Phospholipases: an overview.

Phospholipids are present in all living organisms. They are a major component of all biological membranes, along with glycolipids and cholesterol. Enzymes aimed at cleaving the various bonds in phospholipids, namely phospholipases, are consequently widespread in nature, playing very diverse roles from aggression in snake venom to signal transduction, lipid mediators production, and digestion in humans. Although all phospholipases target phospholipids as substrates, they vary in the site of action on the phospholipids molecules, physiological function, mode of action, and their regulation. Sign

Phospholipases: Where Do We Stand?

Phospholipases are a diverse group of lipolytic enzymes that hydrolyze membrane glycerophospholipid substrates at specific ester bonds at the lipid-water interface. They are involved in numerous functions such as the production of various intracellular signaling molecules, inflammation, and human digestion. These highly widespread enzymes in nature vary considerably in terms of structure, mode of action, and regulation. Research on phospholipases has progressed greatly in recent decades. This review aims to provide a general framework of the current understanding of phospholipases and a discus

Secreted Phospholipases A2 - not just Enzymes.

Secreted phospholipases A2 (sPLA2s) constitute, physiologically and pathologically, a very important family of enzymes. Most actions of sPLA2s have been explained by their phosphatidylglycerol sn-2 hydrolytic activity. However, since pharmacologically active sPLA2 molecules without enzymatic activity have been discovered, first in snake venom and then also in human, it has become increasingly evident that, on many occasions, the action of these proteins has to be considered as arising from the interplay of their receptor-binding and enzymatic functions. The number of known sPLA2-interacting mo

Effects of local anaesthetics on phospholipases.

1. The effects of six local anaesthetics have been studied on the activities of soluble phospholipases A2 (EC 3.1.1.4) and lysophospholipase (EC 3.1.1.5). 2. Phospholipase A2 activity in human seminal plasma towards sonicated radioactively-labelled phosphatidylethanolamine was slightly stimulated a low and inhibited at high concentrations of all anaesthetic compounds employed. The order of decreasing potency was chlorpromazine, dibucaine, tetracaine, lidocaine, cocaine and procaine. In line with previous findings, the mode of inhibition was seen to be competitive with respect to Ca2+. 3. Phosp

Scorpion venom phospholipases A2: A minireview.

Many venomous species, including snakes, bees and scorpions, contain a variety of secreted phospholipases A2 (sPLA2) that contribute to prey digestion and venom toxicity. Based on their primary structures, the different venom sPLA2 have been classified into four groups I, II, III and IX. While the structure of sPLA2 groups II and I has been well characterized, only one crystal structure of group III sPLA2 from bee venom was described. Scorpion venom sPLA2 belong to group III with a particular heterodimeric structure composed of a long enzymatic chain linked by a disulfide bridge to a Short one

Serum phospholipases A2 after aortobifemoral reconstruction.

Phospholipase A2 has been implicated in the pathogenesis of local and distant tissue injury after ischaemia and reperfusion. A common operation inducing ischaemia and reperfusion is aortobifemoral reconstruction, during which the aorta is clamped and the blood supply via the inferior mesenteric artery and iliac arteries is interrupted. The purpose of the present work was to study the catalytic activity concentration of phospholipase A2 and the mass concentrations of group I and group II phospholipases A2 in the sera of patients undergoing aortobifemoral reconstruction. Both the catalytic activ

Serum phospholipases A2 in inflammatory diseases.

Phospholipase A2 (EC 3.1.1.4; PLA2) is detected in serum by determination of either the catalytic activity of the enzyme or the concentration of the enzyme protein by immunoassays. The most sensitive methods for determining PLA2 catalytic activity are radiometric assays, with a substrate of synthetic phospholipid (e.g., phosphatidylcholine or phosphatidylethanolamine) containing a 14C- or 3H-labeled fatty acid at the sn-2-position. Membranes of autoclaved Escherichia coli grown in the presence of radioactive oleic acid may also be used as a substrate. The released fatty acids are separated fro

What can venom phospholipases A(2) tell us about the functional diversity of mammalian secreted phospholipases A(2)?

Most venomous animals including snakes, bees and scorpions contain a variety of venom phospholipases A(2) (vPLA(2)s) which participate in both digestion of prey and venom toxicity. So far, more than 150 vPLA(2)s have been characterized. They all have a conserved fold with several disulfide bridges, can be catalytically active or not, and several of them can display a tremendous array of toxic effects including neurotoxicity and myotoxicity. Furthermore, the molecular diversity of vPLA(2)s found within a single snake venom can result from positive Darwinian selection. Over the last decade, rece

Spider's venom phospholipases D: A structural review.

Spider venoms are complex mixtures of proteins, peptides and small organic and inorganic molecules. Among the proteins, phospholipases D (PLDs) present the major portion, and till now they are the most studied enzymes in spider venom. These PLDs have been divided into two classes, I and II, based on their primary and tertiary structure. Currently, crystal structures of both classes of these enzymes are available in the Protein Data Bank (PDB). Their three-dimensional structure is composed of eight α-helices and eight β-strands forming the ubiquitous fold called triosephosphate isomerase (TIM)

Natural inhibitors of toxic phospholipases A(2).

Endogenous proteins isolated from the serum of snakes have been found to be natural inhibitors displaying anti-hemorrhagic, anti-neurotoxic or anti-myotoxic activity. Some of these proteins inhibit phospholipase A(2) (PLA(2)) activity. We review in brief here the properties, structure and classification of these PLA(2) inhibitors (PLIs), focusing in particular on the mechanism of neutralization of the toxic PLA(2)s by anti-neurotoxic PLIs. We also discuss: 1) the protection provided by these molecules against endogenous snake venom PLA(2)s; 2) their specificity for neurotoxic snake venom PLA(2

Molecular evolution of group II phospholipases A2.

The nucleotide sequences of 13 cDNAs encoding group II phospholipases A2 (PLA2s), which are from viperidae snake venoms and from mammalian sources, were aligned and analyzed by phylogenetic trees constructed using various components of the sequences. The evolutionary trees derived from the combined sequences of the untranslated (5' and 3') region and the signal peptide region of cDNAs were in accord with the consequences from taxonomy. In contrast, the evolutionary trees from the mature protein-coding region sequences of cDNAs and from the amino acid sequences showed random patterns. These obs

Effect of phospholipases on factor-VIII activity.

The effect of the enzymes phospholipases C and D on Factor VIII were investigated. Phospholipase D was found to activate the partially purified intact Factor-VIII molecule maximally at a final concentration of 0.6 U/ml. Neither the dissociated small molecular weight component nor the high molecular weight component were activated. Phospholipase C, on the other hand, inactivated both the intact and the dissociated Factor-VIII molecule. Phospholipase D, however, had no effect on the haemophilic cryoprecipitate or the partially purified haemophilic Factor VIII. The implications of these results f

PTEN regulates phospholipase D and phospholipase C.

PTEN is an ubiquitously expressed tumor suppressor which plays a prominent role in the pathogenesis of many types of sporadic solid tumors, including breast cancer, as well as hematologic malignancies. Germline PTEN mutations cause 85% of Cowden syndrome (CS), characterized by a high risk of breast and thyroid cancers, and 65% of Bannayan-Riley-Ruvalcaba syndrome (BRRS), characterized by lipomatosis, hemangiomas and speckled penis. Historically, PTEN's role in tumor suppression has been linked to the down-regulation of the PI3K/AKT pathway by PTEN's lipid phosphatase activity. Beyond the AKT p

Phospholipases D: An Update on Structure-Function Relationships.

Phospholipases D (PLD) are lipolytic and interfacial enzymes that hydrolyze the distal phosphodiester bond of phospholipid substrates. PLD are widespread in prokaryotes and eukaryotes and have various functions in bacteria, yeast, plants, and mammals. The PLD reaction frees phosphatidic acid (PA), which is an intermediate in the lipid pathway but also a signaling molecule involved in metabolic, cellular, and physiological processes. Depending on their primary structure, PLD belong to two different groups: the HKD family, characterized by an HXKX4D sequence that is often duplicated, and the non

Phospholipases: Paving the Way for a New Life.

Phospholipases are a large group of enzymes with ramifications on diverse physiological processes. These enzymes are involved in releasing phospholipids, which are subsequently utilized in the production of prostaglandin and calcium signaling. Calcium signaling is central to the function of phospholipases and calcium oscillations have an impact on the variety of events of embryo development. Additionally, the diverse isoforms of the enzymes can activate gene expression and regulatory factors in a stage-specific manner. Any alterations in the level of these enzymes or the pathways regulating th

Secreted phospholipases of the lung pathogen Legionella pneumophila.

Legionella pneumophila is an intracellular pathogen and the main causative agent of Legionnaires' disease, a potentially fatal pneumonia. The bacteria infect both mammalian cells and environmental hosts, such as amoeba. Inside host cells, the bacteria withstand the multifaceted defenses of the phagocyte and replicate within a unique membrane-bound compartment, the Legionella-containing vacuole (LCV). For establishment and maintenance of the infection, L. pneumophila secretes many proteins including effector proteins by means of different secretion systems and outer membrane vesicles. Among the

Membrane and inhibitor interactions of intracellular phospholipases A2.

Studying phospholipases A2 (PLA2s) is a challenging task since they act on membrane-like aggregated substrates and not on monomeric phospholipids. Multidisciplinary approaches that include hydrogen/deuterium exchange mass spectrometry (DXMS) and computational techniques have been employed with great success in order to address important questions about the mode of interactions of PLA2 enzymes with membranes, phospholipid substrates and inhibitors. Understanding the interactions of PLA2s is crucial since these enzymes are the upstream regulators of the eicosanoid pathway liberating free arachid
