INTRODUCTION: Sex differences in cardiovascular disease (CVD) risk are examined through biological and clinical factors, with less attention to early-life social exposures. This study examined associations of childhood parental son preference with CVD risk, sex differences, and mediation by modifiable risk factors.
METHODS: This cohort analysis included China Health and Retirement Longitudinal Study participants aged ≥45 years without baseline CVD. Parental son preference was assessed retrospectively in 2014; incident CVD was self-reported physician-diagnosed heart disease or stroke through 2

Restriction fragment length polymorphism (RFLP) was the first reliable genetic marker used in plant molecular studies. It is a DNA-based marker that detects variation in DNA sequence at restriction enzyme sites and produces codominant, locus-specific markers that differentiate homologous and heterozygous genotypes. It provides greater accuracy than trait-based methods. The application of RFLP markers includes developing genetic linkage maps, conducting QTL analysis, characterizing germplasm, and detecting somaclonal variations. Despite being the availability of new molecular markers, RFLP stil

The dynamic flow of heavy metals in ecosystems is disturbing the sustainability of environmental quality and the flow of resources in ecosystems. Detection of genotypes with metal-tolerance ability is a prerequisite for mitigating this burning problem, through restoration of degraded ecosystems, progressive crop productivity, and improvement in efficiency of phytoremediation programs. The application of simple sequence repeat (SSR) markers for tracing metal tolerance in plants is gaining importance among researchers. SSRs, as molecular markers, are highly informative and preferable compared to

De novo peptide sequencing enables the identification of a broad repertoire of major histocompatibility complex class I-associated peptides, providing a valuable source of potential immunotherapeutic targets in cancer. Nevertheless, its practical utility remains limited by the absence of robust methods for confidently determining peptide-spectrum matches and interpreting their proteogenomic information. This chapter presents the pXg workflow, designed to comprehensively and reliably identify peptides derived from de novo sequencing in mass spectrometry-based immunopeptidomics, integrated with

Chromatin immunoprecipitation followed by sequencing (ChIP-seq) is a widely used technique to study the distribution of transcription factors, histones, posttranslational modifications, or other DNA-associated proteins across the genome. This methodology is based on the isolation of DNA fragments bound by the protein of interest using specific antibodies. Computational analysis of sequencing libraries derived from the isolated DNA fragments will delineate genome-wide maps of protein-DNA interactions. ChIP-seq profiling of EWSR1::FLI1 oncogene has enormously contributed to the understanding of

Allergic reactions associated with arthropod bites are a growing concern worldwide. In particular, Alpha-Gal syndrome (AGS) is a red meat allergy associated with IgE antibodies against galactose-alpha-1,3-galactose (alpha-Gal) that is typically linked to tick bites, but other arthropods with alpha-Gal content may also induce allergic reactions. The AGS is underdiagnosed and thus important to evaluate the risks that may be associated with exposure to bites of arthropods with alpha-Gal. To approach this objective, herein are described the materials and methods for the detection of alpha-Gal cont

Alpha-GAL syndrome (AGS) is a type of allergy to mammalian meat and other foods or pharmaceuticals made from mammalian substrates. It is an IgE-mediated reaction caused by sensitization to the polysaccharide galactose-α-1,3-galactose (alpha-GAL) found in tick saliva after a previous tick bite. Alpha-GAL is present on proteins of mammals except primates. Sensitization to alpha-GAL in tick saliva predisposes to cross-reactivity following exposure (oral or non-oral) to glycated galactose-α-1,3-galactose proteins, regardless of their origin. Symptoms of varying severity (from dyspepsia to anaphyla

Allergic diseases have increased significantly in recent decades, with a higher incidence of severe manifestations such as anaphylaxis. This increase is attributed mainly to environmental factors associated with the Western lifestyle that alter the epithelial barrier, promoting sensitization to allergens, such as those from arthropods. Hymenoptera (bees and wasps) are the most implicated arthropods. The sting introduces a venom composed of highly sensitizing proteins that are capable of triggering reactions ranging from local and mild to potentially fatal systemic reactions. The incidence is h

The differential diagnosis of small round cell tumors of bone and soft tissue (SRCT) crucially relies on the synoptic evaluation of morphology, immunohistochemical patterns, and molecular features. Although the implementation of broad RNA sequencing in diagnostic molecular pathology routines has substantially changed the standards for molecular confirmation of diagnoses, fluorescence in situ hybridization (FISH) on formalin-fixed, paraffin-embedded (FFPE) tissue sections remains an essential tool for rapid molecular corroboration of diagnoses, which is essentially required for therapeutic deci

Restriction fragment length polymorphism (RFLP) has been an important molecular tool in advancing cotton (Gossypium spp.) improvement by enabling researchers to detect stable genetic variations across the genome. Using restriction enzymes and labeled probes, RFLP generates highly reproducible, co-dominant markers that help identify differences among cultivars, wild species, and breeding lines. This technique has been widely used to construct early genetic linkage maps, locate quantitative trait loci (QTLS) associated with fiber quality, yield, and disease resistance, and study genome structure

Sesame (Sesamum indicum L.) is an important oilseed crop with considerable agronomic potential, widely cultivated across tropical and subtropical regions. Despite its substantial economic value, genomic research in sesame has progressed more slowly than in major staple crops. The advent of high-throughput single-nucleotide polymorphism (SNP) markers has markedly advanced sesame breeding by enabling the development of dense genetic linkage maps, genome-wide association studies (GWAS), and marker-assisted selection (MAS). This chapter presents detailed protocols for SNP discovery and validation

BACKGROUND: There have not been any studies to date on trends in digital health literacy (DHL) in Germany. We studied trends in DHL and relevant factors that affect it on the basis of two population-based surveys conducted in 2019/2020 and 2024/2025.
METHODS: Data from the second and third Health Literacy Surveys in Germany (HLS-GER 2, 2019/2020; HLS-GER 3, 2024/2025) were evaluated. Subjective difficulty with digital health information was measured with the HLS19-DIGI survey instrument in two personal cross-sectional surveys of the adult, German-speaking population of Germany.
RESULTS: In H

The genus Rhodotorula comprises oleaginous yeasts widely distributed across diverse environments and recognized for their ability to synthesize carotenoids, lipids, and other high-value metabolites with biotechnological relevance. The preservation of these microorganisms is critical to maintain their genetic stability, metabolic potential, and phenotypic traits over time. Among available methods, cryopreservation using glycerol is one of the most effective strategies for long-term storage, as it protects cellular structures during freezing and minimizes metabolic activity at ultra-low temperat

Preserving fungi in liquid media is an essential strategy for maintaining long-term viability, genetic stability, and phenotypic characteristics across diverse fungal species. Among the most widely used methods are storage in sterile distilled water, storage in mineral oil, and cryopreservation. Each technique is based on biological and biochemical principles, such as metabolic suppression, osmotic balance, protection against oxidative stress, and induction of quiescence. These principles minimize cell damage, maintain structural integrity, and ensure the recovery of highly viable cultures. Th

The identification of biomarkers for diseases such as allergies requires analytical strategies capable of handling high-dimensional and heterogeneous biological data. Advances in omics technologies have enabled the systematic exploration of molecular features across multiple biological levels. However, their effective integration and interpretation remain challenging. In this chapter, we present a data-driven bioinformatic framework for biomarker discovery based on single-omics and multi-omics datasets, with a particular focus on allergy-related applications. We describe complementary strategi

The genus Trichoderma comprises potent biocontrol fungi known for their antagonistic activity against diverse phytopathogens. Understanding the genetic diversity among Trichoderma isolates is essential for identifying strains with enhanced biocontrol potential. Sequence-related amplified polymorphism (SRAP) markers, which target open reading frames (ORFs) and associated regulatory regions, offer a robust and efficient molecular tool for diversity analysis. This chapter describes a detailed protocol for genomic DNA extraction, SRAP-PCR amplification, and data analysis for Trichoderma isolates.

Maize is the third most significant cereal crop, and most of its agronomic traits are quantitative in nature. The fine mapping of genes or quantitative trait loci (QTLs) associated with these traits is essential for marker-assisted selection (MAS) in maize breeding. Although significant progress has been made, there remains a lack of high-density, highly polymorphic single-nucleotide polymorphism (SNP) markers, especially competitive allele-specific PCR (KASP) SNP markers suitable for automated genotyping. The development of next-generation sequencing (NGS) platforms has generated vast amounts

This chapter details a comprehensive, step-by-step RAPD protocol for assessing genetic diversity in 16 Iranian Echium amoenum ecotypes, a medicinal plant. The method uses STE (Sucrose-Tris-EDTA) buffer to extract DNA from leaf tissue, followed by spectrophotometric and electrophoretic quality control. PCR amplification with 20 decamer primers yields 248 polymorphic bands (80% polymorphism), separated by agarose gel electrophoresis. NTSYS-pc analysis, using the Dice coefficient, UPGMA clustering, and principal component analysis, reveals geographic-climatic groupings. This approach is applicabl

The molecular characterization of genetically modified plants is commonly based on the detection of foreign DNA sequences introduced during transformation. While this strategy is effective for conventional transgenic plants, it becomes inadequate for cisgenic and intragenic plants in which the introduced gene originates from the host genome and is regulated by native promoter and terminator elements. Following segregation-based elimination of selectable marker cassettes, such cisgenic plants may become indistinguishable from wild-type plants using standard sequence-dependent PCR assays. This c
