Rhabdoid phenotypic change has been described in a number of different neoplasms from diverse organ sites. These tumors share common light and electron-microscopic features, display a polyphenotypic immunohistochemical profile and often show cytogenetic abnormalities of chromosome 22. In the central nervous system (CNS), most rhabdoid tumors occur in the posterior fossa of very young children and are associated with a primitive neuroectodermal tumor (PNET) component and are designated atypical teratoid/rhabdoid tumors. Infrequently, other rhabdoid tumors of the CNS have been described, includi

Glioblastoma multiforme is the most common primary brain tumor of adults, as well as the most malignant. Its etiology is unknown, but the tumor is thought to arise through dedifferentiation of adult astrocytes. It occurs most frequently between the ages of 40 and 60, in men more often than in women (1.5:1). Important early symptoms include subtle personality change, headache, weakness, and intellectual impairment; specific complaints and physical findings depend on the location of the lesion. The initial diagnostic test should be a CT-scan; it will detect more than 90% of malignant astrocytoma

MicroRNAs represent an abundant class of endogenously expressed 18-25 nucleotide non-coding RNA molecules that function to silence gene expression through a process of post-transcriptional modification. They exhibit varied and widespread functions during normal development and tissue homeostasis, and accordingly their dysregulation plays major roles in many cancer types. Gliomas are cancers arising from the central nervous system. The most malignant and common glioma is glioblastoma multiforme (GBM), and even with aggressive treatment (surgical resection, chemotherapy, and radiation), average

For glioblastoma (GBM), therapeutic resistance shows a formidable obstacle to effective treatment. Recent insights into cellular plasticity as a novel paradigm for understanding this resistance suggest that tumor cells undergo phenotypic and molecular alterations during GBM progression, collectively termed cellular plasticity. These alterations arise from complex tumor microenvironment (TME) factors, epigenetic changes, and selective pressures induced by radiotherapy or temozolomide (TMZ), ultimately leading to significant tumor heterogeneity and therapeutic resistance. This review will examin

This study is a histological and clinical investigation of four cases of cerebellar glioblastoma, a rare tumor. The cases included three males and one female, from 33 to 67 years in age (mean 49 years). Tumor resection, postoperative irradiation and chemotherapy were performed in all cases. Two patients died of local tumor recurrence after 14 and 27 months. Another patient relapsed after 10 months; however, after additional tumor resection and second line chemotherapy, she remains disease-free 41 months after the initial treatment. The fourth patient has not relapsed in the 6 months since init

Glioblastoma (GBM) is the most common primary malignancy of the brain with a poor prognosis. Despite advances in therapeutics, the gold standard of treatment remains maximal safe resection followed by temozolomide and radiation therapy, with the inter- and intratumoral heterogeneity of GBM making effective management challenging. As a result, the search for mechanisms underlying this heterogeneity has been paramount. The immunosuppressive and anti-inflammatory microenvironment of GBM has been of particular interest in recent years. Inflammasomes are protein complexes responsible for initiation

Glioblastoma (GBM), the most frequent and malignant primary brain tumor, is characterized by a highly diverse and profoundly immunosuppressive tumor microenvironment (TME) that provides an unconstrained environment for tumor progression and significantly complicates therapeutic interventions. Despite advances in immunotherapeutic approaches, such as chimeric antigen receptor T cell and immune checkpoint inhibitors, efficacy remains limited due to the complexity of the GBM TME and robust immune evasion mechanisms. In this review, we elucidate the intricate interplay among cellular components wi

BACKGROUND AND IMPORTANCE: Although glioblastoma is the most common primary brain tumor, primary intraventricular locations are extremely rare; only 21 cases have been reported to date.
METHODS: A retrospectively acquired database of all intracranial glioblastomas treated in 2 different neurosurgical departments during the last 10 years was queried. Patients with histologically proven intraventricular glioblastomas were included in the study.
RESULTS: Eight patients were identified as having a histologically confirmed intraventricular glioblastoma. Patient age at diagnosis ranged from 6 to 7

Glioblastomas (GB) are highly aggressive brain tumors with poor patient prognosis and low survival rates. To identify novel therapeutic targets, the tumor microenvironment (TME) is increasingly examined, with a particular focus on biomechanical changes in the extracellular matrix (ECM) that contribute to GB aggressiveness. In GB, the ECM stiffens, regulating cell behavior through mechanotransduction. Preclinical in vitro and ex vivo studies generally report increased stiffness in GB relative to healthy brain tissue, whereas clinical in vivo measurements often report decreased stiffness. This r

Glioblastoma multiforme (GBM) is the most common and deadliest form of primary brain tumor. Despite treatment with surgery, radiotherapy, and chemotherapy with the drug temozolomide, the expected survival after diagnosis remains low. The median survival is only 14.6 months and the two-year survival is a mere 30%. One reason for this is the heterogeneity of GBM including the presence of glioblastoma cancer stem cells (GSCs). GSCs are a subset of cells with the unique ability to proliferate, differentiate, and create tumors. GSCs are resistant to chemotherapy and radiation and thought to play an

The clinicopathologic and molecular genetic features of 5 cases of rhabdoid glioblastoma, an extremely rare variant of glioblastoma that tends to affect patients at a young age, were investigated by immunohistochemical analysis and focused molecular genetic studies including array-based comparative genomic hybridization. All 5 cases had supratentorial tumors that immunohistochemical analysis revealed to be robustly positive for epithelial membrane antigen, vimentin, p53, and PDGFRα (platelet-derived growth factor receptor, alpha polypeptide) but only focally positive for glial fibrillary acidi

PURPOSE OF REVIEW: Reirradiation has emerged as a potentially valuable treatment strategy for recurrent glioblastoma, a disease characterized by inevitable local progression despite aggressive multimodal first-line therapy. Recent advances in radiotherapy techniques, improved patient selection, and evolving systemic treatment combinations have renewed clinical interest in this approach. This is reflected by recent publication of the first international consensus guidelines (ESTRO/EANO) and the initiation of an European phase III randomized trial on reirradiation of patients with recurrent glio

Cryptococcus neoformans and C. gattii cause invasive fungal disease, with meningitis being the most common manifestation of central nervous system (CNS) disease. Encapsulated cryptococcomas occur rarely, predominantly in immunocompetent hosts, usually related to C. gattii Our patient was an immunocompetent man who presented with headache and a large cystic CNS lesion thought to be glioblastoma. Biopsy of a concomitant lung lesion confirmed cryptococcoma and empiric antifungal therapy was started for presumed CNS cryptococcoma. Antifungal therapy failed to shrink the CNS lesion, and surgical ex

INTRODUCTION: Peptides occupy a strategic position between small molecules and biologics, offering high target specificity, favourable safety profiles, and chemical tractability. In musculoskeletal medicine, they address diverse pathological processes-including inflammation, degeneration, infection, oncology, and fibrosis-through mechanisms that conventional therapeutics cannot replicate. Two principal frameworks underpin this field. Throughout this review, we differentiate bioactive peptides, defined as naturally occurring or food-derived sequences that exert physiological effects, from thera

Spinal glioblastoma multiforme is a rare entity comprising 1.5% of all spinal cord tumors. We report a 57-year-old man presenting with a 1.5 month history of left sided radiculopathy, lower cranial nerve weakness and difficulty in walking. MRI of the brain and spine showed an exophytic intramedullary lesion extending from the cervicomedullary junction to the lower margin of C4. To our knowledge, we report the first patient with dorsally exophytic cervicomedullary and cervical intramedullary glioblastoma multiforme.

Extracranial metastases arising from primary brain tumors are very unusual. We report a case of glioblastoma with intra-thoracic spread characterized by mediastinal lymph nodes metastasis and bronchial, lung parenchyma and pleura carcinomatous lymphangitis. Pathologic findings are consistent with non-differentiated cell proliferation but Glial Fibrillary Acidic Protein immunodetection can be helpful to relate such a non-differentiated cell proliferation to its glial origin. The mechanisms of extra-cerebral spread are discussed according to autopsic findings and to the data reported in the lite

Clinical & translational oncology : official publication of the Federation of Spanish Oncology Societies and of the National Cancer Institute of Mexico2026eng
Glioblastoma is the most aggressive and common malignant primary brain tumor among adults. Due to its aggressiveness, GBM entails a short median overall survival and fatal outcomes. Incidence notably increases with age, 50% of patients being diagnosed at the age of 65 or older. Given that cognitive loss is commonly confused with age-related cognitive impairment, delays in diagnosis are frequent. Furthermore, the elderly are a vulnerable group, commonly having poorer functional status, more comorbidities, and reduced tolerance to intensive treatments. Therefore, clinical and therapeutic implica
