Barbiturates inhibit progesterone synthesis in cultured Leydig tumor cells and human granulosa cells.

Screening drugs used in obstetrical practice for effects on steroid hormone synthesis revealed that phenobarbital inhibited progesterone synthesis in MA-10 Leydig tumor cells. The inhibition was apparently a drug class effect since it could be reproduced by other barbiturates. Barbiturate blockade was reversible and could be bypassed in the MA-10 cells by using 22-hydroxycholesterol. Human granulosa cell progesterone synthesis was also inhibited in a dose dependent fashion by phenobarbital, secobarbital and barbituric acid. Significant inhibition occurred in dose ranges that would be therapeut

Synergistic activity of purine metabolism inhibitors in cultured human tumor cells.

Cytotoxic effects of 3-deazaguanosine (3-DGUO) result from the inhibition of DNA synthesis and incorporation of the drug into DNA. Synergistic antiproliferative effects of a combination of 3-deazaguanosine and 2-beta-D-ribofuranosylthiazole-4-carboxamide, a potent inhibitor of inosine monophosphate dehydrogenase, was observed in human tumor cells. Inosine reversed the antiproliferative effects of the 3-deazaguanosine but not 2-beta-D-ribofuranosylthiazole-4-carboxamide. 3-Deazaguanosine monophosphate was shown to inhibit the activity of the de novo purine synthesis enzyme, 5'-phosphoribosyl-5-

Photodegraded nifedipine promotes transferrin-independent gallium uptake by cultured tumor cells.

UNLABELLED: It was reported previously that normal soft tissues accumulate 67Ga by a transferrin-dependent route, but uptake by tumors can be transferrin independent. It was also reported that, although overexpression of the transferrin receptor can promote Ga avidity, the transferrin-independent uptake of 67Ga is significant and can be augmented to exceed transferrin-mediated levels by increasing extracellular calcium. In assessing the effect of calcium channel blockers on uptake of 67Ga, it was observed that, after exposure to light (either visible or ultraviolet [UV]), nifedipine strongly p

Tumor promoter receptors regulating neurite formation in cultured human neuroblastoma cells.

The mouse skin tumor promoter phorbol-12,13-dibutyrate (PDBU) reversibly enhanced neurite outgrowth in SH-SY5Y human neuroblastoma cells, whether serum was present or absent. The half-maximum response in serum occurred at 10 nM. The binding of [20-3H]phorbol-12,13-dibutyrate [( 3H]PDBU) was studied. Five mouse skin tumor promoters, which included teleocidin, mezerein, and three structural congeners of phorbol, enhanced neurite outgrowth and inhibited binding of [3H]PDBU, but two nonpromoting phorbol congeners did neither. Saccharin and cyclamate, promoters in rat bladder, did not inhibit [3H]P

Expression of somatostatin messenger RNA and receptor in cultured brain tumor cells.

In order to understand the role of somatostatin in tumor cell growth, the expression of somatostatin in 7 cultured brain tumor cells was investigated. Radioimmunoassay demonstrated the production of somatostatin into the culture supernatant of 6 tumor cells. The expression of somatostatin mRNA and receptor was investigated by reverse transcription polymerase chain reaction (RT-PCR) and a binding assay with 125I-somatostatin-14, respectively. Only two cell lines, GBS-1 and U87MG, expressed somatostatin mRNA, whereas somatostatin receptor was expressed in all cultured cells. These results sugges

Effects of conditioned media from murine lung cancer cells and human tumor cells on cultured myotubes.

Factors secreted from tumors/tumor cells are hypothesized to cause skeletal muscle wasting in cancer patients. We examined whether cancer cells secrete factors to promote atrophy by evaluating the effects of conditioned media (CM) from murine lung cancer cells and primary cultures of human lung tumor cells on cultured myotubes. We evaluated murine Lewis lung carcinoma (LLC) and KRASG12D cells, and primary cell lines derived from tumor biopsies from patients with lung cancer (hTCM; n = 6). In all experiments, serum content was matched across treatment groups. We hypothesized that CM from murine

XPD Functions as a Tumor Suppressor and Dysregulates Autophagy in Cultured HepG2 Cells.

BACKGROUND: Recent clinical studies have linked polymorphisms in the xeroderma pigmentosum group D (XPD) gene, a key repair gene involved in nucleotide excision repair, to increased risk of hepatocellular carcinoma (HCC). However, the cellular effects of XPD expression in cultured HCC cells remain largely uncharacterized. Therefore, the aim of this study was to characterize the in vitro cellular effects of XPD expression on the HCC cell line HepG2. MATERIAL AND METHODS: HepG2 cells were transfected as follows to create four experimental groups: pEGFP-N2/XPD plasmid (XPD) group, EGFP-N2 plasmi

Cultured tumor cells of murine submandibular gland origin: a model to investigate pHi regulation of salivary cells.

Intracellular pH (pHi) and several ion transport mechanisms in cultured murine salivary gland cells (SCA-9) were studied using a videomicrofluorometric method and the H+-specific probe C-SNARF-1. The aim of this study was to test SCA-9 cells' pHi regulation mechanisms and evaluate if this cell line is representative of submandibular gland cells. Resting pHi in unstimulated cells was estimated to be 7.17+/-0.07. To investigate the presence of Na+/H+ and Cl-/HCO3- antiports as well as Na+/K+/2Cl- symports in SCA-9 cells, we used different specific blockers, dimethyl-amiloride, disulfonic stilben

Cationic transfectors increase accumulation in cultured tumor cells of radiolabeled antisense DNAs without entrapment.

While cellular accumulations in culture of oligomers, such as interfering RNA and antisense DNA, are reported to benefit from the addition of transmembrane transfectors (TFs), the extent to which individual TFs improve cellular delivery is usually inferred and rarely measured. The goal of this investigation was to use radioactivity to measure in cells in culture the degree to which accumulations of DNA increased when complexed with TFs and without DNA entrapment in vesicles. The antisense (AS) DNA targeting mdr1 mRNA coding for P-glycoprotein (Pgp) and its sense (S) complement DNA were radiola

Intracellular localization of drugs in cultured tumor cells by ion microscopy and image processing.

Several drugs, containing a halogen atom, F or Br, that are being used in antiviral or anticancer therapy, were studied for their localization in cultured cells by ion microanalysis. The association allows to reduce the exposure time to define the intracellular localization of the studied element. The topography of the cells is given by the image of the polyatomic ion 26CN-. The image of the distribution of 81Br- or 19F-, coded in another color scale, can be superimposed, giving a polychromic image of the cell, thus showing the intracellular localization of the compound. MCF-7 tumor cells were

Inhibition of epidermal growth factor binding to cultured mouse epidermal cells by tumor promoters.

The tumor promotor 12-O-tetradecanoylphorbol-13-acetate (TPA) caused partial inhibition of the binding of [125I]epidermal growth factor ([125I]EGF) to intact mouse epidermal cells (HEL/37). The proportion of [125I]EGF binding which was insensitive to TPA inhibition decreased with increasing EGF concentration. The partial inhibition of [125I]EGF binding was not due to destruction of TPA or to covalent linkage of EGF to receptor sites. The binding of [125I]EGF to HEL/37 cells showed a curvilinear Scatchard plot; this was converted into a linear plot in the presence of TPA. We conclude that TPA a

Effects of tumor cell culture supernatants on macrophages.

It is proved that supernatants of various tumor cell cultures, as well as cell-free ascitic fluid and sera from tumor bearing animals induce the detachment of macrophages from glass, the loss of their ability to form pseudopodia, the inhibition of their migration and the permeability alteration of their membranes. The cytotoxic effect is not produced by normal tissues, excepting placenta. The effect of tumor cell culture supernatants depends on the characteristic evolution time of each tumor; irradiation diminishes this effect. The involvement of macrophages in immune surveillance and response

Modeling the novel SERD elacestrant in cultured fulvestrant-refractory HR-positive breast circulating tumor cells.

PURPOSE: Metastatic hormone receptor-positive (HR+) breast cancer initially responds to serial courses of endocrine therapy, but ultimately becomes refractory. Elacestrant, a new generation FDA-approved oral selective estrogen receptor degrader (SERD) and antagonist, has demonstrated efficacy in a subset of women with advanced HR+breast cancer, but there are few patient-derived models to characterize its effect in advanced cancers with diverse treatment histories and acquired mutations. METHODS: We analyzed clinical outcomes with elacestrant, compared with endocrine therapy, among women who h

Pleomorphic adenoma gene 1 is expressed in cultured benign and malignant salivary gland tumor cells.

The pleomorphic adenoma gene 1 (PLAG1) is activated by reciprocal chromosomal translocations involving 8q12 in a subset of salivary gland pleomorphic adenomas. PLAG1 encodes a zinc finger protein and was initially reported to be expressed in placenta and fetal tissues, with no detectable expression in other normal adult tissues. By Northern blotting we have detected PLAG1 expression in a wide set of normal adult tissues, including heart, placenta, spleen, prostate, testis, ovary, and small intestine. We have performed reverse transcriptase-PCR and Northern blot analyses to study the expression

Tumor-Treating Fields Alter Nanomechanical Properties of Pancreatic Ductal Adenocarcinoma Cells Co-Cultured with Extracellular Matrix.

Tumor-Treating Fields (TTFields), a novel therapeutic avenue, is approved for therapy in Glioblastoma multiforme, malignant pleural mesothelioma, and metastatic non-small cell lung cancer (NSCLC). In pancreatic ductal adenocarcinoma (PDAC), several clinical trials are underway to improve outcomes, yet a significant knowledge gap prevails involving the cell-extracellular matrix (ECM) crosstalk. Herein, we hypothesized that treatment with TTFields influence this crosstalk, which is reflected by the dynamic alteration in nanomechanical properties (NMPs) of cells and the ECM in a co-culture system

Thiol redox modulation of tumor necrosis factor-alpha responsiveness in cultured AIDS-related Kaposi's sarcoma cells.

Both clinical and experimental evidence indicates that AIDS-related Kaposi's sarcoma (AIDS-KS) has a multifactorial pathogenesis with factors such as HIV viral load, latent virus induction, and opportunistic infections contributing to disease progression. However, a consistent feature that unites these apparently diverse putative etiologic agents is sustained serum elevations of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-alpha). While virtually every cell responds to TNF-alpha with gene activation, the extent of TNF-alpha-mediated cellular signaling is regulated by a d

Tumor necrosis factor-α regulates distinct molecular pathways and gene networks in cultured skeletal muscle cells.

BACKGROUND: Skeletal muscle wasting is a debilitating consequence of large number of disease states and conditions. Tumor necrosis factor-α (TNF-α) is one of the most important muscle-wasting cytokine, elevated levels of which cause significant muscular abnormalities. However, the underpinning molecular mechanisms by which TNF-α causes skeletal muscle wasting are less well-understood. METHODOLOGY/PRINCIPAL FINDINGS: We have used microarray, quantitative real-time PCR (QRT-PCR), Western blot, and bioinformatics tools to study the effects of TNF-α on various molecular pathways and gene networks

[Role of proteins in Fas-mediated apoptosis in tumor cells and lymphocytes co-cultured in vitro].

The investigation deals with the role of Fas, FasL, RIP, caspase 3, and PARP taking part in Fas-mediated apoptosis, and contributing to in vitro interaction of hepatoma MH-22a and histiocytic sarcoma J-774 in mice with syngenic splenocytes. Protein expression was identified by means of indirect immunofluorescence. There were two patterns of interaction of tumor cells and splenocytes: apoptosis occurred either in 80% or in an insignificant number of tumor cells. In the latter case, high Fas expression was identified before and when it dropped after the experiment. FasL expression in tumor cells
