Аннотация

OBJECTIVE: Wound healing is a topic of significant interest in current times due to the rising prevalence of chronic illnesses linked to poor healing. The purpose of this work was to develop a guar gum (GG) & chitosan (CS) composite metoprolol succinate-loaded novel scaffold by the lyophilization technique and subsequently evaluate for several characteristics necessary for surgical wound healing.

METHODS: The interaction of GG and chitosan was evaluated by Fourier transform infrared spectroscopy (FTIR) and thermal analysis (DSC, TGA).

RESULTS: The results showed that drug content in four different formulations (F1, F2, F3, F4) was 71.37 %, 83.5 %, 91.37 %, 96.83 %, respectively. The antimicrobial properties of the optimized scaffold (F3) against Staphylococcus aureus MTCC 2940 and Escherichia coli MTCC 1195 were determined by the agar well diffusion method. Drug concentration and drug residence time in the skin remained constant for up to 24 h after topical administration of the F3. In vivo study showed that metoprolol succinate has a potent wound-healing ability, as evidenced by enhanced wound closure and tissue regeneration supported by histopathological parameters.

CONCLUSION: This developed novel scaffold based on the CS-GG conjugate loaded with metoprolol in advancing wound care treatments by use of its several biological applications.
