Quercetin attenuates chondrocyte anoikis via modulation of mesenchymal stromal cell-derived small extracellular vesicles to promote cartilage repair in osteoarthritis.
Аннотация
BACKGROUND
Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degeneration and impaired repair. Current therapies are unable to effectively slow its progression, and there is an urgent need to develop new therapeutic strategies.
METHODS
This study investigated the effects of quercetin-treated mesenchymal stromal cell-derived small extracellular vesicles (QT-MSC-sEV) on chondrocytes. The effects of QT-MSC-sEV on chondrocyte proliferation, migration, and anoikis were evaluated using CCK-8, scratch assays, flow cytometry, and Western blotting. Additionally, the role of QT-MSC-sEV in cartilage repair in vivo was assessed using an OA rat model induced by anterior cruciate ligament transection (ACLT).
RESULTS
QT-MSC-sEV significantly promoted chondrocyte proliferation and migration, inhibited anoikis, and regulated related signaling pathways. In vivo, QT-MSC-sEV reduced osteophyte formation and improved cartilage integrity. Mechanistically, QT-MSC-sEV enhanced cartilage repair by activating the TGF-β signaling pathway through miR-503-5p-mediated downregulation of Smurf1.
CONCLUSION
This study provides the first mechanistic elucidation of OA cartilage degeneration from an anoikis perspective, demonstrating that QT-MSC-sEV inhibit chondrocyte anoikis and promote cartilage repair through the HIF-1α/miR-503-5p/Smurf1/Smad1 axis, thereby establishing a novel therapeutic strategy for OA.
