The numbers of neurons were counted in the ventral and dorsal anterior and ventral and dorsal posterior part of the putamen of 11 Huntington's disease and nine control patients. The process of neuronal cell loss in Huntington's disease proved to be unevenly spread throughout the putamen; the ventral anterior part was relatively spared as compared with the other parts of the putamen. A significantly higher number of neurons was found in the ventral part of the putamen in the controls.

A 55-year-old right-handed man with a history of hypertension suddenly fell and developed right hemiparesis. Neurological examination revealed that he was alert, but did not appropriately respond to verbal questions and commands. Detailed examination revealed that he could correctly respond to written commands. His speech was almost fluent, showing no paraphasia and normal articulation. His written sentences were legible. Pure tone audiometry showed that his auditory acuity was relatively preserved. His brainstem auditory evoked potential components from I to V were recorded bilaterally with n

Gene expression patterns in the postmortem putamen of patients with Tourette syndrome (TS) were investigated using cDNA microarrays. A cDNA neuroarray comprising 1537 genes known to be related to neurological or neuropsychiatric disorders was used to compare patient samples (n=3) with those from control subjects (n=4). Z test and Z ratio were used to analyze results; seven genes were found to be upregulated according to our definition (P<0.1, two-tailed, for Z test; P<0.05 for Z ratio) and three were found to be downregulated. Validation experiments were performed using reverse transcription p

Motor adaptation in cortico-striato-thalamo-cortical loops has been studied mainly in animals using invasive electrophysiology. Here, we leverage functional neuroimaging in humans to study motor circuit plasticity in the human subcortex. We employed an experimental paradigm that combined two weeks of upper-extremity immobilization with daily resting-state and motor task fMRI before, during, and after the casting period. We previously showed that limb disuse leads to decreased functional connectivity (FC) of the contralateral somatomotor cortex (SM1) with the ipsilateral somatomotor cortex, inc

Age is a major risk factor for neurodegenerative diseases. Shortening of leucocyte telomeres with advancing age, arguably a measure of "biological" age, is a known phenomenon and epidemiologically correlated with age-related disease. The main mechanism of telomere shortening is cell division, rendering telomere length in post-mitotic cells presumably stable. Longitudinal measurement of human brain telomere length is not feasible, and cross-sectional cortical brain samples so far indicated no attrition with age. Hence, age-related changes in telomere length in the brain and the association betw

Subcortical plasticity has mainly been studied using invasive electrophysiology in animals. Here, we leverage precision functional mapping (PFM) to study motor plasticity in the human subcortex during 2 weeks of upper-extremity immobilization with daily resting-state and motor task fMRI. We found previously that, in the cortex, limb disuse drastically impacts disused primary motor cortex functional connectivity (FC) and is associated with spontaneous fMRI pulses. It remains unknown whether disuse-driven plasticity pulses and FC changes are cortex specific or whether they could also affect move

To analyze the organization of corticostriatal motor inputs, we examined the neuronal responses in the putamen (Put) to stimulation in the primary motor cortex (MI) and the supplementary motor area (SMA). Stimulating electrodes were chronically implanted in the distal and proximal parts of the forelimb representation of the MI and in the forelimb representation of the SMA in Japanese monkeys (Macaca fuscata). Stimulation in the MI and SMA evoked orthodromic spike discharges in both phasically active and tonically active Put neurons. The latency of excitation evoked by MI stimulation was shorte

In a rodent model of Parkinson's disease (PD), levodopa-induced involuntary movements have been linked to striatal angiogenesis - a process that is difficult to document in living human subjects. Angiogenesis can be accompanied by localized increases in cerebral blood flow (CBF) responses to hypercapnia. We therefore explored the possibility that, in the absence of levodopa, local hypercapnic CBF responses are abnormally increased in PD patients with levodopa-induced dyskinesias (LID) but not in their nondyskinetic (NLID) counterparts. We used H215O PET to scan 24 unmedicated PD subjects (12 L

INTRODUCTION: Gratitude plays a crucial role in promoting affective well-being, yet the neural mechanisms underlying their relationship remains unclear. Given the central role of the nucleus accumbens (NAcc) and amygdala in emotion and reward processing, this study investigated whether resting-state functional connectivity (RSFC) of these subcortical regions correlates with trait gratitude and mediates its association with affective well-being.
METHODS: Resting-state fMRI data were collected from 363 young adults. Seed-based connectivity analyses identified NAcc and amygdala connectivity patt

A woman in her 70s presented with approximately 2 years of sudden-onset gait and cognitive problems. She had been diagnosed with normal pressure hydrocephalus (NPH) and underwent ventriculoperitoneal shunt (VPS) placement 1 year prior. Before VPS placement, brain imaging showed ventriculomegaly and chronic infarction of the right putamen and claustrum. A lumbar drain trial resulted in modest improvement of gait dysfunction. She underwent VPS placement for suspected NPH, but her symptoms remained unchanged. Examination revealed mild cognitive impairment, left-sided and lower body predominant pa

PURPOSE: The purpose of this report is to demonstrate a rare clinical manifestation of apraxia eyelid opening related to a basal ganglia lesion.
CASE REPORT: In this study, we report a 91-year-old woman suffering from difficulty in eyelid opening after being treated for myocardial ischemia with dual antiplatelet medications. She could open her eyelid with fingers touching her forehead. Brain computed tomography revealed a right putamen hemorrhage. Surface electromyography revealed persistent frontalis muscle contraction during relaxation of orbicularis oculi muscles. Apraxia of eyelid opening

BACKGROUND: Similar to the model for Sydenham's chorea, antineuronal antibodies, which develop in response to a preceding streptococcal infection, have been speculated to have a role in the development of Tourette syndrome (TS).
METHODS: Serum antibodies against human caudate, putamen, and globus pallidus (interna and externa) were assayed by enzyme-linked immunosorbent assay (ELISA) and Western blot techniques and results were correlated with clinical characteristics and markers of streptococcal infection.
SUBJECTS: A total of 41 children with TS (mean age, 11.3 years) and 39 controls (mean

Preliminary evidence from postmortem studies of X-linked dystonia-parkinsonism (XDP) suggests tissue loss may occur first and/or most severely in the striatal striosome compartment, followed later by cell loss in the matrix compartment. However, little is known about how this relates to pathogenesis and pathophysiology. While MRI cannot visualize these striatal compartments directly in humans, differences in relative gradients of afferent cortical connectivity across compartments (weighted toward paralimbic versus sensorimotor cortex, respectively) can be used to infer potential selective loss

BACKGROUND: Complex motor stereotypies in children are repetitive rhythmic movements that have a predictable pattern and location, seem purposeful, but serve no obvious function, tend to be prolonged, and stop with distraction, e.g., arm or hand flapping, waving. They occur in both "primary" (otherwise typically developing) and secondary conditions. These movements are best defined as habitual behaviors and therefore pathophysiologically hypothesized to reside in premotor to posterior putamen circuits. This study sought to clarify the underlying neurobiologic abnormality in children with prima

Social coordination in primates relies on parieto-frontal networks encoding self- and others' actions. These areas send convergent projections to the putamen, but its role in representing self- and others' actions remains unknown. We recorded neuronal activity from anatomically characterized putamen regions of two male macaques during a Mutual Action Task (MAT), where a monkey and a human took turns grasping a multi-affordance object based on sensory cues. Cortico-striatal synaptic input, indexed by local field potentials, mirrored known cortical dynamics during sensory instructions and moveme

There is currently debate with regards to the role of phonological deficit in Chinese reading difficulty, even though some researchers have suggested that the deficit of phonological processing is also a signature of developmental dyslexia in Chinese, as has been found in alphabetic languages. In this study, we examined the brain mechanisms of phonological deficit in Chinese children with developmental dyslexia (DD) during an auditory rhyming judgment task. First, we examined structural differences in Chinese dyslexia by comparing gray and white matter volume in Chinese children with DD, age-m

BACKGROUND: Growing evidence indicates that major depressive disorder (MDD) is characterized by accelerated biological aging, including greater age-related changes in physiological functioning. The disorder is also associated with abnormal neural reward circuitry, particularly in the basal ganglia (BG). Here we assessed age-related changes in BG volume in both patients with MDD and healthy control participants.
METHODS: We obtained whole-brain T1-weighted images from patients with MDD and healthy controls. We estimated grey matter volumes of the BG, including the nucleus accumbens, caudate, p

OBJECTIVE: To explore the selectivity of neuronal somatotopic representation in the striatum of patients with unilateral task-specific dystonia of the right arm.
MATERIALS AND METHODS: The authors used fMRI in 14 right-handed dystonic subjects to examine putaminal organization. Subjects performed flexion/extension of the right and left fingers and toes, and contraction of the lips.
RESULTS: Compared to healthy volunteer subjects, dystonic subjects had altered somatotopic organization in the left putamen, contralateral to the affected hand. Disease severity correlated with underactivation and
