Standardized Diagnostic Assays for Omsk Hemorrhagic Fever Virus.

Omsk hemorrhagic fever is an acute zoonotic disease caused by Omsk hemorrhagic fever virus, a member of the genus Flavivirus (family Flaviviridae), with a reported case-fatality rate of approximately 3%. Historically confined to southwestern Siberia, ecological changes raise concerns about possible spread to non-endemic regions. Although no Omsk hemorrhagic fever cases have been reported in the Republic of Korea, the risk of accidental importation highlights the importance of establishing a reliable diagnostic protocol. We established and validated an institutionally developed diagnostic proto

Inferring Evolutionary Timescale of Omsk Hemorrhagic Fever Virus.

Until 2020, there were only three original complete genome (CG) nucleotide sequences of Omsk hemorrhagic fever virus (OHFV) in GenBank. For this reason, the evolutionary rate and divergence time assessments reported in the literature were based on the E gene sequences, but notably without temporal signal evaluation, such that their reliability is unclear. As of July 2022, 47 OHFV CG sequences have been published, which enables testing of temporal signal in the data and inferring unbiased and reliable substitution rate and divergence time values. [The Omsk hemorrhagic fever: research results (1946-2013)].

The main aspects of epidemiology and epizootology of the Omsk hemorrhagic fever (OHF) are analyzed. The discovery of the virus OHF in 1947, as well as the first outbreak of new diseases in the districts of the Omsk region, is described. Comprehensive work for decryption of the etiology of the OHF by specialists from the Omsk and Moscow Institutes is carried out. Long-term dynamics of activity of natural foci of OHF contains four periods of variable intensity of epidemic and epizootic processes. The main reservoir of the virus OHF in natural foci and the source of human infection is muskrat. Me

The deduced evolution history of Omsk hemorrhagic fever virus.

Omsk hemorrhagic fever (OHF) is a severe disease that emerged in the 1940s in Siberia, Russia. It is caused by the OHF virus (OHFV), belonging to the Flavivirus genus. In wildlife, the principal vector of OHFV is the Dermacentor reticulatus tick. However, humans are mainly infected after contact with an infected muskrat Ondatra zibethicus. The evolutionary history of OHFV is not yet clarified. In an attempt to reconstruct the temporal and spatial phylodynamics of OHFV, a collection of 25 OHFV strains was studied. Maximum likelihood analysis, the Bayesian MCMC approach, and procedures based on

[The clinico-epidemiological characteristics of Omsk hemorrhagic fever in 1988-1992].

Materials on morbidity in Omsk hemorrhagic fever at the period of 1988-1992 in the districts of the Novosibirsk region are presented. Cases of the disease were registered mainly in September-October (83.3% of cases). Transmission of the disease by contact played the leading role in the epidemiology of Omsk hemorrhagic fever. The disease took the course of medium severity with faintly pronounced hemorrhagic syndrome.

An animal model for the tickborne flavivirus--Omsk hemorrhagic fever virus.

The tickborne encephalitis (TBE) serocomplex of flaviviruses consists primarily of viruses that cause neurologic disease; these viruses include Omsk hemorrhagic fever virus (OHFV), a virus that is genetically related to other TBE serocomplex viruses but that circulates in an ecologically distinct niche and causes markedly different human disease. The objective of this study was to examine a potential small-animal model for OHFV and to compare the pathology of infection with that of the neurotropic tickborne flavivirus, Powassan virus (POWV). POWV-infected BALB/c mice demonstrated typical arbov

Structure of the envelope protein domain III of Omsk hemorrhagic fever virus.

We have solved the NMR solution structure of domain III from the Omsk hemorrhagic fever virus envelope protein and report the first sequencing of the Guriev strain of this virus. Important structural differences between tick-borne flaviviruses, such as OHFV and TBE, and mosquito-borne flaviviruses, such as West Nile virus, are discussed.

[Laboratory Diagnosis of Omsk Hemorrhagic Fever Virus and Kyasanur Forest Disease Virus].

The Omsk hemorrhagic fever virus and Kyasanur Forest disease virus, which belong to the genus Orthoflavivirus in the family Flaviviridae, are tick-borne viruses. These viruses, which were previously endemic to certain countries and regions, are now at increasing risk of spreading to new areas owing to global climate change and increased human mobility. Despite advances in real-time reverse transcription-polymerase chain reaction and serological assays, there are challenges in distinguishing these viruses from other tick-borne flavivirus infections. There is still a need for accurate and rapid

Trans Complementation of Replication-defective Omsk Hemorrhagic Fever Virus for Antiviral Study.

Omsk hemorrhagic fever virus (OHFV) is a tick-borne flavivirus classified as a biosafety level-4 (BSL4) pathogen. Studies of OHFV are restricted to be conducted within BSL4 laboratories. Currently, no commercial vaccines or antiviral drugs are available against OHFV infection. In this study, we recovered a replication-deficient OHFV with an NS1 deletion (OHFV-ΔNS1) and reporter virus replacing NS1 with the Gaussia luciferase (Gluc) (OHFV-ΔNS1-Gluc). Both the defective OHFV-ΔNS1 and OHFV-ΔNS1-Gluc virus could only replicate efficiently in the BHK21 cell line expressing NS1 (BHK21NS1) but not in

Sub-genomic replicon and virus-like particles of Omsk hemorrhagic fever virus.

Omsk hemorrhagic fever virus (OHFV) is a member of the tick-borne encephalitis serocomplex of flaviviruses, and causes hemorrhagic disease in humans. To investigate the molecular mechanisms involved in OHFV pathogenesis, we constructed several subgenomic OHFV replicons containing large deletions in the structural region. Replicon RNA was introduced into BHK cells by transfection and the production of viral proteins was monitored by IFA. GFP and luciferase genes were inserted into the OHFV replicon, and these reporter genes were expressed in cells harboring replicating replicon RNA. OHFV replic

[Effectiveness of virazol, realdiron and interferon inductors in experimental Omsk hemorrhagic fever].

Efficacy of drug therapy of Omsk hemorrhagic fever (OHF) was tested experimentally using cell culture and laboratory animals infected with the OHF virus strain Ondatra. Screening tests showed that high concentrations of Virazol or interferon inducers Larifan and Rifastin caused moderately pronounced suppression of virus reproduction in cell culture. Realdiron was found to be a high-efficacy preparation causing complete inhibition of virus reproduction in cell. Larifan demonstrated the highest antiviral efficacy against OHF virus in experiments with laboratory animals. This drug prevented the d

Omsk Hemorrhagic Fever Virus: A Comprehensive Review from Epidemiology to Diagnosis and Treatment.

Omsk hemorrhagic fever virus (OHFV) is the etiological agent of a poorly studied acute viral disease, causing several epidemic waves observed in the western Siberia regions of Omsk, Kurgan, Novosibirsk, and Tyumen. OHFV is a flavivirus and shares structural and morphological features with tick-borne encephalitis (TBE) complex viruses. The disease's symptoms show high variability, from flu-like symptoms, hyperesthesia, and petechial rush in the upper body to high fever and hemorrhagic manifestations, with a fatality rate of about 1%. The real number of OHFV-infected people is still unknown due

Evaluation of the European tick-borne encephalitis vaccine against Omsk hemorrhagic fever virus.

This study focused on the antigenic cross-reactivity between tick-borne encephalitis virus (TBEV) and Omsk hemorrhagic fever virus (OHFV) to assess the efficacy of the commercial TBE vaccine against OHFV infection. Neutralization tests performed on sera from OHFV- and TBEV-infected mice showed that neutralizing antibodies are cross-protective. The geometric mean titers of antibodies against TBEV and OHFV from TBEV-infected mice were similar. However, the titers of anti-TBEV antibodies in OHFV-infected mice were significantly lower than those of anti-OHFV antibodies in the same animals. In mous

Analysis of the complete genome of the tick-borne flavivirus Omsk hemorrhagic fever virus.

Omsk hemorrhagic fever virus (OHF) is a tick-borne flavivirus endemic to Western Siberia. This virus is the only known tick-borne flavivirus to cause hemorrhagic disease in humans in the absence of encephalitis. OHF virus circulates within a small, defined niche in which other tick-borne complex flaviviruses are also present. The objectives of this study were to genetically classify OHF virus based on its complete genome and to identify genetic determinants that might be involved in tissue tropism and viral replication leading to the disease state caused by this virus. The OHF virus genome was

Omsk hemorrhagic fever virus is a tick-borne encephalitis virus adapted to muskrat through host-jumping.

Omsk hemorrhagic fever was first described in the early 1940s and is a natural focal infection, spread exclusively in four regions of Western Siberia, and associated with muskrat (Ondatra zibethicus). The etiological agent of this disease is the Omsk hemorrhagic fever virus (OHFV) which is closely related to the tick-borne encephalitis virus (TBEV), and its range entirely lies within the TBEV area. OHFV belongs to the mammalian tick-borne flaviviruses and the ecological group of arboviruses. The problem concerning the origin of OHFV remains unresolved to date. This study analyzed all nucleotid
