the rhizosphere soil of F. bidentis. To obtain the allelopathic substances, the root exudates were successively extracted by N-hexane, dichloromethane, ethyl acetate, and N-butanol, and their allelopathic effects were tested. The results showed that B. frigoritolerans and B. megaterium conside

Quercetin 3,7,3',4'-tetrasulphated isolated from Flaveria bidentis inhibits tissue factor expression in human monocyte.

Sulphated esters of the flavonoids sulphated quercetin 3,7,3',4'-tetrasulphated (QTS) and quercetin 3-acetyl-7,3,4'-trisulphate (ATS), isolated from Flaveria bidentis, have demonstrated anticoagulant and antiplatelet properties. In this study, we examined if both compounds affected the expression of the procoagulant tissue factor (TF) induced by lipopolysaccharide (LPS) on human monocyte. Monocytes were pretreated with different concentrations of each flavonoid (0.1-500 μM), followed by a 4h incubation with LPS in order to induce TF expression. Results of the TF expression showed different beh

Inducibility of cyclic electron transport is linked to the transition from C3-C4 to C4 photosynthesis in Flaveria.

The evolutionary convergence of complex biological features offers valuable insights into the interplay among environmental factors, organismal traits, and evolutionary outcomes. C4 photosynthesis exemplifies an adaptive syndrome derived from ancestral C3 photosynthesis in response to environmental stresses. In this study, we investigated the inducibility of C4 photosynthetic characteristics in various Flaveria species, including one of the youngest C4 species on Earth, under low CO2 conditions (100 ppm). Species used include F. robusta (proto-Kranz type), F. sonorensis (type I C3-C4), F. line

Antisense reduction in NADP-ME in the C4 species Flaveria bidentis alters stomatal sensitivity to intercellular [CO2].

Stomata have the crucial role of balancing the uptake of CO2 for photosynthesis and water loss via transpiration by adjusting their aperture. In C3 plants, coordination between photosynthesis and stomatal conductance can be disrupted by mutations in Calvin-Benson-Bassham (CBB) cycle expression, but it is not clear whether the disruption of the C4 carbon concentrating mechanism would have similar effects. In this study, it was hypothesized that perturbing the C4 cycle, resulting in reduced carbon flux into the bundle sheath cells and overall net CO2 assimilation rates, may alter stomatal regula

Kinetic and anion inhibition studies of a β-carbonic anhydrase (FbiCA 1) from the C4 plant Flaveria bidentis.

Several β-carbonic anhydrases (CAs, EC 4.2.1.1) are present in all land plants examined thus far. Here we report the first detailed biochemical characterization of one such isoform, FbiCA 1, from the C4 plant Flaveria bidentis, which was cloned, purified and characterized as recombinant protein. FbiCA 1 has an interesting CO2 hydrase catalytic activity (kcat of 1.2×10(5) and kcat/Km of 7.5×10(6)M(-1)×s(-1)) and was moderately inhibited by most simple/complex inorganic anions. Potent FbiCA 1 inhibitors were also detected, such as trithiocarbonate, diethyldithiocarbamate, sulfamide, sulfamic aci

From Gas Chromatography-Mass Spectrometry (GC-MS) to Network Pharmacology: System-Level Insights into the Multi-Target Biological Potential of Flaveria trinervia (Spreng.) C. Mohr.

Flaveria trinervia (Spreng) C. Mohr is a plant traditionally used in Mexican medicine. In this study, gas chromatography-mass spectrometry (GC-MS) combined with network pharmacology was employed to characterize volatile and semi-volatile metabolites from F. trinervia leaves and to explore their potential system-level mechanisms of action in inflammatory and tumor-related disorders. A dual extraction strategy (hexane/dichloromethane and acetone/chloroform) was applied, followed by GC-MS-based compound identification. Putative molecular targets were predicted using established pharmacological da

Faster relaxation of nonphotochemical quenching in C4 than in C3 species.

Acceleration of photoprotective nonphotochemical quenching (NPQ) responses to changes in light intensity has been suggested as a strategy to enhance crop yield. Our current understanding of NPQ overwhelmingly comes from C3 species, and despite many key crops being C4, separating the impact of photosynthetic pathway from species-specific effects is complicated by the large evolutionary distance between most C3 and C4 species. By conducting a series of experiments on C3 and C4 species from Alloteropsis, Flaveria, and Cleome, genera which encompass both pathways, we controlled for phylogenetic di

Single and double overexpression of C(4)-cycle genes had differential effects on the pattern of endogenous enzymes, attenuation of photorespiration and on contents of UV protectants in transgenic potato and tobacco plants.

To improve the efficiency of CO(2) fixation in C(3) photosynthesis, C(4)-cycle genes were overexpressed in potato and tobacco plants either individually or in combination. Overexpression of the phosphoenolpyruvate carboxylase (PEPC) gene (ppc) from Corynebacterium glutamicum (cppc) or from potato (stppc, deprived of the phosphorylation site) in potato resulted in a 3-6-fold induction of endogenous cytosolic NADP malic enzyme (ME) and an increase in the activities of NAD-ME (3-fold), NADP isocitrate dehydrogenase (ICDH), pyruvate kinase (PK), NADP glycerate-3-P dehydrogenase (NADP-GAPDH), and P
