Purification and characterization of glutamine synthetase from the archaebacterium Methanobacterium ivanovi.

Glutamine synthetase (GS) was purified to electrophoretic homogeneity from the obligate anaerobic archaebacterium Methanobacterium ivanovi. The 130-fold-purified enzyme was obtained by heat treatment, ion-exchange chromatography, and gel filtration. Like all other eubacterial GSs known so far, the GS of M. ivanovi was found to be a dodecamer of about 600,000 daltons composed of a single type of subunit. The enzyme was stable at 63 degrees C for 10 min and was not sensitive to oxygen. The isoelectric point was 4.6, and the optimum pH of gamma-glutamyltransferase activity was 8.0. The Km values

Methanobacterium congolense sp. nov., from a methanogenic fermentation of cassava peel.

Strain CT, a non-motile, mesophilic, hydrogenotrophic, methanogenic bacterium, was isolated from an anaerobic digester used for the treatment of raw cassava-peel waste in Congo. The cells were rods, 0.4-0.5 x 2-10 microm in size, and stained Gram-positive. Hydrogen and carbon dioxide were the only substrates that supported growth and methane production. Methane production, but not growth, occurred with CO2 in the presence of either 2-propanol, 2-butanol or cyclopentanol as hydrogen donors. The temperature range for growth was 25-50 degrees C, the optimum being between 37 and 42 degrees C. The

Methanobacterium arbophilicum sp.nov. An obligate anaerobe isolated from wetwood of living trees.

The isolation and characterization of a new methanogenic bacterium, Methanobacterium arbophilicum, is described. Isolation from wetwood enrichment cultures, that were obtained from methane-positive trees, required a medium containing inorganic salts, vitamins, and an atmosphere consisting of an 80:20 mixture of hydrogen-carbon dioxide. Isolates of M. arbophilicum were gram-positive, non-motile short rods that occurred singly, in pairs, or chains. The organism was found to be an autotroph and a strick anaerobe, and to have a pH optimum of 7.5-8.0. The optimal temperature for growth was 30 to 37

A plant-type (beta-class) carbonic anhydrase in the thermophilic methanoarchaeon Methanobacterium thermoautotrophicum.

Carbonic anhydrase, a zinc enzyme catalyzing the interconversion of carbon dioxide and bicarbonate, is nearly ubiquitous in the tissues of highly evolved eukaryotes. Here we report on the first known plant-type (beta-class) carbonic anhydrase in the archaea. The Methanobacterium thermoautotrophicum DeltaH cab gene was hyperexpressed in Escherichia coli, and the heterologously produced protein was purified 13-fold to apparent homogeneity. The enzyme, designated Cab, is thermostable at temperatures up to 75 degrees C. No esterase activity was detected with p-phenylacetate as the substrate. The e

Methanobacterium lacus sp. nov., isolated from the profundal sediment of a freshwater meromictic lake.

An autotrophic, hydrogenotrophic methanogen, designated strain 17A1(T), was isolated from the profundal sediment of the meromictic Lake Pavin, France. The cells of the novel strain, which were non-motile, Gram-staining-negative rods that measured 2-15 µm in length and 0.2-0.4 µm in width, grew as filaments. Strain 17A1(T) grew in a mineral medium and its growth was stimulated by the addition of yeast extract, vitamins, acetate or rumen fluid. Penicillin, vancomycin and kanamycin reduced growth but did not completely inhibit it. Growth occurred at 14-41 °C (optimum 30 °C), at pH 5.0-8.5 (optimu

1H NMR metabolomic profiling of Methanobacterium and Methanosarcina reveals unusual intracellular trimethylamine and acetamide production.

OBJECTIVES: Only a limited number of metabolomic datasets are available for pure cultures of methanogenic archaea, and none have reported 1H-NMR-based intracellular profiles for Methanobacterium and Methanosarcina. We aimed to characterize intracellular metabolite profiles in selected strains and to identify metabolites that challenge current metabolic models. MATERIALS AND METHODS: Nine lyophilized biomass samples representing three Methanobacterium species and three Methanosarcina species (including M. acetivorans grown on methanol, acetate and trimethylamine) were analyzed as single biolog

Convenient fluorescence-based methods to measure membrane potential and intracellular pH in the Archaeon Methanobacterium thermoautotrophicum.

New and improved methods to determine the membrane potential (Delta Psi) and the Delta pH in methanogenic archaea were developed and tested in Methanobacterium thermoautotrophicum strain Delta H. The Delta pH measurements took advantage of the pH-dependent fluorescence properties of coenzyme F(420), the major intracellular electron carrier in the organism. The protonophore p-nitrophenol did not show any interference with the F(420) fluorescence spectra and was therefore suitable to equalize internal and external pH. The method developed allowed the determination of the intracellular pH with an

Structural insight on the mechanism of regulation of the MarR family of proteins: high-resolution crystal structure of a transcriptional repressor from Methanobacterium thermoautotrophicum.

Transcriptional regulators belonging to the MarR family are characterized by a winged-helix DNA binding domain. These transcriptional regulators regulate the efflux and influx of phenolic agents in bacteria and archaea. In Escherichia coli, MarR regulates the multiple antibiotic resistance operon and its inactivation produces a multiple antibiotic resistance phenotype. In some organisms, active efflux of drug compounds will produce a drug resistance phenotype, whereas in other organisms, active influx of chlorinated hydrocarbons results in their rapid degradation. Although proteins in the MarR

Production of volatile derivatives of metal(loid)s by microflora involved in anaerobic digestion of sewage sludge.

Gases released from anaerobic wastewater treatment facilities contain considerable amounts of volatile methyl and hydride derivatives of metals and metalloids, such as arsine (AsH(3)), monomethylarsine, dimethylarsine, trimethylarsine, trimethylbismuth (TMBi), elemental mercury (Hg(0)), trimethylstibine, dimethyltellurium, and tetramethyltin. Most of these compounds could be shown to be produced by pure cultures of microorganisms which are representatives of the anaerobic sewage sludge microflora, i.e., methanogenic archaea (Methanobacterium formicicum, Methanosarcina barkeri, Methanobacterium

Production of ethane, ethylene, and acetylene from halogenated hydrocarbons by methanogenic bacteria.

Several methanogenic bacteria were shown to produce ethane, ethylene, and acetylene when exposed to the halogenated hydrocarbons bromoethane, dibromo- or dichloroethane, and 1,2-dibromoethylene, respectively. They also produced ethylene when exposed to the coenzyme M analog and specific methanogenic inhibitor bromoethanesulfonic acid. The production of these gases from halogenated hydrocarbons has a variety of implications concerning microbial ecology, agriculture, and toxic waste treatment. All halogenated aliphatic compounds tested were inhibitory to methanogens. Methanococcus thermolithotro

Enhanced biomethanation in anaerobic membrane bioreactor: decarbonization efficiency, carbon mass flow, and microbial dynamics.

Methanation of carbon dioxide (CO2) is a key process for improving carbon utilization technology. In this study, we investigated a novel approach to biomethanation from hydrogen (H2) and CO2 using an anaerobic membrane bioreactor under mesophilic conditions. System performance was evaluated by varying carbon loading rates, with a focus on decarbonization efficiency, carbon mass flow, and microbial dynamics. The results demonstrated stable operation and strong resilience, achieving a decarbonization efficiency higher than 84 % and a methane (CH4) flow-out rate of 1.1 L/L/d at a carbon loading r

Formylmethanofuran dehydrogenases from methanogenic Archaea. Substrate specificity, EPR properties and reversible inactivation by cyanide of the molybdenum or tungsten iron-sulfur proteins.

Formylmethanofuran dehydrogenases, which are found in methanogenic Archaea, are molybdenum or tungsten iron-sulfur proteins containing a pterin cofactor. We report here on differences in substrate specificity, EPR properties and susceptibility towards cyanide inactivation of the enzymes from Methanosarcina barkeri, Methanobacterium thermoautotrophicum and Methanobacterium wolfei. The molybdenum enzyme from M. barkeri (relative activity with N-formylmethanofuran = 100%) was found to catalyze, albeit at considerably reduced apparent Vmax, the dehydrogenation of N-furfurylformamide (11%), N-methy

H2-forming methylenetetrahydromethanopterin dehydrogenase, a novel type of hydrogenase without iron-sulfur clusters in methanogenic archaea.

A novel hydrogenase has recently been found in methanogenic archaea. It catalyzes the reversible dehydrogenation of methylenetetrahydromethanopterin (CH2 = H4MPT) to methenyltetrahydromethanopterin (CH identical to H4MPT+) and H2 and was therefore named H2-forming methylenetetrahydromethanopterin dehydrogenase. The hydrogenase, which is composed of only one polypeptide with an apparent molecular mass of 43 kDa, does not mediate the reduction of viologen dyes with either H2 or CH2 = H4MPT. We report here that the purified enzyme from Methanobacterium thermoautotrophicum exhibits the following o

Bioelectrochemical carbon monoxide methanation in mixed-culture microbial electrolysis cells: Performance, electron transfer pathways and microbial succession.

Bioelectrochemical methanation offers a promising avenue for carbon resource recovery. However, the direct biocathodic conversion of toxic carbon monoxide (CO) into methane, as well as the relevant interfacial extracellular electron transfer pathways and microbial community succession remain largely unexplored. Here, we systematically investigated the CO methanation process within a mixed-culture microbial electrolysis cell. Decreasing the cathodic potential from -0.90 to -1.05 V triggered a significant enhancement in methanogenic activity, boosting the cumulative methane yield from 0.142 to 1

Characterization of methanogens from landfill samples: implications for sustainable biogas production.

This case study aimed to isolate and identify methanogenic bacteria from landfill soil, mud, and leachate samples to assess their role in anaerobic digestion and biogas production. Anaerobic digestion involves the breakdown of organic matter by a diverse group of bacteria under oxygen-free conditions, resulting in the production of methane and carbon dioxide. The collected samples from the landfill were cultured in a modified mineral salt medium (MSM). Microscopic observations revealed distinct coccus and bacillus morphologies of the isolated methanogenic bacteria. Gas production experiments a
