The stems and leaves of Phytolacca americana were phytochemically investigated, leading to the isolation and identification of nine compounds. Two new compounds, 6,7-dimethoxy-4β-hydroxypeltogynane (1) and phytolaccamide A (2) were characterized, with compound 1 assigned an absolute configuration of 2R, 3S, 4S. Compounds 1, 3 and 4 represrent rare peltogynane-type structures. Preliminary in vitro antibacterial and antifungal screening showed no obvious inhibitory activity against Staphylococcus aureus subsp. aureus ATCC 29213 or Candida albicans ATCC 10231 at 100 μM.

We studied the effects of phytolaccosides, saponins from Phytolacca americana, on the intestinal absorption of heparin in vitro and in vivo. The absorption enhancing activity of these compounds (phytolaccosides B, D2, E, F, G and I) was determined by changes in transepithelial electrical resistance (TEER) and the transport amount of heparin disaccharide, the major repeating unit of heparin, across Caco-2 cell monolayers. With the exception of phytolaccoside G, all of them decreased TEER values and increased the permeability in a dose-dependent and time-dependent manner. In vitro, phytolaccosid

In the present study, the accumulation of Mn and other metals by Phytolacca Americana L. from contaminated soils in Hunan Province, South China, was investigated. Results showed that the average concentrations of Mn in the leaves and roots reached 2198 and 80.4 mg kg(-1) (dry weight), respectively, with a maximum 13,400 mg kg(-1) in the leaves. A significant correlation was found between Mn concentrations in the plant leaves and those in the corresponding soils. Hydroponic experiments were also conducted to study the Cd uptake ability and interactions between Mn and Cd in the plant. It was fou

The experimental results have shown that the toxicity of variously processed Phytolacca americana is lower than that of the original rude drug, i.e. the local irritability tends to decrease by 16.7-83.3%; LD50 to elevate by 1.66-10.47 times; expectoration to increase by 1.10-1.57 times; but diuresis mostly to decrease by 16.0-45.0%. This reveals that the main object of processing Phytolacca americana is to lower toxicity, increase expectoration and relieve diuresis to reduce fluid retention.

BACKGROUND: Phytolacca americana L. (American pokeweed), an alien plant species in China, has become widely distributed in both rural and urban areas. American pokeweed contains toxic substances such as saponins and oxalates, which have led to multiple poisoning incidents in recent years. Here, we describe a fatal case of acute American pokeweed leaf poisoning in China, emphasizing the clinical progression, toxin concentration, and implications for public health interventions. To our knowledge, no such description currently exists in the literature.
METHODS: A comprehensive epidemiological in

In recent years, nano-contamination in the soil environment has aroused concern. But it is still uncertain whether the interactions of nano- and metal-pollutants would have a combined toxic effect on plants. In this study, we investigated the effects of joint exposure to zinc oxide nanoparticles (ZnO NPs) and Cd on the root tissue of Phytolacca americana L. Spin-polarized density functional theory simulations assumed that the plant may undergo metal toxicity or acidosis upon joint exposure to ZnO NPs/Cd. Subsequently, experimental exposure of P. americana verified the combined toxic effects. T

Phytolacca americana L. can accumulate large amounts of heavy metals in its aerial tissues, especially cadmium (Cd) and manganese (Mn). It has great potential for use in phytoextraction of metals from multi-metal-contaminated soils. This study was conducted to further investigate the Cd- and Mn-tolerance strategies of this plant. Concentrations of non-protein thiols (NPTs) and phytochelatins (PCs) in leaves and roots increased significantly as the concentration of Cd in solution increased. The molar ratios of PCs:soluble Cd ranged from 1.8 to 3.6 in roots and 8.1 to 31.6 in leaves, suggesting

BACKGROUND: Phytolacca americana L. (pokeweed) is a medicinal plant containing diverse bioactive compounds, including phenolics, flavonoids, and saponins. Variation in phytochemical accumulation and concerns associated with harvesting from natural populations highlight the need for controlled and sustainable propagation systems. This study compared solid, liquid, and temporary immersion system (TIS) cultures of P. americana nodal explants containing axillary buds and evaluated their effects on plant growth and phytochemical characteristics.
METHODS: Nodal explants were cultured on solid and l

BACKGROUND: In response to herbivory from insects, plants have evolved complex defense strategies. Alien plants often escape control by specialist insects in their introduced range. This release from herbivory pressure may lead to a reallocation of the defensive strategies of alien plants, thereby promoting their invasiveness. However, the molecular mechanisms underlying this resource reallocation process remain unclear. In the present study, we used Phytolacca americana as a model species, and combined transcriptomic and metabolomic analyses to investigate the molecular basis of its invasion

Phytolacca americana L. is of great interest as a traditional additive in various folk remedies in several countries, including Turkey. We aimed to determine the chemical profile (assisted by high-Performance liquid chromatography-electrospray ionization-tandem mass apectrometry (HPLC-ESI-MS/MS) experiments of three extracts obtained by different polarity solvents viz. ethyl acetate (to extract semipolar compounds), methanol and water (to extract highly polar metabolites) from P. americana leaves. Their anti-diabetic effects were investigated in vitro by assessing their inhibition toα-amylase

The presence of glucosamine trisaccharides in the glycocalyx of surface epithelial cells of the endometrium of hyperstimulated pregnant rats was compared with that of normal pregnant animals by means of lectin-avidin-ferritin histochemistry. The lectin Phytolacca americana was used. A statistically significant reduction in ferritin particles and hence, glucosamine trisaccharides, was found in the glycocalyx of the hyperstimulated animals when compared with that of the controls. This reduction could contribute to the failure of implantation in hyperstimulated animals.

Ribosome-inactivating proteins (RIPs) are a class of plant defense proteins with N-glycosidase activity (EC 3.2.2.22). Pokeweed antiviral protein (PAP) is a Type I RIP isolated from the pokeweed plant, Phytolacca americana, thought to confer broad-spectrum virus resistance in this plant. Through a combination of standard molecular techniques and RNA sequencing analysis, we report here that a small RNA binds and cleaves the open reading frame of PAP mRNA. Additionally, sRNA targeting of PAP is dependent on jasmonic acid (JA), a plant hormone important for defense against pathogen infection and

Pokeweed (Phytolacca americana) lectin was found to stain the secretory granules in epithelial Paneth cells of small intestine in mice and rats. The distribution of Paneth cells stained with this lectin was identical to that obtained by another immunohistochemical marker for lysozyme. However, in comparison with other immunohistochemical markers, Pokeweed lectin is a more robust method for identifying Paneth cells in histological sections and for studying their secretory granules. Co-expression of the Pokeweed lectin binding sites in some oligomucous cells within the crypts suggested a close d

BACKGROUND: Oral lichen planus (OLP) is one of the most common oral mucosal lesions affecting 0.5-2% of the adult population. It is difficult to distinguish between OLP and other oral mucosal diseases. Structural changes in the glycans of saliva proteins might be reliable indicators of OLP. However, little is known about the alteration of salivary glycopatterns during OLP.
OBJECTIVE: We aimed to investigate the alterations of salivary protein glycosylation related to OLP.
MATERIAL AND METHODS: Twenty-eight patients with OLP and 30 age- and sex-matched healthy volunteers (HVs) were enrolled i

Pokeweed antiviral protein (PAP) is a plant-derived N-glycosidase ribosomal-inactivating protein isolated from Phytolacca americana. The antiviral activity of PAP has been described in several viruses. This study was to investigate the antiviral activity of PAP against Japanese encephalitis virus (JEV) infection in vitro and in vivo. Antiviral activity of PAP against JEV infection was evaluated in vitro using plaque forming assay, qRT-PCR and Western blot analysis. In vitro results showed that PAP inhibited replication of JEV in a dose-dependent manner with 50% inhibitory concentration (IC(50)
