Antimicrobial resistance of Group A Streptococcus isolates from patients in Shenzhen, China during COVID-19 pandemic.

BACKGROUND: Group A Streptococcus (GAS) is a bacterial pathogen causing various infections and autoimmune diseases, with humans as its only host. Transmission occurs mainly via airborne droplets and skin mucous membrane contact. While previous surveillance studies from our group (2016–2018) established baseline resistance profiles and genotype distributions of GAS in Shenzhen, the profound societal and healthcare disruptions caused by the COVID-19 pandemic presented a unique natural experiment to investigate potential shifts in bacterial epidemiology. This study aimed to conduct a comparative

Proteasome inhibition by VR23 enhances autophagic clearance of FUSP525L-mediated persistent stress granule in SH-SY5Y cells.

Autophagy is a conserved catabolic pathway that preserves cellular homeostasis through lysosomal degradation. Beyond its general role in proteostasis, selective autophagy mediates the clearance of selective cellular targets such as persistent stress granules (SGs), in a process termed granulophagy. SGs are dynamic cytoplasmic assemblies that normally disassemble after stress relief; however, their aberrant persistence has arisen as a pathological feature of neurodegenerative disorders, including amyotrophic lateral sclerosis (ALS). However, the molecular regulation of granulophagy remains inco

Screening and engineering of broad-spectrum aptamers for construction of a dual-mode aptasensor for highly sensitive detection of macrolide antibiotics.

Macrolide antibiotics (MAs) are widely used in the fields of medicine, animal husbandry and aquaculture. However, residues due to the overuse of MAs threaten human health and cause ecological and environmental pollution. Aptasensors offer a promising solution for the simultaneous detection of these residues. In this study, a broad-spectrum aptamer (F1-Apt1) with high affinity for MAs was successfully screened using Capture-SELEX technology. Through secondary structure analysis and molecular docking, a truncated 40-nt G-quadruplex-containing aptamer (F1-apt1-40) was obtained, with the dissociat

Spinosyn A derivatives as ASS1 activators and tumor inhibitors.

Spinosad, a natural-origin pesticide, is extensively employed in agriculture and public health. Its primary component, Spinosyn A, has also attracted considerable interest due to its antiproliferative activity against various cancer cell lines. While ASS1 has emerged as a promising target for anticancer therapy, the development of ASS1 activators remains a largely unexplored research area. Previous work by our group identified Spinosyn A as the first-in-class ASS1 activator. In this study, to further elucidate the structure-activity relationship (SAR) of Spinosyn A as both an ASS1 activator an

Nutrient and spinosad concentrations influence liquid bait consumption and mortality in spinosad-resistant melon flies (Diptera: Tephritidae).

BACKGROUND: The success of insecticide-laced liquid protein bait spray applications relies on attraction and consumption by the target pest. Commercial liquid baits often recommend a broad range of dilution rates, even though many insects adjust their intake of foods based on nutrient concentrations. This compensatory feeding response could influence the amount of toxin ingested, but this impact has not been thoroughly elucidated. We manipulated liquid diet and spinosad concentrations to reveal their effects on consumption and mortality in three lines of melon flies, Zeugodacus cucurbitae, tha

Severe community-acquired pneumonia: current concepts and controversies.

Community-acquired pneumonia, particularly in its severe forms (sCAP), remains a major public health problem due to its frequency, immediate and delayed complications, and the cost of treatment. Although rare, resistant pathogens could make it increasingly difficult to choose an empirical antibiotic treatment. Rapid molecular microbiological diagnostic techniques could help guide this choice, but their role needs to be better evaluated and their cost may be an obstacle to their widespread use. The duration of treatment tends to be decreasing, but could be guided by clinical progression and pos

Discovery of a Fungal HR-PKS Cluster Encoding Biosynthetic Pathways for Macrolides with Two Distinct Ring Sizes.

Through the heterologous expression of a highly reducing polyketide synthase and a thioesterase from the apeml cluster, a putative macrolide biosynthetic gene cluster on the genome of Aspergillus petrakii, we obtained a naturally new 10-membered macrolide (1) and recifeiolide (2), a known 12-membered macrolide. To obtain modified macrolides, feeding experiments using Aspergillus oryzae transformants expressing individual modification enzymes were employed, resulting in the isolation of aspinolide A (3) and 2 new macrolides, petrakilides A (4) and B (5). These findings highlight a promiscuous e

Deciphering the O-Carbamoylation Modification Mechanism in Venturicidin A Biosynthesis.

Herein, VtdB was identified as an essential O-carbamoyltransferase that catalyzes the conversion of venturicidin B (3) to venturicidin A (1). Notably, a 2.8 Å resolution crystal structure of the VtdB-substrate complex (VtdB/VTD-B) revealed that 3 occupies a specific hydrophobic pocket within the Kael-like domain, where His35 plays a critical deprotonation role during enzymatic catalysis. These biochemical and structural research provides the first mechanistic insight into d-olivose-glycosylated macrolide recognition by the carbamoyltransferase, enabling its future engineering.

Plasmids of the multidrug-resistant Citrobacter portucalensis KOS1-1 strain isolated from a wastewater treatment plant harbor antibiotic resistance genes and gene clusters involved in carbon metabolism.

The growing interest in multidrug-resistant (MDR) Citrobacter species stems from their epidemiological significance and their potential to harbor antibiotic resistance genes (ARGs), with mobile genetic elements playing a central role in their dissemination. Wastewater treatment plants play an important role in the formation of such MDR strains due to the high rate of horizontal gene transfer in these environments. In this study, we isolated the MDR strain KOS1-1 of Citrobacter portucalensis from wastewater, sequenced its genome, and characterized its ARG content and plasmid profile. This strai
