Methods in molecular biology (Clifton, N.J.)2015eng
Phoma foveata, Phoma exigua var. exigua, and Phoma eupyrena are fungal pathogens of potato, causing gangrene or pit rot symptoms in tubers, and they are responsible for significant crop losses. Various techniques are available to identify these pathogens in the laboratory. A multiplex Plexor(®) real-time PCR method which can detect and identify these pathogens in a single reaction will be presented.

Chromoblastomycosis is a chronic infection, caused by pigmented fungi affecting skin and subcutaneous tissues characterized by verrucous nodules or plaques. Fonsecaea pedrosoi and Cladophialophora carrionii are the prevalent agents in the endemic areas. Phoma is an uncommon agent of human infection and involved mainly with phaeohyphomycosis cases. The case of a patient with a history of laceration in foot followed by verrucous aspect and scaly lesions, which had evolved for 27 years is presented. On physical examination disease was clinically compatible with chromoblastomycosis and the microsc

Onychomycosis is a nail infection most commonly caused by dermatophytes. However in recent years, non-dermatophyte molds have also been increasingly reported as causative agents of onychomycosis. Phoma glomerata is a saprophytic fungus commonly found in nature, which rarely causes infections in humans and to date, only one case has been reported in another country as a causative agent of onychomycosis. In this case report, a case of onychomycosis caused by P.glomerata was presented. A 67-year-old female patient admitted to the dermatology outpatient clinic of our hospital with a oneyear histor

Three new eremophilane sesquiterpenoids (1-3), together with six known analogues, were isolated from the marine-derived fungus Phoma sp. DXH009. Their structures were elucidated through detailed NMR and MS spectroscopic analysis, and the absolute configurations of 1-4 were determined by conformational analysis and quantum chemical TDDFT-ECD calculation. Their anti-neuroinflammatory activities were evaluated using the lipopolysaccharide (LPS)-induced BV2 microglial cells. The results indicated that compound 5 (dihydrosporogen AO-1) exhibited significant inhibitory effects on the production of n

A potato tuber rot disease of unknown cause, affecting 5 to 15% of the potato tuber, was observed at Gansu Province of China in March 2010. Sunken, round, oval, or irregular lesions formed at the umbilicus or buds of potato tubers after 30 days of storage at 4°C. These lesions gradually expanded to form khaki, lavender sunken lesions ranging from 1 to 3 cm. Small black bodies were observed in the center of the lesions after 45 days. Twenty-six diseased tubers were collected and surface sterilized with 75% alcohol. Diseased tissue was then directly transferred to potato dextrose agar (PDA) medi

Phoma sp, a fungus routinely isolated from the soil and a known plant pathogen, was found to be the cause of an aggressive, deep compartment hand infection in a renal transplant recipient. Previous reports have described minimally invasive Phoma sp infections with isolates recovered from the skin or subcutaneous tissue. This case, however, is the first reported in which Phoma sp was found to be both aggressive and deeply invasive. Histologic sections obtained from the synovium of the fourth and fifth dorsal hand compartments revealed invasive hyphal elements. Detailed examination with Grocott-

A number of species of fungi were isolated from millet (Pennisetum typhoides Staph and Hubb) and grain sorghum (Sorghum vulgare Pers) obtained from the households of patients suffering from onyalai. Several proved to be toxic to rats and chickens, including Phoma sorghina (Sacc.) which was the predominating fungus. Cultures of P. sorghina grown on maize and wheat were added to standard rations at concentrations of 5-30%. When fed to day-old New Hampshire chickens, the majority died within 4 days, exhibiting traces of blood on the beak and the cloaca. Rats given 10% or more of the mouldered mat

Three new alkaloids, phomapyrrolidones A-C (1-3), bearing a cyclopenta[b]fluorene ring system were isolated from the mycelium extract of the endophytic fungal strain Phoma sp. NRRL 46751, inhabiting Saurauia scaberrinae. Methylation of 1 afforded its N-methyl derivative 4. The planar structures and relative configurations of 1-4 were elucidated by extensive spectroscopic analysis. Phomapyrrolidones B (2) and C (3) exhibited weak antitubercular activity at subcytotoxic concentrations.

Three new diphenyl ethers, barceloneic acids A, B, and barceloneic lactone [1, 2, and 3, respectively] were isolated from a fermentation extract of a fungus of the genus Phoma. The structures of compounds 1-3 were determined by a combination of spectroscopic and single-crystal X-ray diffraction methods. The effect of these compounds on the inhibition of farnesyl-protein transferase (FPTase) was evaluated and results are presented. Barceloneic acid A [1] is a novel and modest inhibitor of FPTase with an IC50 value of 40 microM.

The exotic rangeland perennial Lepidium draba occurs as a noxious weed in 22 states, mostly in the western United States. Because chemical control measures against this invasive perennial, a member of the Brassicaceae, have not achieved adequate results, biological control is being pursued. While inventories of arthropods that feed on L. draba have been established, little is known of soilborne pathogens for possible use as biological control agents. To address this deficiency, we have surveyed for diseases of L. draba in the United States and Eurasia to identify and test potential biocontrol

BACKGROUND: Phoma stem canker (PSC) of oilseed rape (Brassica napus) is a disease caused by a complex of two related fungal pathogens: Plenodomus lingam (Leptosphaeria maculans) and P. biglobosus (L. biglobosa). While P. biglobosus is increasingly recognised as an important PSC pathogen, its sensitivity to succinate dehydrogenase inhibitor (SDHI; FRAC group 7) fungicides remains poorly understood.
RESULTS: In vitro sensitivity testing of P. biglobosus isolates from the UK (2025) and Poland (2024) revealed that resistance to boscalid and fluxapyroxad was predominant in the UK, with 95% (19/20)

Two pure fungal strains were isolated and identified from Ficus sycomorus and Morus nigra, namely, Penicillium oxalicum (OR673586) and Phoma herbarum (OR673589), respectively. The extract and fractions of secondary metabolites of each fungus were evaluated for antioxidant, anti-inflammatory, antimicrobial, antibiofilm, antidiabetic, and cytotoxic activities. The chloroform fraction of P. oxalicum showed potent cytotoxic activity (IC50 = 7.695 μg mL-1) against Hep-G2 cell line, alongside moderate antioxidant and anti-inflammatory activities. On the other hand, the P. herbarum chloroform fractio

INTRODUCTION: Endophytic fungi from desert plants are prolific producers of structurally unique stress-responsive metabolites. This study investigates the secondary metabolites of Phoma betae A. B. Frank (Didymellaceae), a desert plant endophytic fungus, aiming to discover novel bioactive compounds through advanced molecular networking strategies.
METHODS: A building blocks-based molecular network (BBMN) strategy was employed to screen the fungal extract. Target compounds were isolated using silica gel and ODS column chromatography, followed by semi-preparative HPLC purification. Structural e

Plant disease occurs simultaneously with insect attack. Arbuscular mycorrhizal fungi (AMF) modify plant biotic stress response. Arbuscular mycorrhizal fungi and pathogens may modify plant volatile organic compound (VOC) production and insect behavior. Nevertheless, such effects are rarely studied, particularly for mesocosms where component organisms interact with each other. Plant-mediated effects of leaf pathogen (Phoma medicaginis) infection on aphid (Acyrthosiphon pisum) infestation, and role of AMF (Rhizophagus intraradices) in modifying these interactions were elucidated in a glasshouse e

Winter oilseed rape (Brassica napus L.) production is constrained by a complex of fungal pathogens, including Alternaria alternata, A. brassicicola, Botrytis cinerea, Phoma lingam and Sclerotinia sclerotiorum, which are conventionally managed with synthetic fungicides such as prothioconazole. This study evaluated the biocontrol and biostimulant potential of two plant extracts (garlic, Allium sativum; Jerusalem artichoke, Helianthus tuberosus) and two bacterial fermentation supernatants (Enterobacter sp. and Paenibacillus sp.) against these pathogens under in vitro, greenhouse and field conditi

A total synthesis of the PAF antagonist phomactin A (1), isolated from the marine fungus Phoma sp. is described. The synthesis is based on a Cr(II)/Ni(II) macrocyclisation from the aldehyde vinyl iodide 14, leading to the key phomactatrienol intermediate 16a, followed by elaboration of 16a to the epoxyketone 21, which undergoes spontaneous pyran and hemiacetal ring formation to 1 on deprotection with DDQ.
