The efficacy of alpha-ketoglutaric acid in the antagonism of cyanide intoxication.

It has been reported that compounds containing carbonyl groups can readily react with cyanide. Pyruvic acid, an alpha-ketocarboxylic acid, has been shown to antagonize the lethal effects of cyanide. It is suggested that its mechanism of action rests in its ability to react with or "bind" cyanide. In this study, alpha-ketoglutaric acid, also an alpha-ketocarboxylic acid, was evaluated for its ability to counteract the lethal effects of cyanide. alpha-Ketoglutaric acid increased the LD50 value of cyanide (6.7 mg/kg) by a factor of five, a value statistically equivalent to that ascertained in mic

Mechanism of antagonizing cyanide-induced lethality by alpha-ketoglutaric acid.

alpha-Ketoglutaric acid (alpha-KG) has been shown to be an effective antagonist for cyanide-induced lethality. The mechanism of this antagonism is hypothesized to result from alpha-KG binding with cyanide. Several investigative approaches were taken to determine the existence of this binding. First, mixtures of various molar ratios of alpha-KG:cyanide were injected into a high pressure liquid chromatograph. The addition of cyanide reduced the peak area of alpha-KG at a molar ratio of greater than 1:5. Second, blood from naive male ICR mice was spiked with alpha-KG and cyanide. Headspace above

Antidotal efficacy of alpha-ketoglutaric acid and sodium thiosulfate in cyanide poisoning.

Alpha-ketoglutaric acid and sodium thiosulfate antagonize the toxic effects of cyanide. The present study was performed to test whether a synergistic effect may occur. The alpha-ketoglutaric acid/sodium thiosulfate solutions were injected intraperitoneally into mice prior to exposure to hydrogen cyanide (HCN) in a dynamic inhalation chamber or preceding an intraperitoneal injection of sodium cyanide (NaCN). All lethal concentration (LCT) and lethal dose (LD) values were determined after a period of 24 h. Alpha-ketoglutaric acid alone provided no protection at 250 mg/kg when challenged with HCN

Rapid and efficient separation of eucalyptus hemicellulose by synergistic catalysis of acid and ketone in α-ketoglutaric acid pretreatment.

The efficient catalytic hydrolysis and mitigation of unstable structural transitions in lignin are essential for the commercialization of organic acid pretreatment. A novel binary carboxylic acid pretreatment utilizing α-ketoglutaric acid (AKA) was developed, which exploits a distinctive ion-pair biomimetic catalytic mechanism along with the activation of the ketone group. Under optimal AKA pretreatment conditions (5 % acid concentration, 150 °C, 45 min), the hemicellulose separation yield reached 85.21 %. The rapid and efficient separation of eucalyptus hemicellulose results from the low pKa

Alpha-Ketoglutarate Dehydrogenase and Citric Acid Cycle Flexibility in Heart Disease and Failure.

Tricarboxylic acid cycle (TCA) enzymes, mitochondrial isocitrate dehydrogenase (IDH2), α-ketoglutarate dehydrogenase (AKGDH), and succinyl-CoA synthetase (SCS) catalyse the conversion of isocitrate to succinate generating reducing equivalents for mitochondrial complex I and ATP by substrate level phosphorylation. Because of their central roles in energy metabolism, TCA enzymes and their metabolites distribute extensively across cellular compartments coordinating intermediary metabolism, cellular redox, and nuclear gene expression. AKGDH activity is regulated by acylation, oxidation, energy cha

Molecular mechanism of hypoxia and alpha-ketoglutaric acid on collagen expression in scleral fibroblasts.

AIM: To investigate the molecular mechanisms underlying the influence of hypoxia and alpha-ketoglutaric acid (α-KG) on scleral collagen expression. METHODS: Meta-analysis and clinical statistics were used to prove the changes in choroidal thickness (ChT) during myopia. The establishment of a hypoxic myopia model (HYP) for rabbit scleral fibroblasts through hypoxic culture and the effects of hypoxia and α-KG on collagen expression were demonstrated by Sirius red staining. Transcriptome analysis was used to verify the genes and pathways that hypoxia and α-KG affect collagen expression. Finally,

[Effects of α-ketoglutaric acid on the cryopreservation of dairy goat semen and sperm fertilization capacity].

The role of alpha-ketoglutarate (AKG), a crucial metabolic intermediate, in enhancing the cryotolerance of mammalian sperm remains poorly understood. This study systematically investigated the effects of AKG on semen cryopreservation in dairy goats and elucidated its underlying mechanisms. Semen samples were collected from 10 healthy Saanen dairy goats, pooled after screening for motility (> 80%), and cryopreserved in liquid nitrogen vapor with extender supplemented with varying AKG concentrations (0, 10, 100, and1 000 μg/mL). Post-thaw assessments included sperm motility parameters, survival

GPT2-mediated Glutamate to α-Ketoglutaric Acid Conversion in Cardiac Fibroblast Promotes Fibrosis.

Cardiac fibroblasts play a key role in heart fibrosis, but how their metabolism changes during this process is unclear. This study shows that mesenchymal stem cell-derived extracellular vesicles reduce fibrosis in mice after heart pressure overload. Under stress, fibroblasts increase mitochondrial adenosine triphosphate by boosting glutamate metabolism, especially through the enzyme GPT2, which converts glutamate to α-ketoglutarate. This leads to fibroblast activation and excess collagen. Inhibiting GPT2 via microRNA-30c-5p delivered by mesenchymal stem cell-derived extracellular vesicles -red

Inhibition of the alpha-ketoglutarate dehydrogenase complex by the myeloperoxidase products, hypochlorous acid and mono-N-chloramine.

Abstract alpha-Ketoglutarate dehydrogenase (KGDHC) complex activity is diminished in a number of neurodegenerative disorders and its diminution in Alzheimer Disease (AD) is thought to contribute to the major loss of cerebral energy metabolism that accompanies this disease. The loss of KGDHC activity appears to be predominantly due to post-translation modifications. Thiamine deficiency also results in decreased KGDHC activity and a selective neuronal loss. Recently, myeloperoxidase has been identified in the activated microglia of brains from AD patients and thiamine-deficient animals. Myeloper

Inactivation of the 2-ketoglutarate and pyruvate dehydrogenase complexes of beef heart by branched chain keto acids.

Incubation of 2-ketoglutarate dehydrogenase complex with 2-ketoisovalerate, 2-keto-4-methylvalerate, or 2-keto-3-methylvalerate leads to the appearance of a lag phase and of a progressive loss of activity in subsequent measurements of the initial rate of oxidation of 2-ketoglutarate. In the case of 2-ketoisovalerate these effects are shown to be due to the formation of an isobutyryllipoate derivative of the enzyme, as a result of the very slow oxidation of 2-ketoisovalerate by the enzyme complex (Vmax congruent to 0.15% of that for 2-ketoglutarate). Incubation of the enzyme complex with 2-keto

Cholangiocarcinoma PDHA1 succinylation suppresses macrophage antigen presentation via alpha-ketoglutaric acid accumulation.

Gemcitabine combined with cisplatin is the first-line chemotherapy for advanced cholangiocarcinoma, but drug resistance remains a challenge, leading to unsatisfactory therapeutic effect. Here, we elucidate the possibility of chemotherapy regimens sensitized by inhibiting succinylation in patients with cholangiocarcinoma from the perspective of post-translational modification. Our omics analysis reveals that succinylation of PDHA1 lysine 83, a key enzyme in the tricarboxylic acid cycle, alters PDH enzyme activity, modulates metabolic flux, and leads to alpha-ketoglutaric acid accumulation in th

Identifying the target, mechanism, and agonist of α-ketoglutaric acid in delaying mesenchymal stem cell senescence.

α-ketoglutaric acid (AKG), a tricarboxylic acid cycle metabolite central to aerobic metabolism and longevity, retains unresolved anti-aging protein targets. Here, we demonstrate that reduced isocitrate dehydrogenase 1 (IDH1) expression during senescence lowers AKG production, accelerating the aging of mesenchymal stem cells (MSCs). Exogenous AKG or IDH1 overexpression restores AKG levels, enabling 2-oxoglutarate and Fe(II)-dependent oxygenase domain-containing protein 1 (OGFOD1)-catalyzed hydroxylation of ribosomal protein S23 (RPS23) at proline 62. Mechanistically, AKG stabilizes the OGFOD1-R

Enhanced α-ketoglutaric acid production and recovery in Yarrowia lipolytica yeast by effective pH controlling.

The replacement of chemical synthesis by environmentally friendly energy-efficient technologies for production of valuable metabolites is a principal strategy of developing biotechnological industry all over the world. In the present study, we develop a method for α-ketoglutaric acid (KGA) production from rapeseed oil with the use of Yarrowia lipolytica yeast. Sixty strains of Y. lipolytica yeasts were tested for their ability to produce KGA, and the strain Y. lipolytica 212 (Y. lipolytica VKM Y-2412) was selected as a promising KGA producer. Using a three-stage pH controlling, in which pH was

[Metabolic characteristics of the Yarrowia lipolytica strain producing alpha-ketoglutaric and citric acids from ethanol and the effect of [NH4+] and [O2] on yeast respiration and biosynthesis].

The comparative studies performed in this work showed that overproduction of alpha-ketoglutaric acid (KGA) from ethanol by the mutant Yarrowia lipolytica strain I requires both a deficiency of thiamine and a relatively high concentration of ammonium ions in the medium, whereas CA overproduction requires an almost zero concentration of ammonium ions. The threshold value of the dissolved oxygen concentration in the medium, pO2, for CA overproduction is considerably higher than for KGA overproduction. The respiration rate of CA-overproducing cells was 2-3.5 times higher than that of KGA-overprodu

Insights into the alleviating effects of exogenous α-ketoglutaric acid on free nitrous acid stress in Tribonema minus and ammonium removal in wastewater.

α-Ketoglutaric acid (α-KG) is a key intermediate metabolite in carbon and nitrogen metabolism, playing important roles in regulating microbial stress tolerance. However, its potential to mitigate free nitrous acid (FNA) toxicity, a primary inhibitor of microalgae-nitrifier consortium, has remained unexplored. This study investigated the effects of exogenous α-KG onTribonema minusunder FNA stress and evaluated its application in wastewater treatment. Results showed that 1.0 mmol/L α-KG markedly alleviated FNA stress, but a higher concentration (2.0 mmol/L) was less effective. α-KG mitigated FNA
