Eukaryotic porins or VDACs (Voltage-Dependent Anion-selective Channels) are integral membrane proteins forming large hydrophilic pores. Three functioning genes for VDAC isoforms have been detected in mouse and the corresponding cDNAs are known also in humans. Tissue-specific VDAC isoform 1 (HVDAC1) deficiency in human skeletal muscle is responsible of a rare mitochondrial encephalomyopathy, fatal in childhood. Since coding sequences are not affected in the patient, we focused our interest in the gene structure. HVDAC1 and 2 have been previously mapped at chromosomes Xq13-21 and 21, respectivel

Exposure of inner mitochondrial membrane resident protein PHB2 (prohibitin 2) during autophagic removal of depolarized mitochondria (mitophagy) depends on the ubiquitin-proteasome system. This uncovering facilitates the PHB2 interaction with phagophore membrane-associated protein MAP1LC3/LC3. It is unclear whether PHB2 is exposed randomly at mitochondrial rupture sites. Prior knowledge and initial screening indicated that VDAC1 (voltage dependent anion channel 1) might play a role in this phenomenon. Through in vitro biochemical assays and imaging, we have found that VDAC1-PHB2 interaction inc

Voltage dependent anion channel 2 (VDAC2) is an outer mitochondrial membrane porin known to play a significant role in apoptosis and calcium signaling. Abnormalities in calcium homeostasis often leads to electrical and contractile dysfunction and can cause dilated cardiomyopathy and heart failure. However, the specific role of VDAC2 in intracellular calcium dynamics and cardiac function is not well understood. To elucidate the role of VDAC2 in calcium homeostasis, we generated a cardiac ventricular myocyte-specific developmental deletion of Vdac2 in mice. Our results indicate that loss of VDAC

Human mitochondrial voltage-dependent anion channel 2 (hVDAC-2), the most predominant isoform seen in brain mitochondria, is not only crucial for cell survival but is also implicated in Alzheimer disease. The abundance of cysteines in this isoform is particularly fascinating, as hVDAC-1 cysteines have no associated functional role. We report a detailed biophysical examination of a Cys-less mutant of hVDAC-2, and its behavioral comparison with the wild type protein. Our findings suggest that cysteine mutation results in the formation of a better barrel at the expense of weakened protein-lipid i

It has been shown that the voltage (V(m)) dependence of ClC Cl(-) channels is conferred by interaction of the protopore gate with H(+) ions. However, in this paper we present evidence which indicates that permeant Cl(-) ions contribute to V(m)-dependent gating of the broadly distributed ClC-2 Cl() channel. The apparent open probability (P(A)) of ClC-2 was enhanced either by changing the [Cl(-)](i) from 10 to 200 mM or by keeping the [Cl(-)](i) low (10 mM) and then raising [Cl(-)](o) from 10 to 140 mM. Additionally, these changes in [Cl(-)] slowed down channel closing at positive V(m) suggestin

Increased pulmonary artery endothelial cell (PAEC) endothelium-dependent nitric oxide synthase (eNOS) activity mediates perinatal pulmonary vasodilation. Compromised eNOS activity is central to the pathogenesis of persistent pulmonary hypertension of the newborn (PPHN). Voltage-derived anion channel (VDAC)-1 was recently demonstrated to bind eNOS in the systemic circulation. We hypothesized that VDAC isoforms modulate eNOS activity in the pulmonary circulation, and that decreased VDAC expression contributes to PPHN. In PAECs derived from an ovine model of PPHN: (1) there is eNOS activity, but

BACKGROUND: Pulmonary fibrosis (PF) is sustained by apoptosis-resistant myofibroblasts that undergo a metabolic shift toward aerobic glycolysis. Hexokinase 2 (HK2) couples these two features: it catalyzes the first committed step of glycolysis and, by occupying voltage-dependent anion channel 1 (VDAC1) on the outer mitochondrial membrane, physically excludes the pro-apoptotic effector Bcl-2 Associated X protein (BAX) from VDAC1, thereby raising the apoptotic threshold. Epigallocatechin gallate (EGCG), a principal green tea catechin, exhibits anti-fibrotic activity in preclinical models, yet it

Triple-negative breast cancer (TNBC) lacks effective molecularly targeted therapies, highlighting an urgent need to identify druggable vulnerabilities. Although the marine-derived ophiobolin MHO7 exhibits potent antitumor activity, its cellular targets remain elusive. By integrating activity-based protein profiling (ABPP) with clinical relevance analysis, we identified voltage-dependent anion channel 2 (VDAC2) as a primary MHO7-interacting protein in TNBC cells. Probe pull-down, cellular thermal shift assays, microscale thermophoresis and colocalization analysis provided convergent evidence of

BACKGROUND: Follicle development takes place under the control of hormonal and environmental stimuli. It suggested that to improve in vitro fertilisation outcomes in poor responders increasing gonadotropin doses be used. Excessive gonadotropin leads to atresia and impairs follicular development, but the molecular mechanisms of follicular atresia remain largely unknown. Recently, it was suggested that autophagy may be an alternative mechanism involved in follicle depletion.
AIMS: In this study, we aimed to clarify the role of autophagic markers such as light chain (LC) 3B and voltage dependent

Effective and reliable prediction for ecotoxicity, especially when affecting different levels of trophic chains, including humans, is increasingly gaining even more prominence as ecosystems face new threats and challenges, as that posed by the per- and poly-fluoroalkyl substances (PFAS). Toxicological prediction of PFAS in aquatic organisms, such as zebrafish, can be efficiently achieved through computational ecotoxicological approaches which are fully aligned with the state-of-the-art of new approach methodologies (NAMs) and current regulatory recommendations. Specifically in this work, the P

Spring viremia of carp virus (SVCV) represents a significant threat to cyprinids, particularly common carp (Cyprinus carpio). The disease caused by this virus is characterized by tissue necrosis and petechial hemorrhages. However, the pathogenesis of SVCV infection remains poorly understood. Pyroptosis, a recently identified form of programmed cell death, plays a crucial role in host-pathogen interactions and provides a novel approach for studying inflammation-related diseases. This study demonstrates that SVCV induces gasdermin Eb-dependent pyroptosis through activation of NLRP3 and initiatio

This study aimed to identify voltage-dependent anion channel 2 (Hdh-VDAC2) and determine its functional role in response to acute thermal stress, H2O2-induced stress, heavy metal toxicity, bacterial and viral infections, and during metamorphosis. Structural analysis confirmed that Hdh-VDAC2 is a pore-forming β-barrel protein. Molecular docking further confirmed the protein-protein interactions of Hdh-VDAC2 with Hdh-BAX, Hdh-caspase 3, and Hdh-BCL2. In the Hdh-VDAC2-inhibited hemocytes (HCY), apoptotic genes (Hdh-caspase-3 and Hdh-BAX) exhibited elevated mRNA expression, while the anti-apoptoti

The voltage-dependent anion-selective channel (VDAC) plays a crucial role in mitochondrial function, and VDAC paralogs are considered to ensure the differential integration of mitochondrial functions with cellular activities. Heterologous expression of VDAC paralogs in the yeast Saccharomyces cerevisiae por1Δ mutant cells is often employed in studies of functional differentiation of human VDAC paralogs (hVDAC1-hVDAC3) regardless of the presence of the yeast second VDAC paralog (yVDAC2) encoded by the POR2 gene. Here, we applied por1Δpor2Δ double mutants and relevant por1Δ and por2Δ single muta

The voltage-dependent anion channel (VDAC) is the most ubiquitous protein in the mitochondrial outer membrane. This channel facilitates the flux of water-soluble metabolites and ions like calcium across the mitochondrial outer membrane. Beyond this canonical role, VDAC has been implicated, through interactions with protein partners, in several cellular processes such as apoptosis, calcium signaling, and lipid metabolism. There are three VDAC isoforms in mammalian cells, VDAC1, VDAC2, and VDAC3, with varying tissue-specific expression profiles. From a biophysical standpoint, all three isoforms

Neurosteroids modulate the function of γ-aminobutyric acid type A (GABAA) receptors in the brain. We showed that GABAA receptors modulate neurosteroid interactions with VDAC. GABA enhanced [3H]6-AziP labeling of VDAC in a concentration-dependent manner. This effect of GABA was prevented by the GABAA receptor antagonists gabazine and picrotoxin. Studies with brain tissue from mice deficient for VDAC showed that GABA enhanced [3H]6-AziP labeling of VDAC2 and VDAC1. GABA enhanced photolabeling of VDAC in HEK cells expressing γ2L-subunit, suggesting that the γ2L-subunit is required for the interac

The cystic fibrosis transmembrane conductance regulator (CFTR) Cl- channel pore is blocked in a voltage-dependent manner by a broad range of anionic substances added to the cytoplasmic side of the membrane. Here we investigate the origin of the voltage dependence of block by intracellular Au(CN)2-, a highly permeant lyotropic anion which also acts as a high-affinity blocker of Cl- permeation. Not only the affinity, but also the voltage dependence of block by intracellular Au(CN)2- ions is strongly dependent on extracellular Cl- concentration; following replacement of most extracellular Cl- by

The transmembrane protein 43 (TMEM43) S358L mutation is the most common mutation found in patients with ARVC5, a severe subtype of arrhythmogenic right ventricular cardiomyopathy (ARVC). Characterizing the interacting proteins of TMEM43 mutants is important for unraveling the underlying mechanisms of the relative pathogenesis. Here, we applied quantitative immunoprecipitation (IP)-mass spectrometry (MS) to screen the differential binding partners between TMEM43 and TMEM43 p.S358L and identified 166 interacting protein candidates. Functional enrichment of these proteins highlighted their involv

There are a number of mammalian anion channel types associated with cell volume changes. These channel types are classified into two groups: volume-activated anion channels (VAACs) and volume-correlated anion channels (VCACs). VAACs can be directly activated by cell swelling and include the volume-sensitive outwardly rectifying anion channel (VSOR), which is also called the volume-regulated anion channel; the maxi-anion channel (MAC or Maxi-Cl); and the voltage-gated anion channel, chloride channel (ClC)-2. VCACs can be facultatively implicated in, although not directly activated by, cell volu

Organelle communication through endoplasmic reticulum-mitochondrial crosstalk is essential for maintaining cellular homeostasis and is tightly regulated by tethering proteins within mitochondria-associated endoplasmic reticulum membranes. In a recent study published in Kidney International, He et al. identified sarcoplasmic/endoplasmic reticulum Ca2+-ATPase 2 as a key regulator of endoplasmic reticulum-mitochondrial calcium homeostasis in proximal tubules and demonstrated that loss of sarcoplasmic/endoplasmic reticulum Ca2+-ATPase 2 drives voltage-dependent anion channel 1 oligomerization, mit

Mitochondrial dysfunction contributes to numerous human diseases. Voltage‑dependent anion channel 2 (VDAC2) is an essential outer mitochondrial membrane porin with distinct structural and functional properties that are non-redundant with those of VDAC1 and VDAC3. VDAC2 precisely controls ATP, ADP, NADPH and Ca2+ transport, thereby acting as a key hub for energy metabolism, calcium homeostasis, redox balance and cell fate. It directly binds BAX/BAK to differentially regulate apoptosis and is also involved in ferroptosis, necroptosis, Parkin‑dependent mitophagy, and lipid transport through prote
